Prospect of Anterior Gradient 2 homodimer inhibition via repurposing FDA-approved drugs using structure-based virtual screening

Anterior Gradient 2 (AGR2) has recently been reported as a tumor biomarker in various cancers, i.e., breast, prostate and lung cancer. Predominantly, AGR2 exists as a homodimer via a dimerization domain (E60-K64); after it is self-dimerized, it helps FGF2 and VEGF to homo-dimerize and promotes the angiogenesis and the invasion of vascular endothelial cells and fibroblasts. Up till now, no small molecule has been discovered to inhibit the AGR2–AGR2 homodimer. Therefore, the present study was performed to prepare a validated 3D structure of AGR2 by homology modeling and discover a small molecule by screening the FDA-approved drugs library on AGR2 homodimer as a target protein. Thirteen different homology models of AGR2 were generated based on different templates which were narrowed down to 5 quality models sorted by their overall Z-scores. The top homology model based on PDB ID?=?3PH9 was selected having the best Z-score and was further assessed by Verify-3D, ERRAT and RAMPAGE analysis. Structure-based virtual screening narrowed down the large library of FDA-approved drugs to ten potential AGR2–AGR2 homodimer inhibitors having FRED score lower than ? 7.8 kcal/mol in which the top 5 drugs’ binding stability was counter-validated by molecular dynamic simulation. To sum up, the present study prepared a validated 3D structure of AGR2 and, for the first time reported the discovery of 5 FDA-approved drugs to inhibit AGR2–AGR2 homodimer by using structure-based virtual screening. Moreover, the binding of the top 5 hits with AGR2 was also validated by molecular dynamic simulation.

Alternate technique for simultaneous measurement of photoionization cross-section of isotopes by TOF mass spectrometer

New measurements of photoionization cross-sections of the lithium isotopes are reported employing a Time of Flight (TOF) mass spectrometer in conjunction with an atomic beam apparatus. Using a two-step selective photoionization and saturation technique, we have simultaneously measured the photoionization cross-section of the 2p excited state of both the isotopes Li⁶ and Li⁷ as 15±2.5 Mb and 18 ±2.5 Mb where as the corresponding number densities have been determined as N₀≈5.3×10¹⁰ atoms/cm³ and N₀≈6.2×10¹¹ atoms/cm³ respectively.     

Oscillator strengths of the 4s4p~^{{\mathsf{1,3}}}P1↦ 4snd~^{\mathsf {1,3}}D2 transitions of neutral calcium

A detailed study has been performed for estimating the orbital energies, positions and shifts of the Lyman lines of C⁵⁺, Al¹²⁺ and Ar¹⁷⁺ under strongly coupled plasma with a view to understand such line positions and shifts obtained in laser produced plasma experiments. The effect of strongly coupled plasma has been treated within the Ion Sphere (IS) model. Both non-relativistic and relativistic methods have been used for estimating the spectral properties. Theoretical estimates with IS model of the plasma are in conformity with the results of laser plasma experiments on these highly stripped ions. The experimental data for the systems have also been compared with the theoretical estimates using Debye screening model of the plasma with spatial confinements which gives additional restrictions to the wave functions at finite boundaries.     

A general approach to specific second order ordinary differential equations using homotopy perturbation method

Homotopy perturbation method is used to solve specific second order ordinary differential equations and tested for different examples. The results obtained demonstrate efficiency of the proposed method.

Comparative study of optical parameters of fullerene C60 film at different temperatures

Fullerene C₆₀ thin films on glass substrate (around 2000 ? thickness) were prepared by thermal evaporation technique. The structural, surface morphology and optical properties of the films were studied. The optical properties of fullerene C₆₀ were investigated in the spectral range 200 nm to 900 nm using a UV-Vis spectrophotometer at room temperature as well as at liquid nitrogen temperature (77 K). The optical band gap at room temperature is found to be 2.30 eV, which gradually decreases with lowering the temperature and reaches to 2.27 at 77 K. The thickness and refractive index of fullerene C₆₀ film were calculated by ellipsometry. From the X-ray analysis, we have calculated the grain size, dislocation density, number of crystallite per unit area, and strain of the film at room temperature. The surface morphology of film was analyzed by scanning electron microscope (SEM). The present result show that the fullerene C₆₀ film becomes more conducting at low temperature.
