The kinetics of metabolic processes can be assessed, in real time by means of MR hyperpolarized (HP) metabolites. [1-13C]pyruvate, hyperpolarized by means of d-DNP, is, by far, the substrate most widely applied to the investigation of several pathologies characterized by deregulated glycolytic metabolic networks, including cancer. Hyperpolarization of [1-13C]pyruvate by means of the cost effective, fast and easy to handle PHIP-SAH (para-hydrogen induced polarization-side arm hydrogenation) method opens-up a pathway for the application of HP metabolites to a wide range of cancer-related studies. Herein, we report the first application of PHIP-SAH hyperpolarized [1-13C]pyruvate in the investigation of upregulated glycolysis in two murine breast cancer cell lines (168FARN and 4T1). The results obtained using HP pyruvate have been validated with a conventional biochemical assay and are coherent with previously-reported lactate dehydrogenase activity measured in those cells.  相似文献

The incorporation of ceramide in phase-separated monolayers of ternary lipid mixtures has been studied by a combination of atomic force microscopy (AFM), fluorescence, and time-of-flight secondary ion mass spectrometry (ToF-SIMS). Replacement of a fraction of the sphingomyelin by ceramide in DOPC/SM/cholesterol monolayers leads to changes in the SM-cholesterol-rich liquid-ordered domains. AFM shows the formation of heterogeneous domains with small raised islands that are assigned to a ceramide-rich gel phase. ToF-SIMS provides conclusive evidence for the localization of SM and ceramide in ordered domains and shows that ceramide is heterogeneously distributed in small islands throughout the domains. The results indicate the utility of combining AFM and ToF-SIMS for understanding compositions of phase-separated membranes.  相似文献

Quantum dots (QDs) have shown promise as imaging agents in cancer, heart disease, and gene therapy research. This review focuses on the design of QDs, and modification using peptides and proteins for mediated targeting of tissues for fluorescence imaging of tumors in vivo. Recent examples from the literature are used to illustrate the potential of QDs as effective imaging agents. The distribution and ultimate fate of QDs in vivo is considered, and considerations of designs that minimize potential toxicity are presented.  相似文献

The effect of microwave (MW) irradiation and ionic liquids (IL) on the cycloaddition of azomethine ylides to [60]fullerene has been investigated by screening the reaction protocol with regard to the IL medium composition, the applied MW power, and the simultaneous cooling of the system. [60]Fullerene conversion up to 98 % is achieved in 2–10 min, by using a 1:3 mixture of the IL 1‐methyl‐3‐n‐octyl imidazolium tetrafluoroborate ([omim]BF4) and o‐dichlorobenzene, and an applied power as low as 12 W. The mono‐ versus poly‐addition selectivity to [60]fullerene can be tuned as a function of fullerene concentration. The reaction scope includes aliphatic, aromatic, and fluorous‐tagged (FT) derivatives. MW irradiation of IL‐structured bucky gels is instrumental for the functionalization of single‐walled carbon nanotubes (SWNTs), yielding group coverages of up to one functional group per 60 carbon atoms of the SWNT network. An improved performance is obtained in low viscosity bucky gels, in the order [bmim]BF4> [omim]BF4> [hvim]TF2N (bmim=1‐methyl‐3‐n‐butyl imidazolium; hvim=1‐vinyl‐3‐n‐hexadecyl imidazolium). With this protocol, the introduction of fluorous‐tagged pyrrolidine moieties onto the SWNT surface (1/108 functional coverage) yields novel FT‐CNS (carbon nanostructures) with high affinity for fluorinated phases.  相似文献

Silver(I) complexes with sulfur‐donor ligands have a broad range of pharmacological applications. One of the most important factors for tuning the biological activity is the type of donor atom and the ease of ligand replacement. Silver thiosaccharinates display a wide range of structures from mono‐ to polynuclear complexes. We report the synthesis, crystal structure and vibrational spectroscopic analysis of a two‐dimensional AgI–thiosaccharinate coordination polymer, namely poly[tris(μ2‐4,4′‐bipyridine‐κ2N:N′)bis(μ3‐1,1‐dioxo‐1,2‐benzisothiazole‐3‐thiolato‐κ3N:S3:S3)bis(μ2‐1,1‐dioxo‐1,2‐benzisothiazole‐3‐thiolato‐κ2S3:S3)tetrasilver(I)], [Ag2(C7H4NO2S2)2(C10H8N2)1.5]n, with 4,4′‐bipyridine acting as a spacer. A relevant feature of the structure is the presence of an unusually short Ag…Ag separation of 2.8859 (10) Å, well within the range of argentophilic interactions and confirmed as such by Raman analysis of the low‐frequency spectrum. From a topological point of view, the structure presents interpenetration in the form of a threefold entangled 2D→2D mesh (2D is two‐dimensional).  相似文献

