Bacterial kidney disease (BKD) is a major health problem of salmonids, affecting both wild and cultured salmon. The disease is caused by Renibacterium salmoninarum (Rs), a fastidious, slow-growing and strongly Gram-positive diplobacillus that produces chronic, systemic infection characterized by granulomatous lesions in the kidney and other organs, often resulting in death. Fast detection of the pathogen is important to limit the spread of the disease, particularly in hatcheries or aquaculture facilities. Aptamers are increasingly replacing conventional antibodies as platforms for the development of rapid diagnostic tools. In this work, we describe the first instance of isolating and characterizing a ssDNA aptamer that binds with high affinity to p57 or major soluble antigen (MSA), the principal antigen found on the cell wall surface of Rs. Specifically, in this study a construct of the full-length protein containing a DNA binding domain (MSA-R2c) was utilized as target. Aptamers were isolated from a pool of random sequences using GO-SELEX (graphene oxide-systematic evolution of ligands by exponential enrichment) protocol. The selection generated multiple aptamers with conserved motifs in the random region. One aptamer with high frequency of occurrence in different clones was characterized and found to display a strong binding affinity to MSA-R2c with a Kd of 3.0 ± 0.6 nM. The aptamer could be potentially utilized for the future development of a sensor for rapid and onsite detection of Rs in water or in infected salmonids, replacing time-consuming and costly lab analyses.  相似文献

A new method for mass fabrication of silver ink conductivity detector electrodes for poly(methylmethacrylate) (PMMA) microchip electrophoretic systems has been developed based on screen‐printing technology. Printing of silver conductivity electrodes was performed through a patterned stencil on thin PMMA sheets. Following the electrode fabrication, the PMMA sheets are cut into cover sheets, and are aligned and sealed to the channel plate thus establishing a complete microchip separation device. The effects of the electrode width and spacing on the response and resolution have been investigated and the optimized electrode performance was compared to commonly used aluminum electrodes in the determination of ammonium, methyl ammonium, and sodium. The utility of the screen‐printed contactless conductivity detector (SPCCD) electrodes is further demonstrated for the separation and detection of organic acids with excellent reproducibility (RSD values of 3.7% and 4.1% for oxalate and tartrate, respectively). The thick‐film fabrication of the electrode material demonstrates the ability to mass‐fabricate detection devices with total process of device fabrication requiring less than 4 h (including the fabrication of channel plate, cover sheet with the electrodes, and subsequent bonding). The fabrication method described here is convenient and does not compromise the detector performance, hence offers great promise for producing single use field deployable analytical microsystems.  相似文献

The synthesis and magnetic properties of three new N(2)(3-) radical-bridged dilanthanide complexes, {[(Me(3)Si)(2)N](2)(THF)Ln}(2)(μ-η(2):η(2)-N(2))(-) (Ln = Tb, Ho, Er), are reported. All three display signatures of single-molecule-magnet behavior, with the terbium congener exhibiting magnetic hysteresis at 14 K and a 100 s blocking temperature of 13.9 K. The results show how synergizing the strong magnetic anisotropy of terbium(III) with the effective exchange-coupling ability of the N(2)(3-) radical can create the hardest molecular magnet discovered to date. Through comparisons with non-radical-bridged ac magnetic susceptibility measurements, we show that the magnetic exchange coupling hinders zero-field fast relaxation pathways, forcing thermally activated relaxation behavior over a much broader temperature range.  相似文献

Abstract— Five eudistomins, β-carboline derivatives isolated from a Caribbean tunicate, were tested for phototoxicity against several viruses, bacteria, yeast, and mammalian cells. The five compounds showed varying degrees of UVA dependant phototoxicity (i.e. long wavelength UV dependant) against murine cytomegalovirus (MCMV), Sindbis virus (SV) and mouse 3T3 cells, although the relative order of potency was the same for these three organisms. Eudistomin N was the most active (approximately the same as the β-carboline, harmine), while eudistomins M and O were moderately phototoxic, and H and I had little activity. To some degree the relative phototoxicity was correlated with fewer side chain substituents. A similar relative order of phototoxic potency was seen against phage T4, but in this case the magnitude of the effect was considerably reduced, in contrast to harmine. The antibacterial and antifungal activities were not correlated with antiviral effects, and some UVA-independent activities were seen. Thus the eudistomins may possess different mechanisms of action against different organisms, depending upon the presence or absence of UVA.  相似文献
