Long-chain polyunsaturated fatty acids n-3 series (n-3 LC-PUFAs), especially eicosapentaenoic and docosahexaenoic acids, are known to exert preventive effects on obesity and metabolic syndrome. Mainly consumed in the form of fish oil, LC-PUFAs n-3 are also found in significant quantities in other sources such as certain microalgae. The aim of this study was to evaluate the effects of Diacronema lutheri (Dia), a microalga rich in n-3 LC-PUFAs, on metabolic disorders associated with obesity. Three groups of male Wistar rats (n = 6 per group) were submitted for eight weeks to a standard diet or high-fat and high-fructose diet (HF), supplemented or not with 12% of Dia (HF-Dia). Compared to HF rats, HF-Dia rats showed a 41% decrease in plasma triacylglycerol (TAG) and an increase in plasma cholesterol (+35%) as well as in high-density lipoprotein cholesterol (+51%) without change to low-density lipoprotein cholesterol levels. Although fasting glycemia did not change, glucose and insulin tolerance tests highlighted an improvement in glucose and insulin homeostasis. Dia supplementation restored body weight and fat mass, and decreased levels of liver TAG (−75%) and cholesterol (−84%). In HF-Dia rats, leptin was decreased (−30%) below the control level corresponding to a reduction of 68% compared to HF rats. Similarly, the anti-inflammatory cytokines interleukin-4 (IL-4) and IL-10 were restored up to control levels, corresponding to a 74% and 58% increase in HF rats, respectively. In contrast, the level of IL-6 remained similar in the HF and HF-Dia groups and about twice that of the control. In conclusion, these results indicated that the D. lutheri microalga may be beneficial for the prevention of weight gain and improvement in lipid and glucose homeostasis.  相似文献

Polymer-bound sulfonylhydrazide was prepared from commercial sulfonic acid resin through a straightforward, one-step synthetic route avoiding chlorosulfonic intermediates. The simplex self-directing optimization method was used to determine rapidly the ideal operating conditions under microwave irradiation. Polymer-bound sulfonylhydrazide has found an application as scavenging agent in winemaking therefore, this clean preparative approach is expected to be more acceptable by winemakers. An efficient recycling protocol of the used supports has also been established.  相似文献

Aqueous solution properties of amphiphilic P(AA‐cotBA)‐b‐PPO‐b‐ P(AA‐cotBA) copolymers having various tBA contents are presented in this article. These copolymers show pH‐sensitive behavior depending on tBA/AA ratio. Hydrophobic interactions between tBA units leading to pH‐dependent macroscopic aggregates were evidenced by turbidimetry. The aggregation behavior of the PPO middle block was concealed in presence of tBA units. The formation of water‐soluble aggregated objects was characterized by Asymmetrical Flow Field Flow Fractionation (AsF4). Total synthesis of enantiopure (R)-(−)-massoialactone was achieved. The key step includes the asymmetric hydrogenation of an achiral β-keto ester using a ruthenium-SYNPHOS® catalyst to set the hydroxyl function in a stereocontrolled manner with excellent enantioselectivity (>99% ee). A remarkable feature of the spin-crossover process in several dinuclear iron(II) compounds is a plateau in the two-step transition curve. Up to now, it has not been possible to analyse the spin state of dinuclear pairs that constitute such a plateau, due to the relative high temperatures at which the transition takes place in complexes investigated so far. We solved this problem by experimentally studying a novel dinuclear spin-crossover compound [[Fe(phdia)(NCS)(2)](2)(phdia)] (phdia: 4,7-phenanthroline-5,6-diamine). We report here on the synthesis and characterisation of this system, which exhibits a two-step spin transition at T(c1)=108 K and T(c2)=80 K, displaying 2 K and 7 K wide thermal hysteresis loops in the upper and the lower steps, respectively. A plateau of approximately 20 K width centred at about 90 K, which corresponds to the 50 % of the spin conversion, separates the two transitions. The composition of the plateau was identified in metastable state after quenching to 4.2 K by means of M?ssbauer spectroscopy in an external magnetic field. Such experiments revealed that the plateau consists mainly of [HS-LS] pairs (HS=high spin, LS=low spin) and confirmed the hypothesis that the spin conversion in dinuclear entities proceeds through [LS-LS]<-->[HS-LS]<-->[HS-HS] pairs. The results are discussed in terms of a thermodynamic model based on the regular solution theory adapted for dinuclear spin-crossover compounds.  相似文献

