Ferrocene-bis(thymine/uracil) conjugates: base pairing directed, spacer dependent self-assembly and supramolecular packing

X-ray crystallographic studies of methylene linked Ferrocene-bis(thymine/uracil) conjugates Fc(T:T)(M) and Fc(U:U)(M) reveal base dependent 2-D supramolecular assemblies generated via wobble self-pairing for bis-thymine and reverse wobble self-pairing for bis-uracil conjugates, differing in architecture from the corresponding butylene spacer linked conjugates.     

Standard molar Gibbs free energy of formation of Pb5CrO8(s), Pb2CrO5(s), and PbCrO4(s)

Standard molar Gibbs free energy of formation of ternary oxides Pb₅CrO₈(s), Pb₂CrO₅(s), and PbCrO₄(s) were determined by measuring equilibrium oxygen partial pressures over relevant phase fields using manometry and solid oxide electrolyte based emf methods and are given by: ${\mathrm{\Delta }}_f{G}_m^{°}{\text{Pb}}_5{\text{CrO}}_8(s)\pm 0.55/(\text{kJ}·{\text{mol}}^{-1})=-1809.4+0.6845(T/\text{K})(837\le T/\text{K}\le 1008)\text{,}$${\mathrm{\Delta }}_f{G}_m^{°}{\text{Pb}}_2{\text{CrO}}_5(\text{s})\pm 0.30/(\text{kJ}·{\text{mol}}^{-1})=-1161.3+0.4059(T/\text{K})(859\le T/\text{K}\le 1021)\text{,}$${\mathrm{\Delta }}_{\text{f}}{G}_m^{°}{\text{PbCrO}}_4(\text{s})\pm 0.17/(\text{kJ}·{\text{mol}}^{-1})=-909.8+0.3111(T/\text{K})(863\le T/\text{K}\le 1093)\text{,}$     

1,3-Dipolar cycloaddition of nitrile oxides to (R)-1-(1-phenylethyl)-3,5-bis[(E)-arylmethylidene]tetrahydro-4(1H)-pyridinones: synthesis and antimycobacterial evaluation of novel enantiomerically pure di- and trispiroheterocycles

Enantiomerically pure (R)-(1-phenylethyl)-3,5-bis[(E)-arylmethylidene]tetrahydro-4(1H)-pyridinones were synthesized for the first time, and their 1,3-dipolar cycloaddition with nitrile oxides affording di- and trispiroheterocycles regio- and stereoselectively in moderate yields was investigated. These compounds were evaluated against Mycobacterium tuberculosis H37Rv (MTB) and multi-drug-resistant M. tuberculosis (MDR-TB). Among the compounds screened, the dispiroheterocycle, namely, (5R,6R,10S)-3,9-bis(4-chlorophenyl)-10-(2,4-dichlorophenyl)-14-[(E)-(2,4-dichlorophenyl)methylidene]-12-[(R)-1-phenylethyl]-1,4,7-trioxa-2,8,12-tri-azadispiro[4.0.4.4]tetradeca-2,8-diene 5m was found to possess the maximum activity with MIC of 0.49 μM against MTB, being 9.6 and 15.6 times more potent than ciprofloxacin and ethambutol, respectively. Against MDR-TB, 5m displayed maximum activity with an MIC of 0.49 μM, with it thus being more active than rifampicin, isoniazid, ciprofloxacin and ethambutol by 7.8, 23, 77 and 124 times, respectively.     

Stereoselective Total Synthesis of Dodoneine

A simple and highly efficient stereoselective total synthesis of dodoneine ( 1 ), a naturally occurring bioactive 5,6‐dihydro‐2H‐pyran‐2‐one, was achieved. The synthesis involved Keck's asymmetric allylation, iodine‐induced electrophilic cyclization, and Grubbs' catalyzed ring‐closing metathesis as key steps.     

Post-assembly Functionalization of Organoplatinum(II) Metallacycles via Copper-free Click Chemistry

We describe the use of a strain-promoted copper-free click reaction in the post-self-assembly functionalization of organoplatinum(II) metallacycles. The coordination-driven self-assembly of a 120° cyclooctyne-tethered dipyridyl donor with 60° and 120° di-Pt(II) acceptors forms molecular rhomboids and hexagons bearing cyclooctynes. These species undergo post-self-assembly [3+2] Huisgen cycloaddition with a variety of azides to give functionalized ensembles under mild conditions.     

Thermal rearrangements of spiro[2.4]hepta-1,4,6-trienes

Thermolysis of spiro[2.4]hepta-1,4,6-triene (1a) at 50 degrees C yielded bicyclo[3.2.0]hepta-1,3,6-triene (5), which dimerized in two different fashions to form cyclobutanes. The 1,2-dimethyl and 1-propyl derivatives of 1a also rearranged at 50 degrees C, but at a faster rate, each yielding a pair of cyclobutane dimers. The structures of these symmetrical dimers were investigated by 1D and 2D NMR and NOE difference spectroscopy. Ab initio calculations indicated that the two strained olefins 1a and 5 had comparable energies about 50 kcal/mol lower than norborna-1(7),2,5-triene, which was thus excluded as a reaction intermediate.     

A novel three-component reaction for the diastereoselective synthesis of 2H-pyrimido[2,1-a]isoquinolines via 1,4-dipolar cycloaddition

[reaction: see text] The 1,4-dipole derived from isoquinoline and DMAD has been shown to react readily with N-tosylimines resulting in the diastereoselective synthesis of 2H-pyrimido[2,1-a]isoquinoline derivatives.     

Formation of a tris-capsule and a tris-carceplex from a cyclic six-bowl assembly

A macrocycle composed of six cavitands was assembled into a tris-capsule and a tris-carceplex, each of which encapsulates three guest molecules.     

Palladium(0)-catalyzed regioselective synthesis of alpha-dehydro-beta-amino esters from amines and allyl acetates: synthesis of a alpha-dehydro-beta-amino acid derived cyclic peptide as a constrained beta-turn mimic

Acetates derived from the adducts of the Baylis-Hillman reaction can be reacted in a regioselective manner with amines in the presence of palladium(0) catalyst to afford alpha-dehydro-beta-amino esters (2 and 3) in good yields. The regioselectivity of the reaction can be controlled by temperature and reaction medium leading to the synthesis of regioisomers 2 or 3. The alpha-dehydro-beta-amino acid 3 is a turn inducer, and the dipeptides 6 derived from it show the presence of an eight-membered intramolecular hydrogen bond. Also, cobalt(II) chloride catalyzes the cleavage of epoxy peptides with alpha-dehydro-beta-amino acid derivative 3b to afford the corresponding dipeptide derivatives 8, which exhibit an intramolecular hydrogen bond and thus mimic a beta-turn. This intramolecular hydrogen bonding preorganizes the corresponding diallylated peptide 8c for cyclization via ring-closing metathesis to afford the cyclic peptide 9 as a constrained mimic of a beta-turn.
