An efficient and facile one-pot synthesis is described for the preparation of novel 2-amino-5-hydroxy-4H-chromene derivatives with a spirooxindole nucleus using Ca(OH)2-mediated three-component reactions of substituted resorcinols with isatins and malononitrile. This simple method provided a variety of biologically interesting diverse 2-amino-5-hydroxy-4H-chromene derivatives in moderate yields under mild reaction conditions.  相似文献

As agonists of TLR7/8, single‐stranded RNAs (ssRNAs) are safe and promising adjuvants that do not cause off‐target effects or innate immune overactivation. However, low stability prevents them from mounting sufficient immune responses. This study evaluates the adjuvant effects of ssRNA derived from the cricket paralysis virus intergenic region internal ribosome entry site, formulated as nanoparticles with a coordinative amphiphile, containing a zinc/dipicolylamine complex moiety as a coordinative phosphate binder, as a stabilizer for RNA‐based adjuvants. The nanoformulated ssRNA adjuvant was resistant to enzymatic degradation in vitro and in vivo, and that with a coordinative amphiphile bearing an oleyl group ( CA‐O ) was approximately 100 nm, promoted effective recognition, and improved activation of antigen‐presenting cells, leading to better induction of neutralizing antibodies following single immunization. Hence, CA‐O may increase the efficacy of ssRNA‐based adjuvants, proving useful to meet the urgent need for vaccines during pathogen outbreaks.  相似文献

Nanocapsules containing hinokitiol (HKL) were prepared by an emulsion‐diffusion method. Poly(?‐caprolactone) (PCL), synthesized according to a ring‐ opening method, was used as a wall material, and HKL dissolved in octylsalicylate (OS), was used as a core material. Under a microscope, submicron‐sized microcapsules were observed. The apparent shape was either spherical or amorphous. According to the image analyzer analysis, the size ranges from tenths of a nanometer to one micron, and more than 50% in number were less than 300 nm. In a calorimetric study, HKL exhibited an endothermic peak around 52°C, but no peak for the melting of HKL was observed when it was encapsulated in microcapsules. This suggests HKL remained dissolved in the nanocapsule cores but did not exist as a solid crystalline. In an in vitro permeation study, the flux of HKL encapsulated in the nanocapsules was somewhat higher than that of HKL dissolved in either propylene glycol or ethanol. This is probably because that PCL is hydrophobic, so the nanocapsules would readily interact with skin lipid.  相似文献

In a number of previous communications, we reported on the utility of the solvatochromic linear solvation energy relationship (LSER) method for the correlation of a number of solute and solvent-dependent properties. In those studies, it was our practice to examine the effect of a variety of solvents on a given solute or a number of solutes in a given solvent. Here we report on a novel generalized LSER in which the solute and solvent were both simultaneously varied so as to assess the validity of the entire LSER concept and define its limits. The Hildebrand solubility parameter, δH, the Kamlet–Taft solvatochromic parameters, π*, α, β and the solute molar volume, V2, were used as the explanatory variables. The gas–liquid partition coefficient (K) was the property of interest. We have found that the correlation using the generalized linear solvation energy relationship is statistically as good as the previous LSER correlations despite the use of a far smaller number of freely adjustable parameters. Furthermore, the new approach is able to give reasonable predictions of K values of systems not included in the data set upon which the regression is based.  相似文献

This paper describes new and efficient synthetic approaches for biologically interesting tetrahydroquinoline analogues. The key strategies involve the ethylenediamine diacetate-catalyzed cyclization of 1,3-dicarbonyls to aminobenzaldehydes through domino Knoevenagel/hetero Diels–Alder reactions.
