Light-harvesting complex I (LHCI), which serves as a peripheral antenna for photosystem I (PSI) in green plants, consists mainly of four polypeptides, Lhca1-4. We report room temperature emission properties of individual reconstituted monomeric Lhca proteins (Lhca1, -2, -3, and -4) and dimeric Lhca1/4, performed by steady-state and time-resolved fluorescence techniques. The emission quantum yields of the samples are approximately 0.12, 0.085, 0.081, 0.041, and 0.063 for Lhca1, -2, -3, -4, and the -1/4 dimer, respectively, which is considerably lower than the value of 0.22 found for light-harvesting complex II (LHCII), the main peripheral antenna complex of photosystem II in green plants. The decay components of LHCI proteins can be divided in two categories: Lhca1 and Lhca3 have decay times of 1.1-1.6 ns and 3.3-3.6 ns, and Lhca2 and Lhca4 have decay times of 0.7-0.9 ns and 3.1-3.2 ns. These categories seem to correlate with the pigment composition of the samples. All decay times are faster than that observed previously for LHCII. When the absolute emission yields and the lifetimes of the Lhca samples are combined, the overall emission properties of the individual Lhca proteins are expressed in terms of their emitting dipole moment strength. In the samples without extreme red states, that is, Lhca1 and Lhca2, the emitting dipole moment has a value close to unity (relative to monomeric chlorophyll in acetone), which is similar to that for LHCII, whereas, in the samples with the red-most state (F-730), that is, Lhca3, -4, and the -1/4 dimer, the emitting dipole moment has a value less than unity (0.6-0.8), which can be explained by mixing the red-most (exciton) state with a dark charge-transfer state, as suggested in previous PSI red pigment studies. In addition, we find a lifetime component of approximately 50-150 ps in all red-pigment-containing samples, which cannot be due to "slow" energy transfer, but is instead assigned to an unrelaxed state of the pigment-protein, which, on this time-scale, is converted into the final emitting state.  相似文献

Extended Hückel theory calculations have been carried out to investigate the possible formation of stable π and σ complexes between olefins and some hydrocarbonylirons postulated as active catalysts in Reppe synthesis. It was found that H2Fe(CO)4 and HFe(CO)4- do not coordinate ethylene, unlike the corresponding CO deficient species H2Fe(CO)3 and HFe(CO)3-, which interact with the olefin to give stable π complexes. Moreover ethylene—H2Fe(CO)3 is more stable than ethylene—HFe(CO)3-, in line with the conclusions based on the experimental results. Stable alkyl intermediates are predicted starting from the coordinatively saturated hydrocarbonyls.  相似文献

The Maillard reaction occurring between sugars and amino groups is important in living systems. When amino groups belonging to protein chains are involved, the Maillard reaction has been invoked as responsible for protein cross-linking and the production of 'toxic' compounds. The reaction leads to the production of a heterogeneous group of substances, usually called advanced glycation end products (AGEs). Classical analytical approaches, such as spectroscopic (ultraviolet, fluorescence) and mass spectrometric (matrix-assisted laser desorption/ionization, liquid chromatography/electrospray ionization mass spectrometry) methods, have shown that the digestion mixture is highly complex. However, there are clear differences between the digestion mixtures of glycated and unglycated human serum albumin (HSA). In the former case, possible glycated peptides belonging to the AGE peptide class may be identified. Tandem mass spectrometric experiments on selected species seemed to be promising as regards structural information, but it was thought of interest to undertake the present investigation, based on liquid chromatography/electrospray ionization Fourier transform mass spectrometry, in order to obtain definitive results on their elemental composition. Using this approach, about 20 glycated peptides were detected and their possible structures were postulated by examining the known sequence of HSA.  相似文献

Structural studies of a synthetic melanin, obtained by means of performic acid oxidation of tryptophan, were carried out by pyrolysis gas chromatography/mass Spectrometry (Py-GC/MS). To identify the best pyrolysis conditions, both Curie-point pyrolysis and filament pyrolysis were employed and the effects of pyrolysis temperatures and times were studied. Using the first approach, various compounds were identified: toluene, ethylbenzene, styrene, indole, methylindole, ethylindole, phenol, cresol and ethylphenol. Using filament pyrolysis some interesting differences could be observed. Whereas toluene, ethylbenzene, phenol, cresol and methylindole were found, neither indole nor ethylindole was detected. Instead, new pyrolysis products were evident, such as methylpyrrole and indolin-2-one. Hence filament pyrolysis seems to activate different thermal decomposition pathways of the melanin under study. It is suggested that tryptophan melanin is a polymer containing indole and hydroxyindole derivatives as subunits.  相似文献

