Fifty 2-[(arylidene)amino]-4,5-cycloalkyl[b]thiophene-3-carbonitrile derivatives were screened for their in vitro antifungal activities against Candida krusei and Cryptococcus neoformans. Based on experimentally determined minimum inhibitory concentration (MIC) values, we conducted computer-aided drug design studies [molecular modelling, chemometric tools (CPCA, PCA, PLS) and QSAR-3D] that enable the prediction of three-dimensional structural characteristics that influence the antifungal activities of these derivatives. These predictions provide direction with regard to the syntheses of new derivatives with improved biological activities, which can be used as therapeutic alternatives for the treatment of fungal infections.  相似文献

Three aminodi(hetero)arylamines were prepared via a palladium-catalyzed C-N Buchwald-Hartwig coupling of methyl 3-aminothieno[3,2-b]pyridine-2-carboxylate with different bromonitrobenzenes, followed by reduction of the nitro groups of the coupling products to the corresponding amino compounds. The aminodi(hetero)arylamines thus obtained were evaluated for their growth inhibitory effect on four human tumor cell lines MCF-7 (breast adenocarcinoma), A375-C5 (melanoma), NCI-H460 (non-small cell lung cancer) and HepG(2) (hepatocellular carcinoma). The toxicity to non-tumor cells was also evaluated using a porcine liver primary cell culture (PLP1), established by us. The aminodi(hetero)arylamine with the NH(2) group in the ortho position and an OMe group in the para position to the NH of the di(hetero)arylamine, is the most promising compound giving the lowest GI(50) values (1.30-1.63 μM) in all the tested human tumor cell lines, presenting no toxicity to PLP1 at those concentrations. The effect of this compound on the cell cycle and induction of apoptosis was analyzed in the NCI-H460 cell line. It was observed that it altered the cell cycle profile causing a decrease in the percentage of cells in the G0/G1 phase and an increase of the apoptosis levels.  相似文献

An experimental and theoretical study of the photoionization energies (IE's) of Ba(H(2)O)(n) clusters containing up to n = 4 water molecules has been performed. The clusters were generated by a pick-up source combining laser vaporization with pulsed supersonic expansion, and then photoionized by radiation of 272.5-340 nm. The experimentally determined IE(e)'s for n = 1 to 4 are 4.56 ± 0.05, 4.26 ± 0.05, 3.90 ± 0.05 and 3.71 ± 0.05 eV. This cluster size dependence of IE is reproduced within ±0.06 eV employing the mPW1PW91 density-functional and CCSD(T, Full) quantum-chemical methods combined with the 6-311++G(d,p) basis set for the H and O atoms and three different relativistic effective core potentials for Ba atoms. The calculations indicate that the lowest energy hydration structures represent the most relevant contributions to both the vertical and adiabatic ionization energies. Experimental and theoretical evidence correlates with the progressive surface-delocalization of the electron from the hydration cavity around the Ba atom and suggests that the intra-cluster electron transfer is possible even for small aggregates.  相似文献

