This study aimed to evaluate and compare the effects of co-treatment with purified annatto oil (PAO) or its granules (GRA, Chronic®) with that of testosterone on the orchiectomy-induced osteoporosis in Wistar rats. After surgery, rats were treated from day 7 until day 45 with testosterone only (TES, 7 mg/kg, IM) or TES + PAO or GRA (200 mg/kg, p.o.). The following parameters were evaluated: food/water intake, weight, HDL, LDL, glucose, triglycerides (TG), total cholesterol (TC), alkaline phosphatase levels, blood phosphorus and calcium contents, femur weight, structure (through scanning electron microscopy), and calcium content (through atomic absorption spectrophotometry). Our results show that orchiectomy could significantly change the blood lipid profile and decrease bone integrity parameters. Testosterone reposition alone could improve some endpoints, including LDL, TC, bone weight, and bone calcium concentration. However, other parameters were not significantly improved. Co-treatment with PAO or GRA improved the blood lipid profile and bone integrity more significantly and improved some endpoints not affected by testosterone reposition alone (such as TG levels and trabeculae sizes). The results suggest that co-treatment with annatto products improved the blood lipid profile and the anti-osteoporosis effects of testosterone. Overall, GRA had better results than PAO.  相似文献

Nowadays, there is increasing interest in natural antioxidants from food by‐products. Astaxanthin is a potent antioxidant and one of the major carotenoids in crustaceans and salmonids. An ultra‐high pressure liquid chromatographic method was developed and validated for the determination of astaxanthin in shrimp by‐products, and its migration from new packaging materials to food simulants was also studied. The method uses an UPLC® BEH guard‐column (2.1 × 5 mm, 1.7 µm particle size) and an UPLC® BEH analytical column (2.1 × 50 mm, 1.7 µm particle size). Chromatographic separation was achieved using a programmed gradient mobile phase consisting of (A) acetonitrile–methanol (containing 0.05 m ammonium acetate)–dichloromethane (75:20:5, v/v/v) and (B) ultrapure water. This method was evaluated with respect to validation parameters such as linearity, precision, limit of detection, limit of quantification and recovery. Low‐density polyethylene films were prepared with different amounts of the lipid fraction of fermented shrimp waste by extrusion, and migration was evaluated into food simulants (isooctane and ethanol 95%, v/v). This study investigated the effects of low‐level laser therapy (LLLT) in the liver function, structure and inflammation in a experimental model of carbon tetrachloride (CCl4)‐induced liver cirrhosis. Wistar rats were divided into Control, LLLT, CCl4 and CCl4+LLLT groups. CCl4 groups received CCl4 (0.4 g kg?1; i.p.), three times a week, for 12 weeks. A 830 nm LLLT was performed with a continuous wave, 35 mW, 2.5 J cm?2 per point, applied to four points of the liver (right and left upper and lower extremities, in the four lobes of the liver) for 2 weeks. Liver structure and inflammation (cirrhotic areas, collagen deposition, inflammation, density of Kupffer and hepatic stellate cells) and function (aspartate aminotransferase, alkaline phosphatase, gamma glutamyltransferase, lactate dehydrogenase, total proteins and globulins) were evaluated. LLLT significantly reduced CCl4‐increased aspartate aminotransferase (P < 0.001), alkaline phosphatase (P < 0.001), gamma‐glutamyl transferase (P < 0.001) and lactate dehydrogenase (P < 0.01) activity, as well as total proteins (P < 0.05) and globulins (P < 0.01). LLLT also reduced the number of cirrhotic areas, the collagen accumulation and the hepatic inflammatory infiltrate. Of note, LLLT reduced CCl4‐increased number of Kupffer cells (P < 0.05) and hepatic stellate cells (P < 0.05). We conclude that LLLT presents beneficial effects on liver function and structure in an experimental model of CCl4‐induced cirrhosis.  相似文献