Topical anti-inflammatory and analgesic effect for the treatment of rheumatoid arthritis is of major interest because of their fewer side effects compared to oral therapy. The purpose of this study was to prepare different types of topical formulations (ointments and gels) containing synthetic and natural anti-inflammatory agents with different excipients (e.g.,: surfactants, gel-forming) for the treatment of rheumatoid arthritis. The combination of Non-Steroidal Anti-Inflammatory Drugs (NSAIDs), diclofenac sodium, a topical analgesic agent methyl salicylate, and a lyophilized extract of Calendula officinalis with antioxidant effect were used in our formulations. The aim was to select the appropriate excipients and dosage form for the formulation in order to enhance the diffusion of active substances and to certify the antioxidant, analgesic, and anti-inflammatory effects of these formulations. To characterize the physicochemical properties of the formulations, rheological studies, and texture profile analysis were carried out. Membrane diffusion and permeability studies were performed with Franz-diffusion method. The therapeutic properties of the formulations have been proven by an antioxidant assay and a randomized prospective study that was carried out on 115 patients with rheumatoid arthritis. The results showed that the treatment with the gel containing diclofenac sodium, methyl salicylate, and lyophilized Calendula officinalis as active ingredients, 2-propenoic acid homopolymer (Synthalen K) as gel-forming excipient, distilled water, triethanolamine, and glycerol had a beneficial analgesic and local anti-inflammatory effect.  相似文献

A multidisciplinary research was conducted by the University of Salento in collaboration with the Lecce Provincial Museum, in order to study different forms of art widespread in the Salento peninsula (Southern Italy) very valuable from an artistic point of view and important as driving force for the tourism of the area. In this research, the archaeometrical analysis was used to study the first cycle of paintings of the church of Santa Maria delle Cerrate, an italo‐greek monastery located in the country about 15 km north‐east of Lecce, probably built in the 12th century. Microscopic, chromatographic and spectrometric techniques were used: optical microscopy was used to study samples and the relevant stratigraphy, micro‐Raman Spectroscopy to identify pigments and Gas Chromatography with Mass Spectrometric Detection to investigate the techniques masters used to decorate the monastery church. Further information on organic and inorganic materials present in the samples were obtained from Fourier transform infrared analysis in attenuated total reflectance. Materials and techniques were clearly ascertained, and, interestingly, pigments were applied both by fresco and egg‐based tempera. Among the various pigments detected, the identification of both lapis lazuli and lead white opened new perspectives both from the historical and conservative points of view. Metal‐filled composites of a commercial PVC (polyvinyl chloride) powder (mean particle size dp ≈ 100 microns) and a metal powder (mean particle size df about 100 microns for copper, Cu, and about 10 microns for nickel, Ni) prepared by mechanical mixing in a ball mill, subsequent hot‐pressing at 443 K and rapid cooling to 300 K, were characterized by the room‐temperature measurements of electrical conductivity σ, density ρ and microhardness H. The sudden jumps of about 17 orders of magnitude followed by a much slower growth up to the limiting filler fraction ϕ* on the log σ vs. ϕ plots are the evidence for the onset of percolation transitions, at filler volume contents ϕc1 = 0.05 and 0.04 for PVC/Cu and PVC/Ni, respectively. For both systems, the values of H exhibited an initial steep increase up to ϕc2 = 0.07, followed by an apparent plateau extending up to ϕ = 0.18. However, drastic differences in the patterns of composition dependence of H were observed at higher metal loadings, i.e., a continuous increase of H up to the leveling‐off at ϕ* for PVC/Cu, in contrast to a sudden drop of H at ϕ = 0.20 and subsequent slow increase for PVC/Ni. For both composites the apparent density ρ′ of a polymer matrix remained the same as that of the neat PVC in the composition interval ϕ < 0.20, while at ϕ* > 0.20 a precipitous drop of ρ1 was observed due to the formation of polymer‐free voids between filler particles (crowding effect) as ϕ approaches ϕ*. The observed effects were analyzed in terms of a tentative model envisaging cross‐overs from “dilute suspension regime” to “semi‐dilute suspension regime” in the concentration range ϕc1 to ϕc2, and from “semi‐dilute suspension regime” to “concentrated suspension regime” above ϕ = 0.20. Different behavior in this latter regime was explained by intrinsic differences in the structure of conductive infinite clusters between mixtures of particles of about the same size (PVC/Cu) and of widely different sizes (PVC/Ni).  相似文献
