Single molecule fluorescence measurements have recently been used to probe the orientation of fluorescent lipid analogs doped into lipid films at trace levels. Using defocused polarized total internal reflection fluorescence microscopy (PTIRF-M), these studies have shown that fluorophore orientation responds to changes in membrane surface pressure and composition, providing a molecular level marker of membrane structure. Here we extend those studies by characterizing the single molecule orientations of six related BODIPY probes doped into monolayers of DPPC. Langmuir-Blodgett monolayers transferred at various surface pressures are used to compare the response from fluorescent lipid analogs in which the location of the BODIPY probe is varied along the length of the acyl chain. For each BODIPY probe location along the chain, comparisons are made between analogs containing phosphocholine and smaller fatty acid headgroups. Together these studies show a general propensity of the BODIPY analogs to insert into membranes with the BODIPY probe aligned along the acyl chains or looped back to interact with the headgroups. For all BODIPY probes studied, a bimodal orientation distribution is observed which is sensitive to surface pressure, with the population of BODIPY probes aligned along the acyl chains increasing with elevated surface pressure. Trends in the single molecule orientations for the six analogs reveal a configuration where optimal placement of the BODIPY probe within the acyl chain maximizes its sensitivity to the surrounding membrane structure. These results are discussed in terms of balancing the effects of headgroup association with acyl chain length in designing the optimal placement of the BODIPY probe.  相似文献

This review provides analysts with critical insights of current approaches for quantification by laser ablation-inductively coupled plasma-mass spectrometry in the field of elemental imaging. This encompasses both calibration strategies that have been used with success in imaging biological samples, as well as those with potential to improve analytical accuracy and precision if applied to imaging. Methods reviewed include the use of CRMs, laboratory prepared matrix matched standards, internal standardisation, online standard addition and a variety of novel approaches that makes elemental imaging accessible to a wider base of analysts. [(Me(2)Si{NAr}(2))-κ(2)N,N']Sn, reacts with PtCl(2)(L(2)) and [PtCl(μ-Cl)(L)](2) to afford products containing Pt-Sn bonds. In the absence of supporting ligands L, coordination of the stannylene and rearrangement to a structurally unique PtSn(2)N(2)Si metallacycle occurs. We report a combined experimental and computational comparative study of the reactions of the homologous titanium dialkyl- and diphenylhydrazido and imido compounds Cp*Ti{MeC(N(i)Pr)(2)}(NNR(2)) (R = Me (1) or Ph (2)) and Cp*Ti{MeC(N(i)Pr)(2)}(NTol) (3) with silanes, halosilanes, alkyl halides and [Et(3)NH][BPh(4)]. Compound 1 underwent reversible Si-H 1,2-addition to Ti=N(α) with RSiH(3) (experimental ΔH ca. -17 kcal mol(-1)), and irreversible addition with PhSiH(2)X (X = Cl, Br). DFT found that the reaction products and certain intermediates were stabilised by β-NMe(2) coordination to titanium. The Ti-D bond in Cp*Ti{MeC(N(i)Pr)(2)}(D){N(NMe(2))SiD(2)Ph} underwent σ-bond metathesis with BuSiH(3) and H(2). Compound 1 reacted with RR'SiCl(2) at N(α) to transfer both Cl atoms to Ti; 2 underwent a similar reaction. Compound 3 did not react with RSiH(3) or alkyl halides but formed unstable Ti=N(α) 1,2-addition or N(α) protonation products with PhSiH(2)X or [Et(3)NH][BPh(4)]. Compound 1 underwent exclusive alkylation at N(β) with RCH(2)X (R = H, Me or Ph; X = Br or I) whereas protonation using [Et(3)NH][BPh(4)] occurred at N(α). DFT studies found that in all cases electrophile addition to N(α) (with or without NMe(2) chelation) was thermodynamically favoured compared to addition to N(β).  相似文献

Objective

Chemotherapy commonly causes liver injury through sinusoidal obstructive syndrome and steatosis. Chemotherapy-induced liver injury may make it more difficult to detect metastases secondary to reduced contrast between the injured liver and metastases. The aim of this study was to determine the sensitivity of superparamagnetic iron oxide (SPIO) contrast-enhanced imaging in patients who have undergone chemotherapy prior to liver surgery.

Methods

Local ethics committee approval was obtained. Thirty-one patients with hepatic metastases completing preoperative chemotherapy were prospectively recruited. Images were reviewed independently by two blinded observers who identified and localized lesions with a four-point confidence scale. The alternative free-response receiver operator characteristic method was used to analyze the results.

Results

The sensitivity in detecting colorectal metastases following chemotherapy was 78% and 76%, respectively, for observers 1 and 2 (95% confidence interval: 71%–85% and 68%–82%). The areas under the alternative free-response receiver operator curves were 0.73 and 0.80 for observers 1 and 2, respectively.

