An electromembrane extraction followed by HPLC–UV technique was developed and validated for quantification of leuprolide and triptorelin in rabbit plasma. The influencing parameters on the extraction efficiency were optimized using experimental design methodology. The optimized conditions were found to be; supported liquid membrane: a mixture of 1‐octanol and 2‐ethyl hexanol (1:1) containing 10% v/v di(2‐ethylhexyl) phosphate, applied voltage: 5 V, extraction time: 5 min, pH of the donor phase: 4.5 and pH of the acceptor phase: 1.0. The optimized method was validated for linearity, intraday and interday precision, and accuracy in rabbit plasma. The range of quantification for both peptides was 0.5–1000 ng/mL with regression coefficients higher than 0.994. Relative recoveries of leuprolide and triptorelin were found to be 80.3 and 75.5%, respectively. Limits of quantification and detection for both peptides were found to be 0.5 and 0.15 ng/mL, respectively. The validated method was successfully applied to pharmacokinetic study of the 1‐month depot formulations of each peptide after subcutaneous administration to rabbits. 相似文献

Temperature-sensitive hydrogels based on N-t-butylacrylamide (TBA), acrylamide (AAm), and sodium alginate were prepared by free radical polymerization method. Methylenebisacrylamide (MBA) and amonium persulfate (APS) were applied as water soluble crosslinker and initiator, respectively. The chemical structure of the hydrogels was confirmed by FT-IR spectroscopy and thermogravimetric analysis (TGA) methods. Morphology of the samples was examined by scanning electron microscopy (SEM). By changing the initial TBA/AAm mole ratios, hydrogels with different swelling properties were obtained. The rate parameters were found to be 2.0, 2.4, and 3.5 min for the superabsorbents with AAm/TBA weight ratio of 1.0, 1.3 and 2.0 respectively. The swelling behavior in distilled water and different pH solutions was investigated. A preliminary swelling kinetics and the absorbency under load (AUL) were also studied. At the applied pressure (2.07 kPa), maximum swelling was found to be 17, 19, and 21 (g/g) for the superabsorbent hydrogels with AAm/TBA weight ratios of 1.0, 1.5 and 2.0, respectively. 相似文献

Polystyrene (PS)/organoclay nanocomposites were prepared via free radical suspension polymerization. Two kinds of organoclay were used, labeled KT and KD, modified by trimethyloctadecyl ammonium (TM) and dimethyldioctadecyl ammonium (DM) ions, respectively. Nanocomposites containing various amounts of both of the organoclay nanoparticles (1, 3, and 5 wt%) were prepared. The wide angle X-ray diffraction (WAXD) results revealed intercalation in both of the nanocomposites. The greatest improvement in thermal stability of the nanocomposites was achieved with 5 wt% of organo-MMT for both of the clays. The nanocomposite containing 3 wt% of KT organo-MMT showed the greatest improvement of storage modulus. When the organoclay content exceeded 3 wt%, the storage moduli decreased compared to the nanocomposite filled with 3 wt% of the organoclay. D-spacing calculations using Bragg's law and WAXD data showed that the KT and KD nanoparticles were intercalated within the PS matrix, but with different extents of intercalation. The styrene conversions of the as-polymerized nanocomposite samples were obtained by a gravimetric method. The results showed that conversion decreased with incorporation of organoclay in the reaction recipe. Particle size was also increased by increasing nanoclay content. 相似文献

Novel pinacolophanes tethered by methyl and ethyl chains were synthesised and characterised. The stereochemistry of these small-sized cyclophanes was determined by NMR spectral data as well as by single-crystal X-ray analysis. Intramolecular hydrogen bondings in the crystal structure of the molecules play an important role in holding their configuration, while the intermolecular hydrogen bondings lead to a designed arrangement of the molecules. The stereoselective transformation of pinacolophanes to stilbenophanes in improved yield is also reported. 相似文献