Paraquat (PQ) is an herbicide implicated in numerous fatalities, mainly caused by voluntary ingestion. Several methods have been used to quantify PQ in plasma and urine samples of intoxicated humans as a predictor of clinical outcome. There is no validated method for the analysis of PQ in postmortem samples. Therefore, the aim of this study was to develop an analytical method, using gas chromatography–ion trap mass spectrometry (GC‐IT/MS) after solid‐phase extraction, to quantify PQ in postmortem samples, namely in whole blood, urine, liver, lung and kidney, to cover the routes of distribution, accumulation and elimination of PQ. The method proved to be selective as there were no interferences of endogenous compounds with the same retention time as PQ and ethyl paraquat (internal standard). The regression analysis for PQ was linear in the range 0–10 µg/mL. The detection limits ranged from 0.0076 µg/mL for urine to 0.047 µg/mL for whole blood, and the recoveries were suitable for forensic analysis. The proposed GC‐IT/MS method provided an accurate and simple assay with adequate precision and recovery for the quantification of PQ in postmortem samples. The proof of applicability was performed in two fatal PQ intoxications. A review of the analytical methods for the determination of quaternary ammonium herbicides is also provided for a better understanding of the presently available techniques. Cutinases are versatile carboxylic ester hydrolases with great potential in many biocatalytic processes, including biodiesel production. Genome sequence analysis of the model organism Aspergillus nidulans reveals four genes encoding putative cutinases. In this work, we purified and identified for the first time a cutinase (ANCUT2) produced by A. nidulans. ANCUT2 is a 29-kDa protein which consists of 255 amino acid residues. Comparison of the amino acid sequence of ANCUT2 with other microbial cutinase sequences revealed a high degree of homology with other fungal cutinases as well as new features, which include a serine-rich region and conserved cysteines. Cutinase production with different lipidic and carbon sources was also explored. Enzyme activity was induced by olive oil and some triacylglycerides and fatty acids, whereas it was repressed by glucose (1%) and other sugars. In some conditions, a 22-kDa post-translational processing product was also detected. The cutinase nature of the enzyme was confirmed after degradation of apple cutin.  相似文献

The crystal structure and dielectric properties of slowly cooled A-site-deficient perovskites Li(x)Sr(x)La(2/3-x)□(1/3-x)TiO(3) (0.04 ≤ x ≤ 0.33) have been investigated by powder X-ray diffraction (XRD), impedance spectroscopy, and (7)Li NMR techniques. In this series, nominal vacancies decrease with Li content, but the total amount of A-site vacancies, n(t) = Li + □, participating in conduction processes remains basically constant. Rietveld analysis of the XRD patterns showed a change of symmetry from orthorhombic to tetragonal when the lithium and strontium contents increased above x = 0.08 and from tetragonal to cubic above x = 0.16. Structural modifications are mainly due to the cation vacancy ordering along the c axis, which disappear gradually when the lithium content increases. In agreement with the structural information, two lithium signals with different quadrupole constants are detected in (7)Li NMR spectra of orthorhombic/tetragonal phases, which have been associated with lithium in two crystallographic z/c = 0 and 1/2 planes of perovskites. In cubic samples, only a single narrow component, indicative of mobile species, was detected. Lithium motion was thermally activated, with activation energies going from 0.35 to 0.38 eV. Evolution of the bulk dc-conductivity preexponential factors along the series showed a maximum that has been first related to the dependence of lithium hopping on the lithium and vacancy concentrations. Finally, changes in the vacancy ordering, produced along the series, affect the dimensionality of the conductivity, indicating that not only the amount of vacancies but also its distribution are relevant.  相似文献

Incorporation of long path length liquid waveguide capillary cell (LWCC or LCW) into spectrometric detection systems can increase the sensitivity of these by orders of magnitude (up to 500 times), and consequently can reduce the detection limits. The combination of the long path length spectrophotometry with flow methodologies can provide analytical solutions for various challenges in the field of environmental, biochemical and food chemistry.  相似文献

Based on green analytical chemistry principles, an efficient approach was applied for the simultaneous determination of galantamine, a widely used cholinesterase inhibitor for the treatment of Alzheimer's disease, and its major metabolites in serum samples. After a simple serum deproteinization step, second-order data were rapidly obtained (less than 6 min) with a chromatographic system operating in the isocratic regime using ammonium acetate/acetonitrile (94:6) as mobile phase. Detection was made with a fast-scanning spectrofluorimeter, which allowed the efficient collection of data to obtain matrices of fluorescence intensity as a function of retention time and emission wavelength. Successful resolution was achieved in the presence of matrix interferences in serum samples using multivariate curve resolution-alternating least-squares (MCR-ALS). The developed approach allows the quantification of the analytes at levels found in treated patients, without the need of applying either preconcentration or extraction steps. Limits of detection in the range between 8 and 11 ng mL−1, relative prediction errors from 7 to 12% and coefficients of variation from 4 to 7% were achieved.  相似文献
