A fungal isolate, identified as Penicillium citrinum S2, produced ≈1 U/mL of PHB depolymerase by 72 h when grown in BHM containing 0.2%, w/v PHB, pH 6.0 at 30 °C. Partial purification of an extracellular poly(-β-)hydroxybutyrate (PHB) depolymerase PhaZ Pen from P. citrinum S2 by two steps using ammonium sulphate (80% saturation) and affinity chromatography using concanavalin A yielded 16.18-fold purity and 21.53% recovery of protein. The enzyme was composed of three polypeptide chains of 66, 43 and 20 kDa, respectively, as determined by sodium dodecyl sulphate-polyacrylamide gel electrophoresis. All the three bands stained positive for glycoprotein by PAS staining. Optimum enzyme activity was detected at pH 6.0 and 50 °C. The enzyme was stable between pH 4.0 and 7.0 at 50 °C, 2 h. β-hydroxybutyrate monomer was detected as the major end product of PHB hydrolysis. The enzyme also showed distinct behaviour towards different inhibitors tested, which suggests the role of serine, serine residue, carboxyl group, tyrosine and sulfhydryl groups in its active site.  相似文献

The paper reports the detailed analysis of photoconductivity and photo-detecting properties of vacuum deposited zinc selenide (ZnSe) thin films. The vacuum deposited ZnSe films were found to have high absorption coefficient and showed peak photo-response at 460 nm. The photocurrent and photo-response time of the films were measured as a function of substrate temperature and annealing conditions. Considerable increase in photocurrent and much faster photo-response was observed in films deposited at high substrate temperatures. Annealing at moderate temperatures also improved the photoconductivity and response time of the films.  相似文献

The ternary blends of acrylate rubber (ACM), poly(ethyleneterephalate) (PET), and liquid crystalline polymer (LCP) were prepared by varying the amount of LCP, but fixing the ratio of ACM and PET using melt mixing procedure. The compatibility behavior of these blends was investigated with infrared spectroscopy (IR), differential scanning calorimetry (DSC), and dynamic mechanical analyzer (DMA). The IR results revealed the significant interaction between the blend components. Glass transition temperature (Tg) and melting temperature (Tm) of the blends were affected depending on the LCP weight percent in the ACM/PET, respectively. This further suggests the strong interfacial interactions between the blend components. In the presence of ACM, the nucleating effect of LCP was more pronounced for the PET. The thermogravimetric (TGA) study shows the improved thermal stability of the blends.  相似文献

The thermoelectromotive force of vacuum deposited Sb2Te3 films on glass substrates has been measured by the integral method in the temperature range 30–250 °C. The film thickness varied between 1200–9600 Å. Thick antimony pads evaporated onto the sample at both ends served as contacts. The thermoelectric power (α) is found to be positive and increases with temperature for thinner samples and at higher temperatures a steady value is achieved. Further increase in the temperature decreases the thermoelectric power. When the film thickness is increased, α decreases. This is explained as due to the reduction in intercrystalline barrier at elevated temperatures and at higher film thicknesses.  相似文献
