In this work, a downsized flow set up designed based on multicommutation concept for photometric determination of iron(II)/iron(III) and nitrite/nitrate is surface water is described. The flow system network comprised a set of three-way solenoid valves, reaction coil and a double-channel flow cell, which were nested in order to obtain a compact and small-size instrument. To accomplish the downsizing requirement light source (LED) and radiation detection (phototransistor) were coupled to the flow cell. In order to demonstrated the effectiveness of the system, the photometer methods based on Griess reaction and 1-10-phenantroline for nitrite and iron(II) determination, respectively, were selected. Under computer control the set up provided facilities to handle four reagent solutions employing a single pumping channel, thus permitting also the determination of nitrate and iron(III) after its reduction to nitrite and to iron(II), respectively. The overall system performance was demonstrated working several days running standard solution, no significant variation of base line, linear response range and slop (less than 1%) were observed. The usefulness of the downsized system was ascertained by analyzing a set of surface water. Aiming to access the accuracy sample were also analyzed employing reference procedures and no significant difference at 95% confidence level were observed for the four analytes. Other profitable features such as analytical throughput of 40 determination per hour; relative standard deviation of 1%; linear response range between 50 and 300 μg l−1 for nitrite and nitrate, 0.5-6.0 mg l−1 iron(II) and iron(III); low reagent consumption 75 μg for nitrate/nitrite and 0.6 mg for iron(II)/iron(III) per determination; and 2.4 ml waste generation per determination were also achieved. 相似文献

An automatic system that performs two analytical procedures, allowing the evaluation of the relative antioxidant capacity of wine samples, was developed. Automation was carried out using a sequential injection analysis (SIA) system that allowed, thanks to its versatility, the development of two methodologies. One is based on the decolorization assay of the 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical, using a spectrophotometric detector. A second methodology allowed the evaluation of the hydrogen peroxide scavenging activity by measuring the oxidation of homovanylic acid (HVA) to its fluorescent dimer, using a fluorescent detector.The developed automatic methodologies were evaluated using trolox as standard and subsequently using other antioxidant substances as gallic acid, caffeic acid, ascorbic acid, catechin and taxifolin which are abundant in wine and whose antioxidant activities were compared to that shown by trolox. The spectrophotometric and fluorimetric assays showed linearity intervals between 0.001 and 0.01 mM, and 0.001 and 0.008 mM of trolox, respectively.The evaluation of the antioxidant power of 20 white and red wine samples, from different Portuguese wine producing regions, was carried out sequentially, in the automatic system. The results were expressed in trolox equivalent antioxidant capacity (TEAC) and presented, for the ABTS and hydrogen peroxide scavenging activity methodologies, detection limits of 8.4 × 10−7 and 1.4 × 10−4 mM and relative standard deviation (R.S.D. In flow injection analysis, several plugs can be injected simultaneously into the same carrier system, in order to achieve overlapped zones. The potentialities of such sequential injections are studied in model systems with spectrophotometric detection; the procedure is shown to give better precision than the use of concentration gradients. Vanadium(IV or V) complexes with N,O- or O,O-ligands, i.e., [VO{N(CH2CH2O)3}], Ca[V(HIDPA)2] (synthetic amavadine), Ca[V(HIDA)2], or [Bu4N]2[V(HIDA)2] [HIDPA, HIDA = basic form of 2,2'-(hydroxyimino)dipropionic or -diacetic acid, respectively], [VO(CF3SO3)2], Ba[VO(nta)(H2O)]2 (nta = nitrilotriacetate), [VO(ada)(H2O)] (ada = N-2-acetamidoiminodiacetate), [VO(Hheida)(H2O)] (Hheida = 2-hydroxyethyliminodiacetate), [VO(bicine)] [bicine = basic form of N,N-bis(2-hydroxyethyl)glycine], and [VO(dipic)(OCH2CH3)] (dipic = pyridine-2,6-dicarboxylate), are catalyst precursors for the efficient single-pot conversion of methane into acetic acid, in trifluoroacetic acid (TFA) under moderate conditions, using peroxodisulfate as oxidant. Effects on the yields and TONs of various factors are reported. TFA acts as a carbonylating agent and CO is an inhibitor for some systems, although for others there is an optimum CO pressure. The most effective catalysts (as amavadine) bear triethanolaminate or (hydroxyimino)dicarboxylates and lead, in a single batch, to CH3COOH yields > 50% (based on CH4) or remarkably high TONs up to 5.6 x 103. The catalyst can remain active upon multiple recycling of its solution. Carboxylation proceeds via free radical mechanisms (CH3* can be trapped by CBrCl3), and theoretical calculations disclose a particularly favorable process involving the sequential formation of CH3*, CH3CO*, and CH3COO* which, upon H-abstraction (from TFA or CH4), yields acetic acid. The CH3COO* radical is formed by oxygenation of CH3CO* by a peroxo-V complex via a V{eta1-OOC(O)CH3} intermediate. Less favorable processes involve the oxidation of CH3CO* by the protonated (hydroperoxo) form of that peroxo-V complex or by peroxodisulfate. The calculations also indicate that (i) peroxodisulfate behaves as a source of sulfate radicals which are methane H-abstractors, as a peroxidative and oxidizing agent for vanadium, and as an oxidizing and coupling agent for CH3CO* and that (ii) TFA is involved in the formation of CH3COOH (by carbonylating CH3*, acting as an H-source to CH3COO*, and enhancing on protonation the oxidizing power of a peroxo-VV complex) and of CF3COOCH3 (minor product in the absence of CO). 相似文献

