Pyrrolopyrimidine nucleosides VII. A study on electrophilic and nucleophilic substitution at position six of certain pyrrolo[2,3-d] pyrimidine nucleosides

A study involving the reactivity of the pyrrolo[2,3-d] pyrimidine ring system at position 6 with another exocyclic group (CN or -NH₂) already residing at C5 has established that hydrogen and bromine are susceptible to electrophilic and acid-catalyzed nucleophilic substitution, respect-tively. In one instance a strong nucleophile (hydrazine) gave nucleophilic substitution at position 6 which was followed by a reaction with the o-nitrile group to afford the tricyclic nucleoside 4,5-diamino-8-(β-D-ribofuranosyl)pyrazolo[3′, 4′ :5,4] pyrrolo[2,3-d] pyrimidine (4).     

Detection of a biomarker for Alzheimer's disease from synthetic and clinical samples using a nanoscale optical biosensor

A nanoscale optical biosensor based on localized surface plasmon resonance (LSPR) spectroscopy has been developed to monitor the interaction between the antigen, amyloid-beta derived diffusible ligands (ADDLs), and specific anti-ADDL antibodies. Using the sandwich assay format, this nanosensor provides quantitative binding information for both antigen and second antibody detection that permits the determination of ADDL concentration and offers the unique analysis of the aggregation mechanisms of this putative Alzheimer's disease pathogen at physiologically relevant monomer concentrations. Monitoring the LSPR-induced shifts from both ADDLs and a second polyclonal anti-ADDL antibody as a function of ADDL concentration reveals two ADDL epitopes that have binding constants to the specific anti-ADDL antibodies of 7.3 x 10(12) M(-1) and 9.5 x 10(8) M(-1). The analysis of human brain extract and cerebrospinal fluid samples from control and Alzheimer's disease patients reveals that the LSPR nanosensor provides new information relevant to the understanding and possible diagnosis of Alzheimer's disease.     

Configuration of MFI-type zeolite interacting with the probe molecule P(CH<Subscript>3</Subscript>)<Subscript>3</Subscript> – a theoretical study

With P(CH₃)₃ as the probe molecule adsorbed on titanium silicalite (TS-1) zeolite, the special and important role of T12 site in MFI-type zeolite was clearly elucidated. There are altogether three active sites present in TS-1 zeolite with Ti at the T12 site. Owing to the preferential adsorption of probe molecules on the first Brönsted acidic site, the Ti12 center will probably fail to show Lewis acidity. The ionic [HP(CH₃)₃]⁺ species can be stabilized by the first or second Brönsted acidic site, with the former energetically favored. The latter was formed through the transfer of the ionic [HP(CH₃)₃]⁺ species from the first to the second Brönsted acidic site.     

Seasonal variation and activity size distribution of <Superscript>7</Superscript>Be in ambient air

The seasonal variation of the ⁷Be activities in air and the size distribution of the ⁷Be aerosols were studied by using a continuous air sampler and a five stage cascade impactor. The mean monthly activity level of ⁷Be at the Korea Atomic Energy Research Institute (KAERI) site varied from 0.5 to 4.8 Bq·m⁻³ and revealed a seasonal variation, in which the ⁷Be activity levels were high in winter and low in summer. The mass size distribution showed a bi-modal distribution with a higher peak around 0.49 μm and a smaller peak between 3 μm and 7.2 μm. The activity median aerodynamic diameter (AMAD) decreased with increasing ambient ⁷Be concentrations. The mean residence times by using a mean growth rate of 0.004 μm·h⁻¹ were estimated to be 2.5–6.4 days. The AMAD has an increasing tendency with higher relative humidity. It seemed that the high humidity condition facilitated the growth of the aerosol, resulting in increased deposition rates of the aerosols and the low ⁷Be content in the surface air. The AMAD of the ⁷Be aerosols increased with an increasing temperature, but the temperature dependency of AMAD should be explained with geological and meteorological conditions.     

Exo-metal coordination by a tricyclic [(P(mu-N-2-NC5H4))2(mu-O)]2 dimer in [(P(mu-N-2-NC5H4))2(mu-O)]2(CuCl x (C5H5N)2)4 (2-NC5H4 = 2-pyridyl, C5H5N = pyridine)

The in situ reaction of the phosphazane dimer [CIP(mu-N-2-NC5H4)]2 (2) with CuCl in the presence of CsH5N/H2O gives the title complex [(P(mu-N-2-NC5H4))2(mu-O)]2(CuCl x (C5H5N)2)4 (1), containing a tricyclic [(P(mu-N-2-NC5H4))2(mu-O)]2 ligand which is isoelectronic with species of the type [(P(mu-NR))2NR]2.     

Structure cristalline et conformation en solution aqueuse de la carnosine (β-alanyl-L-histidine)

Carnosine (β-alanyl-L-histidine) is a biologically active molecule involved in muscular metabolism. It crystallises in the C; space group with a = 24.725 Å b = 5,427 Å c = 8,004 Å β = 100,2° (Z = 4)

In the crystal, acid and basic groups are engaged in hydrogen bonds whose strength is evaluated through IR frequencies. Molecular conformation in the solid state is defined by τ₁ = /t-177° τ₂ = −38° φ = −96° ψ = +131° χ₁ = 181° χ₂₁ = 62°

NMR study of carnosine in aqueous solution indicates that rotation about CH₂-CH₂ is free and that the other angles take the following values: Ø −150° or −90° and X₁ = 165° or 315°. Infrared and Raman spectra suggest that τ₂ undergoes small changes when going from crystal to solution while ψ is close to +150°.     

Resonance-enhanced multiphoton ionization of molecular hydrogen via the E,F1Σg+ state: Photoelectron energy and angular distributions

Photoelectron energy and angular distributions are measured for the 2+1 multiphoton ionization process H₂ X¹Σ_g⁺ (ν = 0,J) + 2hv → E,F¹Σ_g⁺(ν_E,J_E = J) + hν → H₂⁺X²Σ_g⁺ (ν⁺) + e^?, for ν_E = 0, 1, or 2 and for J_E = 0 or 1 of the inner well of the double-minimum E,F state. Although a strong preference is found for ν⁺ = ν_E, the detailed H₂⁺ vibrational distribution does not exhibit Franck-Condon behavior, and the photoelectron angular distributions vary markedly with both the J_E value of the intermediate state and the ν⁺ value of the ion.     

On the structure of CH5+; A study of hydron transfer reactions from CH4H+ and CD4H+ tandem-ICR

Relative rates of proton and deuteron transfer from CH₄D⁺ and CD₄H⁺ to a number of molecules were examined in a tandem-ion cyclotron resonance instrument. The results were in conflict with the recent work of Sefcik et al. and support a randomized model.     

Steric control in the oligomerisation of phosphazane dimers; towards new phosphorus-nitrogen macrocycles

The reaction of [ClP(mu-NtBu)]2 (1) with H2O (1 : 2 equivalents) in the presence of excess Et3N gives the new chain compound [(mu-O)[P(mu-NtBu)2P(H)=O]2] (3), consisting of two P2N2 rings linked by a mu-O atom and terminating in P(V)(H)=O groups. A similar chain species is obtained from the reaction of the lithiate of [(tBuNH)P(mu-NtBu)2P(H)=O] (5) with [ClP(mu-NtBu)2P(NHtBu)] (2), the product being [(mu-O)[P(mu-NtBu)2P(NHtBu)]2] (6). Compounds 3 and 6 are the first examples of O-bridged chain phosphazanes and potential precursors to new phosphorus-nitrogen macrocycles. The syntheses and X-ray structures of 3, 5 and 6 are reported.