The 4-halo-3,5-dimethyl pyrazoles have been synthetisized in good yields in short reaction times in the absence of a catalyst by reaction of 3,5-dimethyl pyrazoles with N-halosuccinimides (NBS, NCS and NIS) under ultrasound irradiation. The synthesis of derivatives of 2,3‐dihydroimidazo[1,5,4‐ef][1,2,5]benzothiadiazepin‐6(4H,7H)‐thione 1,1‐dioxide is reported starting from N‐substituted ethyl 2‐(5‐chloro‐2‐nitrobenzenesulfonamido)‐2‐alkyl‐acetates. Fundamental steps of the synthetic pathway were: i) intramolecular cyclization of N‐substituted 2‐(2‐amino‐5‐chlorobenzenesulfonamido)‐2‐alkylacetic acids in the presence of N‐(3‐dimethyl‐aminopropyl)‐N′‐ethyl carbodiimide hydrochloride‐N,N‐dimethylaminopyridine complex; ii) building of imidazole ring from 2‐alkyl‐8‐chloro‐2,3‐dihydro‐3‐methyl‐1,2,5‐benzothiadiazepin‐4(5H)‐one 1,1‐dioxide to achieve 2‐alkyl‐9‐chloro‐2,3‐dihydro‐3‐methylimidazo[1,5,4‐ef][1,2,5]benzothiadiazepin‐6(4H,7H)‐one 1,1‐dioxide; iii) preparation of thiocarbonyl derivative by treatment with Lawesson's reagent. Introduction of a 3‐methyl‐2‐butenyl chain at position 2 of above imidazobenzothiadiazepinone required protection at the 7 position with thermally removable tert‐butoxycarbonyl moiety, due to the fact that alkylation of unprotected structure proved to be regioselective for the 7 position.  相似文献

The treatment of plasma with a sodium dodecy sulphate solution before deproteinization with sulphosalicylic acid is described. With this treatment, the liberation of tryptophan from plasma albumin is complete and its determination can be made directly, together with that of other amino acids and related compounds, by automatic ion-exchange chromatography, eliminating the need for an extra analysis for the accurate determination of this amino acid. Results of tryptophan recovery tests, carried out both on plasma samples simply deproteinized with sulphosalicylic acid and on other samples pre-treated with sodium dodecylsulphate solution are reported. These results are also compared with those obtained when trichloroacetic acid was used as deproteinizing agent. A recovery study on a human albumin (fraction V) solution containing a known amount of tryptophan is also described.  相似文献

By measuring the relative CO quantum yields from ketene photolysis as a function of photolysis wavelength we have determined the threshold energy at 25° for CH2CO(1A1) → CH2(3B1) + CO(1Σ+) to be 75.7 ± 1.0 kcal/mole. This corresponds to a value of 90.7 ± 1.0 kcal/mole for ΔHf2980[CH2(3B1)]. By measuring the relative ratio of CH2(1A1)/CH2(3B1) from ketene photolysis as a function of photolysis wavelength we have determined the threshold energy at 25°C for CH2CO(1A1) → CH2(1A1) + CO(1Σ+) to be 84.0 ± 0.6 kcal/mole. This corresponds to a value of 99.0 ± 0.6 kcal/mole for ΔHf2980[CH2(1A1)]. Thus a value for the CH2(3B1) ? CH2(1A1) energy splitting of 8.3 ± 1 kcal/mole is determined, which agrees with three other recent independent experimental estimates and the most recent quantum theoretical calculations.  相似文献

Analogous to the formation of CH(2)[(t-Bu)(2)C(6)H(2)O](2)P(Ph)(O(2)C(6)Cl(4)) (1), the new bicyclic tetraoxyphosphoranes CH(2)[(t-Bu)(2)C(6)H(2)O](2)P(Et)(O(2)C(6)Cl(4)) (3) and CH(2)[ClC(6)H(3)O](2)P(Ph)(O(2)C(6)Cl(4)) (4) were synthesized by the oxidative addition of the appropriate cyclic phosphines with o-tetrachlorobenzoquinone. For the formation of CH(2)[(t-Bu)(2)C(6)H(2)O](2)P(Ph)(O(2)C(2)Ph(2)) (2), a similar reaction was followed with the use of benzil (PhCOCOPh) in place of o-tetrachlorobenzoquinone. X-ray analysis of 1-3 revealed trigonal bipyramidal geometries and provided evidence for the first series of complexes in the absence of ring strain in which the least electronegative group, ethyl or phenyl, is located in an axial position, in violation of the electronegativity rule. Thus, the two oxygen-containing ring systems occupied two different sets of positions in the trigonal bipyramid (TBP) with the eight-membered rings at diequatorial sites. X-ray analysis of 4 revealed a trigonal bipyramidal geometry with electron-withdrawing chlorine substituents on each ring assumed the more conventional geometry with the rings occupying axial-equatorial positions and the phenyl group located in the remaining equatorial site. The fact that molecular mechanics calculations favorably reproduced the observed geometries suggests that a steric contribution associated with the ring tert-butyl groups for 1-3 is partly responsible in favoring diequatorial ring occupancy for the eight-membered ring. NMR data supported rigid pentacoordinated structures in solution at 23 degrees C. Phosphorane 1 crystallizes in the orthorhombic space group Fdd2 with a = 44.787(5) ?, b = 34.648(8) ?, c = 10.3709(9) ?, and Z = 16. Phosphorane 2 crystallizes in the orthorhombic space group Pna2(1) with a = 20.658(8) ?, b = 10.342(2) ?, c = 19.879(6) ?, and Z = 4. Phosphorane 3 crystallizes in the orthorhombic space group Pcmn with a = 9.807(2) ?, b = 16.632(4) ?, c = 23.355(3) ?, and Z = 4. Phosphorane 4 crystallizes in the monoclinic space group C2/c with a = 35.699(5) ?, b = 12.187(2) ?, c = 14.284(3) ?, beta = 107.08(1) degrees, and Z = 8. The final conventional unweighted residuals are 0.0395 (1), 0.0518 (2), 0.0540 (3), and 0.0868 (4).  相似文献
