Summary An overview of the activities of the Polish Institute of Nuclear Chemistry and Technology in the field of preparation and certification of reference materials for multielement trace analysis is presented. A general strategy has been worked out and is briefly described with more detailed emphasis on the problem of data evaluation. Two geological-environmental materials recently issued [i.e. an Apatite Concentrate (CTA-AC-1) and Fine Fly Ash (CTA-FFA-1)] which were certified for 25 and 40 elements, respectively, are described. The preparation and testing of a new CRM of biological origin Oriental Tobacco Leaves (CTA-OTL-1) is presented in detail, including study of the optimum methods for drying, checking homogeneity, radiation sterilization etc. Interlaboratory comparison, in which 61 laboratories participated, yielded results enabling recommended (certified) values for 29 elements and information values for further 11 elements to be established. 相似文献

We compared cryopreservation of mammalian neural stem cells (NSCs) cultured as neurospheres by slow-cooling (1 C/min) in 10% (v/v) DMSO and cryopreservation by immersion into liquid nitrogen in ethylene glycol (EG)-sucrose solutions that support vitrification (40% (v/v) EG, 0.6 M sucrose) or that do not (37% v/v) EG, 0.6 M sucrose and 30% (v/v) EG, 0.6 M sucrose); the concentration of penetrating cryoprotectant in the last two solutions was lowered with the intention to reduce their toxicity towards NSCs. To protect against contamination a straw-in-straw technique was employed. Vitrification offered the best combination of preservation of structural integrity of neurospheres, cell viability (>96%), multipotency and karyotype. Rapid cooling in 37% (v/v) EG, 0.6 M sucrose afforded good viability but did not preserve structural integrity. Rapid cooling in 30% (v/v) EG, 0.6 M sucrose additionally reduced cell viability to 77%. Slow-cooling reduced cell viability to 65% and damaged the neurospheres. This study suggests that, in contrast to freezing, vitrification has immense potential for the cryopreservation of stem cells cultured as neurospheres or in other structured cultures. 相似文献

The effect of effective microorganisms (EM) on internal organ morphology, intestinal morphometry, and serum biochemical activity in Japanese quails under Clostridium perfringens challenge was determined. After 30 days of EM addition, one group of quails was orally inoculated with Clostridium perfringens. The second group did not receive EM and was inoculated with C. perfringens. In the gut, EM supplementation reduced the number of lesions, enhanced gut health, and protected the mucosa from pathogenic bacteria. EM showed an anti-inflammatory effect and fewer necrotic lesions in villi. In the internal organs, EM showed a protective effect against a typical lesion of C. perfringens infection. Necrosis and degeneration of the hepatocytes, necrosis of bile ducts, and bile duct proliferation were more severe in the infected group without EM. Morphometric evaluation showed significantly higher villi in the jejunum after EM addition. A greater crypt depth was observed in the C. perfringens group. Biochemical analysis of the blood indicated lower cholesterol on the 12th day of the experiment and between-group differences in total protein, lactate dehydrogenase (LDH), and albumin levels in the EM group. Further studies are needed to improve EM activity against pathologic bacteria as a potential alternative to antibiotics and to develop future natural production systems. 相似文献

Recent studies on the PrkC, serine-threonine kinase show that that the enzyme is located at the inner membrane of endospores
and is responsible for triggering spore germination. The activity of the protein increases considerably after phosphorylation
of four threonine residues placed on the activation loop and one serine placed in the C-terminal lobe of the PrkC. The molecular
relationship between phosphorylation of these residues and enzyme activity is not known. In this work molecular dynamics simulation
is performed on four forms of the protein kinase PrkC from B. subtilis—phosphorylated or unphosphorylated; with or without ATP bound—in order to gain insight into phosphorylation and ATP binding
on the conformational changes and functions of the protein kinase. Our results show how phosphorylation, as well as the presence
of ATP, is important for the activity of the enzyme through its molecular interaction with the catalytic core residues. Three
of four threonine residues were found to be involved in the interactions with conservative motifs important for the enzyme
activity. Two of the threonine residues (T167 and T165) are involved in ionic interactions with an arginine cluster from αC-helix.
The third residue (T163) plays a crucial role, interacting with His-Arg-Asp triad (HRD). Last of the threonine residues (T162),
as well as the serine (S214), were indicated to play a role in the substrate recognition or dimerization of the enzyme. The
presence of ATP in the unphosphorylated model induced conformational instability of the activation loop and Asp-Phe-Gly motif
(DFG). Based on our calculations we put forward a hypothesis suggesting that the ATP binds after phosphorylation of the activation

A series of mono‐, bis‐, and tris(phenoxy)–titanium(IV) chlorides of the type [Cp*Ti(2‐R? PhO)nCl3?n] (n=1–3; Cp*=pentamethylcyclopentadienyl) was prepared, in which R=Me, iPr, tBu, and Ph. The formation of each mono‐, bis‐, and tris(2‐alkyl‐/arylphenoxy) series was authenticated by structural studies on representative examples of the phenyl series including [Cp*Ti(2‐Ph? PhO)Cl2] ( 1 PhCl2 ), [Cp*Ti(2‐Ph? PhO)2Cl] ( 2 PhCl ), and [Cp*Ti(2‐Ph? PhO)3] ( 3 Ph ). The metal‐coordination geometry of each compound is best described as pseudotetrahedral with the Cp* ring and the 2‐Ph? PhO and chloride ligands occupying three leg positions in a piano‐stool geometry. The mean Ti? O distances, observed with an increasing number of 2‐Ph? PhO groups, are 1.784(3), 1.802(4), and 1.799(3) Å for 1 PhCl2 , 2 PhCl , and 3 Ph , respectively. All four alkyl/aryl series with Me, iPr, tBu, and Ph substituents were tested for ethylene homopolymerization after activation with Ph3C+[B(C6F5)4]? and modified methyaluminoxane (7% aluminum in isopar E; mMAO‐7) at 140 °C. The phenyl series showed much higher catalytic activity, which ranged from 43.2 and 65.4 kg (mmol of Ti?h)?1, than the Me, iPr, and tBu series (19.2 and 36.6 kg (mmol of Ti?h)?1). Among the phenyl series, the bis(phenoxide) complex of 2 PhCl showed the highest activity of 65.4 kg (mmol of Ti?h)?1. Therefore, the catalyst precursors of the phenyl series were examined by treating them with a variety of alkylating reagents, such as trimethylaluminum (TMA), triisobutylaluminum (TIBA), and methylaluminoxane (MAO). In all cases, 2 PhCl produced the most catalytically active alkylated species, [Cp*Ti(2‐Ph? PhO)MeCl]. Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are considered the standard of care for type 2 diabetes in many countries worldwide. These molecules have profound anti-hyperglycaemic actions with a favourable safety profile. They are now being considered for their robust cardiovascular (CV) protective qualities in diabetic patients. Most recent CV outcome trials have reported that GLP-1 RAs reduce major adverse cardiovascular events (MACE). Furthermore, the GLP-1 RAs seem to target the atherosclerotic CV disease processes preferentially. GLP-1 RAs also improve a wide range of routinely measured surrogate markers associated with CV risk. However, mediation analysis suggests these modest improvements may contribute indirectly to the overall anti-atherogenic profile of the molecules but fall short in accounting for the significant reduction in MACE. This review explores the body of literature to understand the possible mechanisms that contribute to the CV protective profile of GLP-1 RAs. 相似文献
