Polyphenol Oxidase as a Promising Alternative Therapeutic Agent for Cancer Therapy

Cancers have always been the most difficult to fight, the treatment of cancer is still not considered. Thus, exploring new anticancer drugs is still imminent. Traditional Chinese medicine has played an important role in the treatment of cancer. Polyphenol oxidase (PPO) extracted from Edible mushroom has many related reports on its characteristics, but its role in cancer treatment is still unclear. This study aims to investigate the effects of PPO extracted from Edible mushroom on the proliferation, migration, invasion, and apoptosis of cancer cells in vitro and explore the therapeutic effects of PPO on tumors in vivo. A cell counting kit-8 (CCK8) assay was used to detect the effect of PPO on the proliferation of cancer cells. The effect of PPO on cancer cell migration ability was detected by scratch test. The effect of PPO on the invasion ability of cancer cells was detected by a transwell assay. The effect of PPO on the apoptosis of cancer cells was detected by flow cytometry. Female BALB/c mice (18–25 g, 6–8 weeks) were used for in vivo experiments. The experiments were divided into control group, model group, low-dose group (25 mg/kg), and high-dose group (50 mg/kg). In vitro, PPO extracted from Edible mushroom significantly inhibited the proliferation, migration, and invasion capability of breast cancer cell 4T1, lung cancer cell A549, and prostate cancer cell C4-2, and significantly promoted the apoptosis of 4T1, A549, and C4-2. In vivo experiments showed PPO inhibitory effect on tumor growth. Collectively, the edible fungus extract PPO could play an effective role in treating various cancers, and it may potentially be a promising agent for treating cancers.     

MD Simulation Studies for Selective Phytochemicals as Potential Inhibitors against Major Biological Targets of Diabetic Nephropathy

Diabetes is emerging as an epidemic and is becoming a public health concern worldwide. Diabetic nephropathy is one of the serious complications of diabetes, and about 40% of individuals with diabetes develop diabetic nephropathy. The consistent feature of diabetes and its associated nephropathy is hyperglycemia, and in some cases, hyperamylinemia. Currently, the treatment includes the use of medication for blood pressure control, sugar control, and cholesterol control, and in the later stage requires dialysis and kidney transplantation, making the management of this complication very difficult. Bioactive compounds, herbal medicines, and extracts are extensively used in the treatment and prevention of several diseases, and some are reported to be efficacious in diabetes too. Therefore, in this study, we tried to identify the therapeutic potential of phytochemicals used in in silico docking and molecular dynamic simulation studies using a library of 5284 phytochemicals against the two potential targets of type 2 diabetes-associated nephropathy. We identified two phytochemicals (i.e., gentisic acid and michelalbine) that target human amylin peptide and dipeptidyl peptidase-4, respectively, with good binding affinity. These phytochemicals can be further evaluated using in vitro and in vivo studies for their anti-hyperglycemia and anti-hyperamylinemia effects.

Two-dimensional subband Steiglitz–McBride algorithm for automatic analysis of two-dimensional nuclear magnetic resonance data

Rapid, accurate, and automatic quantitation of two-dimensional nuclear magnetic resonance(2D-NMR) data is a challenging problem. Recently, a Bayesian information criterion based subband Steiglitz–McBride algorithm has been shown to exhibit superior performance on all three fronts when applied to the quantitation of one-dimensional NMR free induction decay data. In this paper, we demonstrate that the 2D Steiglitz–McBride algorithm, in conjunction with 2D subband decomposition and the 2D Bayesian information criterion, also achieves excellent results for 2D-NMR data in terms of speed, accuracy, and automation—especially when compared in these respects to the previously published analysis techniques for 2D-NMR data.

NMR Spectroscopic Study of Camphanic Acid,Rh2[Camphanate]4, and its Adducts with 1,4-Benzodiazepines

The H and ¹³C NMR spectra of chiral camphanic acid, lactonc of (1S,3R)-1-hydroxy-2,2,3-trimethyl-cyclopentan-1,3-dicarboxylic acid (1), and dirhodium tetracamphanate complex (2) were completely assigned on the basis of one- and two-dimensional NMR experiments. The NMR spectra of the adducts of dirhodium tetracamphanate 2 with 5-pyrido-1, 4-benzodiazepin-2-ones 3 and 4, complexes with catalytic activity, support the assignation and also revealed two different types of axial complexation of these nitrogen ligands.
