In this study, a genetically encoded bioluminescent indicator for ERK2 dimer was developed with the split Renilla luciferase complementation method, in which the formation of ERK2 dimer induces a spontaneous emission of bioluminescence in living cells. In response to extracellular stimuli, such as epidermal growth factor (EGF) or 17β‐estradiol (E2), extracellular signal‐regulated kinase 2 (ERK2) is phosphorylated by its upstream kinase MEK, and also phosphorylates its substrates in various regions of the cell, including the nucleus. Phosphorylated ERK2 is led to form its dimer, thereby transporting itself into the nucleus. We demonstrated with the indicator that stimulation with EGF or E2 induces the formation of ERK2 dimer in living MCF‐7 cells. The dynamics of this dimer formation was examined and discussed.  相似文献

In order to emphasize functional thinking in mathematical education, arguments are brought forward for an early and frequent use of tree diagrams in teaching stochastics. Since set and tree diagrams can be regarded as two sides of the same matter, Bayes’ theorem, which is strongly associated with predicative set diagrams, need not be dealt with explicitly any longer. Set diagrams are replaced by tree diagrams that are labelled in detail and by tree inversion as a functional instrument of dealing with conditioned probabilities. This technique is demonstrated by three typical problems in the context of conditioned probability. Finally, two letters by Pascal to Fermat are analysed in order to illustrate that predicative and functional approaches were already pursued in the early stages of probability calculus in the 17th century.  相似文献

Functional spontaneous gradient copolymers of allyl methacrylate (A) and butyl acrylate (B) were synthesized via atom transfer radical polymerization. The copolymerization reactions were carried out in toluene solutions at 100 °C with methyl 2‐bromopropionate as the initiator and copper bromide with N,N,N′,N″,N″‐pentamethyldiethylenetriamine as the catalyst system. Different aspects of the statistical reaction copolymerizations, such as the kinetic behavior, crosslinking density, and gel fraction, were studied. The gel data were compared with Flory's gelation theory, and the sol fractions of the synthesized copolymers were characterized by size exclusion chromatography and nuclear magnetic resonance spectroscopy. The copolymer composition, demonstrating the gradient character of the copolymers, and the microstructure were analyzed. The experimental data agreed well with data calculated with the Mayo–Lewis terminal model and Bernoullian statistics, with monomer reactivity ratios of 2.58 ± 0.37 and 0.51 ± 0.05 for A and B, respectively, an isotacticity parameter for A of 0.24, and a coisotacticity parameter of 0.33. © 2006 Wiley Periodicals, Inc. Reproducibility for successive determinations with a hanging mercury drop electrode is assessed in relation to solution stirring, drop size and back-diffusion to the mercury thread. The effect of experimental parameters such as drop size, deposition time and gallium concentration on the observed stripping current is investigated. The interference of zinc present in a 0.5M NaSCN + 4.2M NaClO(4) supporting electrolyte on the gallium detection limit and calibration plots is described. Formation of an intermetallic compound with a Zn:Ga ratio of 2:3 and its destruction by co-deposition of zinc with Sb are reported. A detection limit of 10(-8)M gallium was obtained in the presence of 10(-5)M Sb(III).  相似文献

Starting from optically active methylferrocene-- and--carboxylic acids (1 a and1 b) of known absolute configuration and enantiomeric purity about 15 chiral ferrocenes (of each isomeric series— and ) were obtained by suitable ligand transformations. Thereby (almost) all possible chiral combinations of the ligands CH3, COOH (COO), COOCH3, CN and NH2 (NH3 +) were accessible which are necessary for a potential test of approximations of chirality functions for compounds with basic symmetry C5v. The chiroptical properties of these disubstituted ferrocenes are recorded.Preliminary tests using a shortened Ansatz revealed large discrepancies between calculated () and found [M]D-values. Possible reasons for this failure are discussed.

Zusammenfassung Um der oft gehörten Meinung entgegenzutreten, die energetischen Wirkungsgrade lebender Organismen seien wesentlich höher als die technischer Verfahren, werden entsprechende Werte miteinander verglichen. Der sehr niedrige Wirkungsgrad der Photosynthese zusammen mit der kleinen Energiedichte der Sonnenstrahlung erzwang ab einer kritischen Größe die Aufspaltung in Energiesammler, die Pflanzen, und Verbraucher chemischer Energie, den Tieren. Dank den 10 mal höheren Wirkungsgraden in der Technik, lassen sich Solarmobile bauen, die von der auf sie fallenden Sonnenstrahlung angetrieben werden. Wegen des Multiplikationsgesetzes der Teilwirkungsgrade ergeben sich sowohl für die Speicherung der Sonnenenergie in den fossilen Brennstoffen wie in der Nahrungskette extrem niedrige Wirkungsgrade.
Efficiencies in the world of life and in engineering
One often hears the opinion, that the efficiencies in the world of life are higher than in engineering. To disprove this, many values of efficiency of corresponding processes are compared. The very low efficiency of photosynthesis in living plants together with the low density of solar radiation has led the world of life to split into collectors of energy, the plants, and consumers of chemical energy, the animals. Thanks to the ten times higher efficiencies of solar cells, it is possible, to produce solar mobiles, vehicles that move driven by solar radiation.