Dipalmitoylphosphatidylcholine (DPPC) monolayers were deposited onto a germanium attenuated total reflectance (ATR) crystal using the Langmuir–Blodgett technique. The DPPC-coated crystal was then exposed to human serum albumin or human fibrinogen solutions while measuring the protein adsorption by recording FTIR spectra. The effect of the zwitterionic nature of the DPPC polar headgroup towards protein adsorption has been ascertained by exposing either the phospholipid headgroup or the acyl chains to the protein solution; this was possible by the use of a silanized or a bare germanium crystal. Calibration curves have been made to measure the protein surface concentrations. After 3 h, the albumin surface concentration on DPPC monolayers was about three times higher when the proteins were exposed to the lipid acyl chains instead of the polar headgroups (e.g. 3 vs. 1 μg cm−2). As for fibrinogen (FGN) adsorption, when the lipid polar headgroups were exposed to the protein solution, the FGN adsorption was low reaching a maximum value of 0.5 μg cm−2. When interacting with the lipid acyl chains, the FGN adsorption reached a plateau at a value of 2.1 μg cm−2 after 3 h. Clearly, both albumin and FGN showed a low tendency to adsorb on surfaces where the lipid polar headgroups are exposed toward the protein solution.  相似文献

To evaluate the role of hydrophobic and electrostatic or other polar interactions for protein-ligand binding, we studied the interaction of human serum albumin (HSA) and beta-lactoglobulin with various aliphatic (C10-C14) cationic and zwitterionic detergents. We find that cationic detergents, at levels that do not cause unfolding, interact with a single site on beta-lactoglobulin and with two primary and five to six secondary sites on HSA with an affinity that is approximately the same as that with which zwitterionic (dimethylamineoxide) detergents interact, suggesting the absence of significant electrostatic interactions in the high-affinity binding of these compounds. The binding affinity for all of the groups of compounds was dependent upon hydrocarbon chain length, suggesting the predominant role of hydrophobic forces, supported by polar interactions at the protein surface. A distinct correlation between the binding energy and the propensity for micelle formation within the group of cationic or noncharged (nonionic and zwitterionic) detergents indicated that the critical micellar concentration (CMC) for each of these detergent groups, rather than the absolute length of the hydrocarbon chain, can be used to compare their hydrophobicities during their interaction with protein. Intrinsic fluorescence data suggest that the two primary binding sites on serum albumin for the zwitterionic and cationic compounds are located in the C-terminal part of the albumin molecule, possibly in the Sudlow II binding region. Comparisons with previous binding data on anionic amphiphiles emphasize the important contribution of ion bond formation and other polar interactions in the binding of fatty acids and dodecyl sulfate (SDS) by HSA but not by beta-lactoglobulin. Electrostatic interactions by cationic detergents played a significant role in destabilizing the protein structure at high binding levels, with beta-lactoglobulin being more susceptible to unfolding than HSA. Zwitterionic detergents, in contrast to the cationic detergents, had no tendency to unfold the proteins at high concentrations.  相似文献

We reported a method of determination of enantiomeric purity of the new potential antiviral agents by direct analytical HPLC. Those agents are nucleoside analogs, having one chiral center. They are synthesized as a single enantiomer (R or S) by an asymmetric pathway. The chiral stationary phases chosen are silica-based cellulose tris-3,5-dimethylphenylcarbamate (Chiralcel OD-H), or tris-methylbenzoate (Chiralcel OJ). Resolution was achieved using normal-phase chromatography with a mobile phase consisting of n-hexane-alcohol (ethanol or 2-propanol) in various percentages. Furthermore the effects of structural features on retention, selectivity and resolution, as well as on the elution order were thoroughly studied. Differences in the lipophilicity of the compounds were also examined.  相似文献