Conclusion

Compared to previously published work on chemotherapy-naïve patients, it is clear that the sensitivity of SPIO-enhanced magnetic resonance imaging (MRI) in detecting colorectal metastases following chemotherapy is reduced. It is therefore critical that all imaging — pre-, during and postchemotherapy — is reviewed when reporting liver MRI prior to surgery.  相似文献

The structures of heavy and light water at 300?K were investigated by using a joint approach in which the method of neutron diffraction with oxygen isotope substitution was complemented by path integral molecular dynamics simulations. The diffraction results, which give intra-molecular O-D and O-H bond distances of 0.985(5) and 0.990(5)??, were found to be in best agreement with those obtained by using the flexible anharmonic TTM3-F water model. Both techniques show a difference of ??0.5% between the O-D and O-H intra-molecular bond lengths, and the results support a competing quantum effects model for water in which its structural and dynamical properties are governed by an offset between intra-molecular and inter-molecular quantum contributions. Further consideration of the O-O correlations is needed in order to improve agreement with experiment.  相似文献

Nonlinearly constrained optimization problems can be solved by minimizing a sequence of simpler unconstrained or linearly constrained subproblems. In this paper, we consider the formulation of subproblems in which the objective function is a generalization of the Hestenes-Powell augmented Lagrangian function. The main feature of the generalized function is that it is minimized with respect to both the primal and the dual variables simultaneously. The benefits of this approach include: (i) the ability to control the quality of the dual variables during the solution of the subproblem; (ii) the availability of improved dual estimates on early termination of the subproblem; and (iii) the ability to regularize the subproblem by imposing explicit bounds on the dual variables. We propose two primal-dual variants of conventional primal methods: a primal-dual bound constrained Lagrangian (pdBCL) method and a primal-dual 1 linearly constrained Lagrangian (pd 1LCL) method. Finally, a new sequential quadratic programming (pdSQP) method is proposed that uses the primal-dual augmented Lagrangian as a merit function.  相似文献

Polypyrrole is a material with immensely useful properties suitable for a wide range of electrochemical applications, but its development has been hindered by cumbersome manufacturing processes. Here we show that a simple modification to the standard electrochemical polymerization method produces polypyrrole films of equivalently high conductivity and superior mechanical properties in one-tenth of the polymerization time. Preparing the film as a series of electrodeposited layers with thorough solvent washing between layering was found to produce excellent quality films even when layer deposition was accelerated by high current. The washing step between the sequentially polymerized layers altered the deposition mechanism, eliminating the typical dendritic growth and generating nonporous deposits. Solvent washing was shown to reduce the concentration of oligomeric species in the near-electrode region and hinder the three-dimensional growth mechanism that occurs by deposition of secondary particles from solution. As artificial muscles, the high density sequentially polymerized films produced the highest mechanical work output yet reported for polypyrrole actuators.  相似文献

Methyl ethyl ketone (MEK) and methyl iso-butyl ketone (MIBK) constitute significant proportion of the total VOC emissions from manufacturing and application processes of surface coatings. Biodegradation of MEK and MIBK using an acclimatized mixed culture was evaluated, under aerobic condition. Biodegradation studies were carried out using MEK and MIBK as single substrates and in combination. Mixed-pollutant studies were conducted in MEK-dominated system, MIBK-dominated system, and MEK?CMIBK equi-concentration systems to understand the concentration-dependent interaction of these compounds in a biosystem. Experimental data obtained from single-pollutant system was used to estimate the biokinetic parameters, viz. ?? max, K s , K i , and Y T , for these compounds. Among the several bio-kinetic models tested, Monod inhibition model was best suited for predicting the biodegradation of these two VOCs. Four multiple-substrate models, viz. no-interaction, competitive, un-competitive, and non-competitive were used to study the nature of inhibition for different combinations of these compounds. The biodegradation of MEK and MIBK mixtures was found to be best described by competitive inhibition model. However, the predictions were not very good for systems where MEK concentration was higher than MIBK concentration.  相似文献

Fluorescent nanoparticles have a variety of biomedical applications as diagnostics and traceable drug delivery agents. Highly fluorescent porous silica nanoparticles were synthesized in a water/oil phase by a microemulsion method. What is unique about the resulting porous silica nanoparticles is the combination of a single-step, efficient synthesis and the high stability of its fluorescence emission in the resulting materials. The key of the success of this approach is the choice of a lipid dye that functions as a surrogate surfactant in the preparation. The surfactant dye was incorporated at the interface of the inorganic silica matrix and organic environment (pore template), and thus insures the stability of the dye?Csilica hybrid structure. The resulting fluorescent silica materials have a number of properties that make them attractive for biomedical applications: the availability of various color of the resulting nanoparticle from among a broad spectrum of commercially dyes, the controllablity of pore size (diameters of ~5?nm) and particle size (diameters of ~40?nm) by adjusting template monomer concentration and the water/oil ratio, and the stability and durability of particle fluorescence because of the deep insertion of surfactant??s tail into the silica matrix.  相似文献