In this work, we propose the utilization of scCO2 to impregnate ibuprofen into the mcl-PHA matrix produced by Pseudomonas chlororaphis subs. aurantiaca (DSM 19603). The biopolymer has adhesive properties, is biocompatible and has a melting temperature of 45 °C. Several conditions, namely, pressure (15 and 20 MPa) and impregnation time (30 min, 1 h and 3 h) were tested. The highest ibuprofen content (90.8 ± 6.5 mg of ibuprofen/gPHA) was obtained at 20 MPa and 40 °C, for 1 h, with an impregnation rate of 89 mg/(g·h). The processed mcl-PHA samples suffered a plasticization, as shown by the decrease of 6.5 °C in the Tg, at 20 MPa. The polymer’s crystallinity was also affected concomitantly with the matrices’ ibuprofen content. For all the impregnation conditions tested the release of ibuprofen from the biopolymer followed a type II release profile. This study has demonstrated that the mcl-PHA produced by P. chlororaphis has a great potential for the development of novel topical drug delivery systems. 相似文献

Purpose: Vascular targeted photodynamic therapy (VTP) is a nonsurgical tumor ablation approach used to treat early-stage prostate cancer and may also be effective for upper tract urothelial cancer (UTUC) based on preclinical data. Toward increasing response rates to VTP, we evaluated its efficacy in combination with concurrent PD-1 inhibitor/OX40 agonist immunotherapy in a urothelial tumor-bearing model. Experimental design: In mice allografted with MB-49 UTUC cells, we compared the effects of combined VTP with PD-1 inhibitor/OX40 agonist with those of the component treatments on tumor growth, survival, lung metastasis, and antitumor immune responses. Results: The combination of VTP with both PD-1 inhibitor and OX40 agonist inhibited tumor growth and prolonged survival to a greater degree than VTP with either immunotherapeutic individually. These effects result from increased tumor infiltration and intratumoral proliferation of cytotoxic and helper T cells, depletion of Treg cells, and suppression of myeloid-derived suppressor cells. Conclusions: Our findings suggest that VTP synergizes with PD-1 blockade and OX40 agonist to promote strong antitumor immune responses, yielding therapeutic efficacy in an animal model of urothelial cancer. 相似文献

Nitrogen-bearing rings are common features in the molecular structures of modern drugs, with chiral δ-lactams being an important subclass due to their known pharmacological properties. Catalytic dearomatization of preactivated pyridinium ion derivatives emerged as a powerful method for the rapid construction of chiral N-heterocycles.
