DNA-encoded combinatorial synthesis provides efficient and dense coverage of chemical space around privileged molecular structures. The indole side chain of tryptophan plays a prominent role in key, or “hot spot”, regions of protein–protein interactions. A DNA-encoded combinatorial peptoid library was designed based on the Ugi four-component reaction by employing tryptophan-mimetic indole side chains to probe the surface of target proteins. Several peptoids were synthesized on a chemically stable hexathymidine adapter oligonucleotide “hexT”, encoded by DNA sequences, and substituted by azide-alkyne cycloaddition to yield a library of 8112 molecules. Selection experiments for the tumor-relevant proteins MDM2 and TEAD4 yielded MDM2 binders and a novel class of TEAD-YAP interaction inhibitors that perturbed the expression of a gene under the control of these Hippo pathway effectors.  相似文献

The behaviour of four biologically relevant selenium compounds (Se(VI), Se(IV), selenomethionine and selenocystine) in capillary zone electrophoresis (CZE) was investigated. Parameters which affect the separation, detection and sample introduction were investigated to improve the sensitivity of the analysis. Formal acceptance of the results of chemical laboratories is increasingly organized through a) accreditation of measuring laboratories nationally and b) mutual recognition of accreditation internationally (through formal Multilateral Recognition Agreements, MRAs). However, real comparability of results of measurements is realized by using common (internationally agreed) measurement scales which make these results traceable to this scale, i.e. "traceable" to the same (internationally agreed) value of the unit of that scale. In addition, the criterion against which the evaluation is done, should be "external" to the measurement laboratories which are being evaluated. This is realized in IRMM's International Measurement Evaluation Programme (IMEP) where evaluation is performed against values which are anchored using "metrology", the science of measurement with its own rules, which offers a sound foundation for measurement in all scientific disciplines. It is argued in this paper that the demonstration of measurement capability against values on such scales provides a result-oriented rather than a procedure-oriented evaluation. Thus, competence can be "demonstrated" rather than just "designated" and this can be shown to both customers and regulators. It inspires more confidence.  相似文献

Summary The conformation in dimethylsulfoxide of the somatostatin derivative angiopeptin and of three disulfide-free analogs was estimated by two-dimensional nuclear magnetic resonance spectroscopy at room temperature. The resulting 3D molecular graphics were compared and shown to reflect the observed differences in the inhibition of restenosis after rat aorta balloon injury by these octapeptide inhibitors. Angiopeptin and its active analog 2 displayed a relatively rigid conformation of the cyclic hexapeptide backbone due to the presence of two well-defined hydrogen bonds, further stabilized by a third hydrogen bond outside the ring. No such constraints were detected for the two biologically inactive analogs, which, compared to 2, had a two-atom longer or shorter hexapeptide ring. The well-defined structure of compound 2 may serve as an improved pharmacophore for this new class of drugs.  相似文献

The simultaneous spectrofluorimetric determination of mixtures of aluminium, gallium and indium as their 8-hydroxyquinoline complexes following extraction into chloroform is studied. The high collinearity of the spectra hindered their resolution by multiple linear regression (MLR) methodology; therefore, experimental data were processed by partial least-squares regression (PLSR) methodology. A previous step in the study of three-dimensional fluorescence spectrum is possible to select the best information to quantify this system with high collinearity. Finally the optimal conditions for quantitation, the best data preprocessing procedure and the most suitable spectral mode for calibration were established. Using an external set allowed the three analytes to be determined simultaineously at concentrations below 1 mug ml(-1) with errors less than 10% for aluminium and indium, and 15% for gallium.  相似文献

The goal of this work was to synthesize new molecular bricks which could be used as precursors of heterobimetallic low-dimensional compounds. Along this line, four compounds have been synthesized and structurally characterized, namely (NBu(4))(2)[Ni(Cl(2)opba)] (1), (NBu(4))(2)[Cu(Cl(2)opba)] (2), (NBu(4))(5)[Mn(Cl(2)opba)(DMSO)(2)](4) (3), and Cu(en)(2)[Mn(Cl(2)opba)(H(2)O)(2)](2).2DMSO (4), with Cl(2)opba = (4,5-dichloro-o-phenylene)bis(oxamato), NBu(4) = tetra-n-butylammonium, en = ethylenediamine, and DMSO = dimethyl sulfoxide. Compounds 1 and 2 are isostructural; they crystallize in the monoclinic system, space group C2/c, Z = 4, with a = 18.708(2) ?, b = 17.525(2) ?, c = 14.763(9) ?, and beta = 92.03(4) degrees for 1 and a = 18.928(2) ?, b = 17.634(2) ?, c = 14.704(9) ?, and beta = 92.38(3) degrees for 2. 3 crystallizes in the tetragonal system, space group P&fourmacr;2(1)c, Z = 2, with a = 26.295(10) ? and c = 12.342(7) ?. The structure shows a random occupation of the metal site by Mn(III) and Mn(II) ions in 3/4 and 1/4 ratios, respectively. 4 crystallizes in the triclinic system, space group P&onemacr;, Z = 1, with a = 7.066(7) ?, b = 11.844(1) ?, c = 14.292(5) ?, alpha = 105.64(2) degrees, beta = 97.67(5) degrees, and gamma = 102.13(3) degrees. The structure consists of Mn(III)Cu(II)Mn(III) trinuclear species, with Cu-O-Mn bridges involving oxygen atoms of the oxamato groups already linked to the metal atom. The magnetic properties of compounds 1-4 have been investigated and quantitatively interpreted. For 3, this magnetic investigation has been performed on a single crystal, which allows us to determine unambiguously the sign of the axial zero-field splitting parameter for the Mn(III) ion. The potentialities of these new molecular bricks have been discussed.  相似文献