We present an experimental study of the transient periodic structures appearing in the nematic director field in the magnetically induced reorientation of the director in the vicinity of the twist Fréedericksz geometry. Thin nematic samples (50?µm thick) were exposed to magnetic fields of variable intensity and orientation relative to the surface aligning direction of the sample. The director reorientation was induced by a rapid rotation of the sample in the static magnetic field producing a misalignment between the director and the magnetic field. The director field was optically monitored during the reorientation process and the transient periodic structures were characterized. Two types of periodic structures could be identified, namely bands and walls. Walls grow from bands close to the twist Fréedericksz geometry. The time dependence of the wave length and inclination of the periodic structures was obtained as a function of the magnetic field intensity and orientation relative to the surface aligning direction of the sample. The results for the bands are compared with the predictions of a model that we specifically developed to account for the non-orthogonal field orientations. It is seen that our model can account rather well for the experimental results considering that it uses only the field rotation time as adjustable parameter. The use of microbial biomass in biosorption is already being studied as a potential alternative to (or combined with) conventional processes, where several algae and microorganisms have already shown promising ability to uptake metals. Cyanobacteria (blue-green algae) are widespread organisms, with specific properties, such as high nutrient removal capacity and tolerance to highly variable conditions which make them well-suited for wastewater and remediation purposes. The main aim of this work was to evaluate the use of a marine cyanobacterium LEANCYA 21 (Synechocystis sp.), collected from the Portuguese southern border, for the removal of selected trace metals when in natural seawater culture medium. It was observed, for the first time, that this particular strain is capable of uptaking Pb, Ni and Zn (at nM levels) from seawater solutions using small amounts of biomass. Uptake values for Pb were up to 90% (0.75 mg g?1 biomass) in 6 h. The specific biosorption curves of Ni and Zn showed that these metals follow a first order kinetics biosorption in batch conditions. Solutions containing multimetals have revealed that Ni uptake is affected by the presence of Pb and Zn. The calculated specific absorption values were high enough to predict a possible application in aquaculture where such low levels of metals may inhibit microalgae growth.  相似文献

The present study describes the simple and fast preparation of Cerium (III) hexacyanoferrate (II) (CeHCF) solid nanoparticles at three different water/formamide (%) ratios used as solvent (v/v) (100:0, 80:20, 0:100). CeHCF nanoparticles (Nps) were characterized by fourier transform infrared pectroscopy (FTIR), x-ray diffraction (XRD), scanning electron microscopy (SEM), zeta potential and cyclic voltammetry (CV). Electrodes modified with CeHCF presented a well-defined redox pair with formal potential (Eo′) of approximately 0.29 V (vs. Ag/AgCl(sat) attributed to the Fe2 +/Fe3+ redox pair in the presence of cerium (III)). The Nps in the three systems investigates, presents a random size distribution to different surface, where most were distributed between 20 and 160 nm. Considering the three investigated systems, only CeHCF-1 (100:0) was sensitive to L-dopamine, presenting a linear signal region as a function of L-dopamine concentrations, with a limit of detection (LD) of 0.125 mmol L−1, limit of quantification (LQ) of 0.419 mmol L−1 and amperometric sensitivity (S) of 148.16 μA mmol L−1.  相似文献

The unbridled dissemination of multidrug-resistant pathogens is a major threat to global health and urgently demands novel therapeutic alternatives. Antimicrobial photodynamic therapy (aPDT) has been developed as a promising approach to treat localized infections regardless of drug resistance profile or taxonomy. Even though this technique has been known for more than a century, discussions and speculations regarding the biochemical mechanisms of microbial inactivation have never reached a consensus on what is the primary cause of cell death. Since photochemically generated oxidants promote ubiquitous reactions with various biomolecules, researchers simply assumed that all cellular structures are equally damaged. In this study, biochemical, molecular, biological and advanced microscopy techniques were employed to investigate whether protein, membrane or DNA damage correlates better with dose-dependent microbial inactivation kinetics. We showed that although mild membrane permeabilization and late DNA damage occur, no correlation with inactivation kinetics was found. On the other hand, protein degradation was analyzed by three different methods and showed a dose-dependent trend that matches microbial inactivation kinetics. Our results provide a deeper mechanistic understanding of aPDT that can guide the scientific community toward the development of optimized photosensitizing drugs and also rationally propose synergistic combinations with antimicrobial chemotherapy.  相似文献

The silicoaluminophosphate SAPO-37 as well as ZSM-20 zeolite have a faujaside-like structure and are potentially useful as catalysts. We report in this work different procedures to decompose the organic template occluded in SAPO-37. The texture of the final products was analyzed by means of nitrogen adsorption. The acid properties of SAPO-37 and ZSM-20 were also compared by IR measurements of adsorbed pyridine and related to the acidic properties of the presently commonly used cracking catalyst zeolite Y.
