Paracoccidioidomycosis (PCM) is the most prevalent human mycosis in Latin America. The infection is thought to take place firstly in the lungs and then may disseminate to other organs and tissues. Treatment by currently available antifungals is lengthy, the drugs may have undesirable side effects, and some are costly. Occasional resistant strains of Paracoccidioides brasiliensis, the causative agent of PCM, have been reported. So, the search for more efficient treatments or adjuvant therapies has to be continued. In this work, we evaluated the effects of HeNe laser irradiation on cutaneous inflammatory lesions caused by the inoculation of 5 x 10(6)/0.1ml yeasts cells into the back footpad of Balb/c mice. HeNe irradiation (lambda=632.8nm, 3mW, incident energy of 3J/cm(2)) was applied at days 7, 8 and 9 post-infection and histological and immunohistochemical analysis were done. Unirradiated animals were used as controls. The results showed that laser-treated mice presented reduction of footpad edema, faster cutaneous wound healing, confluent granuloma, diffuse- and more loosely distributed immunolabeling for TNF-alpha, enhanced labeling of IFN-gamma and any P. brasiliensis form detected, whereas multiple viable fungi were seen in diffuse widespread granulomas obtained from non-treated mice foot-pad. Fungi that were harvested from laser-treated animals presented no capability of growth in vitro as compared to those obtained from non-treated mice. We conclude that HeNe laser irradiation was able to inhibit the progress of inflammatory local reaction produced by P. brasiliensis infection and influence local cytokines production. We suggest that this treatment modality can be a useful coadjuvant tool to be combined with antifungal agents in the treatment of PCM ulcerations. The mechanisms involved in laser therapy of PCM lesions need further investigation. 相似文献

The formation of a new hybrid material based on titanium dioxide as inorganic support and containing an iron organochelator (ICL670) is described. An organophosphorous coupling agent was used to graft the organic molecule on the oxide surface. The attachment of the organic substrate was well-confirmed by FTIR (DRIFT), solid-state 31P and 13C CPMAS NMR, thermal analysis and the integrity of the structural and morphological parameters were verified using XRD and TEM analyses. The interaction between the material and dissolved iron(III) was also investigated through potentiometric measurements and demonstrated the interest of this new non-siliceous based hybrid material. The obtained linear evolution of the open circuit potential from 10−2 to 10−6 mol L−1 can be used for the analytical detection of iron(III). 相似文献

This paper presents an optimization model for downsizing a multi-product supply chain facing bankruptcy risk, where multi-functional production facilities are shared for producing a group of substitutable products. In order to determine the potential demand after discontinuation of certain product types, the proposed mixed integer programming model incorporates a new general formulation of demand substitution, which allows arbitrary demand diversion and arbitrary replacement rates between products under investigation. The new substitution formulation enables considering uneven substitutions for supply chain network design and therefore it extends currently used substitution approaches. A series of systematically generated downsizing cases confirm the validity of the here proposed approach, and illustrate how downsized chains can successfully adopt to the new circumstances. 相似文献

The advanced development of cell carriers for regenerative medicine and cell therapy demands materials able to sustain cell viability prior to their delivery to the target tissue, an ability that can be controlled by the shape, size and degradability of the matrix. TEMPO-oxidized nanofibrillar cellulose (ToNFC) macromolecules are negatively charged and therefore can be easily formulated by ionotropic gelation into beads of varying sizes that can release their payload through an erosion-controlled process. We report here for the first time on the preparation of ToNFC beads via ionic gelation using CaCl2 and on their loading with OSTEO-1 rat bone cells, with a view to examine their capacity of sustaining the cell viability and of releasing the bone cells in a controlled manner. The initial results obtained demonstrate that ToNFC is able to protect the OSTEO-1 cells and to maintain their viability for at least 2 weeks. Following gradual disintegration of the beads, a significant cell release and subsequent proliferation was observed after 7 days. These results indicate the considerable potential of nanofibrillar cellulose (ToNFC) for applications in cell therapy and regenerative medicine. 相似文献
