Military training commonly results in land degradation, but protocols for assessing and predicting long-term environmental impact are lacking. An ability to assess the impact of repeated disturbance and subsequent recovery is needed to balance training requirements against environmental quality. To develop methodology for assessing soil quality, a study evaluating disturbance resulting from tank maneuvers was initiated on Fort Riley Military Installation, Kansas. The objectives were to identify and quantify soil-quality indicators on two soil types exposed to controlled tank traffic. We examined physical, chemical, and biotic indicators after treatments were applied during wet and dry soil conditions. A randomized complete-block design, with three blocks per soil type and three treatments per block, was used. Treatments consisted of disturbance created by a 63-ton M1A1 tank making five passes in a figure-8 pattern during either dry or wet soil conditions. The M1A1 was operated at a speed of approximately 8 km/h. Control plots received no tank traffic. Soil-quality indicators evaluated were soil compaction, soil penetration resistance, rut depth, soil bulk density, soil texture, soil chemical composition, plant biomass, soil microbial diversity, and nematode and earthworm taxa. Soil-quality indicators were sampled within one week after tank disturbance. Preliminary data indicate soil-texture-dependent treatment effects (p  0.05) for bulk density and porosity. Bulk density increased and porosity decreased on trafficked areas, in the silt loam soil, but showed no change in the silty clay loam soil. Disturbance during wet soil conditions raised penetrometer resistance and gravimetric water content more than disturbance during dry soil conditions (p  0.05). A significant difference in disturbance was measured between the outside and inside portion of the same track (p  0.01 and 0.001, respectively). The outside track caused the greatest amount of disturbance, as measured by the height of the disturbed soil ridge above the track bed. Tank disturbance significantly reduced total vegetative biomass (p  0.05) compared with that of un-trafficked areas. Disturbance under wet soil conditions significantly reduced grass biomass (p = 0.040), whereas disturbance under dry soil conditions significantly reduced forb biomass (p = 0.0247) compared to un-trafficked areas. Total earthworm abundance (p = 0.011) was reduced by 82% when disturbance occurred during wet soil conditions regardless of soil type.  相似文献

Genetically engineered spider silk‐like block copolymers were studied to determine the influence of polyalanine domain size on secondary structure. The role of polyalanine block distribution on β‐sheet formation was explored using FT‐IR and WAXS. The number of polyalanine blocks had a direct effect on the formation of crystalline β‐sheets, reflected in the change in crystallinity index as the blocks of polyalanines increased. WAXS analysis confirmed the crystalline nature of the sample with the largest number of polyalanine blocks. This approach provides a platform for further exploration of the role of specific amino acid chemistries in regulating the assembly of β‐sheet secondary structures, leading to options to regulate material properties through manipulation of this key component in spider silks.

In 1899, Albert Sexton, a Philadelphia mechanical engineer, received the John Scott Medal of the Franklin Institute for his invention of the omnimetre. This inexpensive circular slide rule was one of a host of computing devices that became common in the United States around 1900. It is inscribed “NUMERI MUNDUM REGUNT”. In part because of instruments such as the omnimetre, numbers increasingly ruled the practical world of the late 19th and early 20th century. DNA-based aptamers are commonly used recognition elements in biosensors for a range of target molecules. Here, the development of a wavelength-shifting optical module for a DNA-based adenosine-binding aptamer is described. It applies the combination of two photostable cyanine-styryl dyes as covalent modifications. This energy-transfer pair is postsynthetically attached to oligonucleotides via a copper(I)-catalyzed azide–alkyne cycloaddition by two structurally different approaches: 1) as nucleotide modifications at the 2′-position of uridines and 2) as nucleotide substitutions using (S)-amino-1,2-propanediol as acyclic linker between the phosphodiester bridges. Both dyes exhibit a remarkable photostability. A library of DNA aptamers consisting of different combinations of the two dyes in diagonal orientations were evaluated by their emission color contrast as readout. Further optimization led to aptasensors with improved fluorescent readout as compared with previously reported aptasensors. This approach described is synthetically facile using simple propargylated phosphoramidites as DNA building blocks. As such, this approach could be applied for other dyes and other chemical/biological applications.  相似文献

In molecular imaging, multimodal imaging agents can provide complementary information, for improving the accuracy of disease diagnosis or enhancing patient management. In particular, optical/nuclear imaging may find important preclinical and clinical applications. To simplify the preparation of dual‐labeled imaging agents, we prepared versatile monomolecular multimodal imaging probe (MOMIP) platforms containing both a fluorescent dye (BODIPY) and a metal chelator (polyazamacrocycle). One of the MOMIP was conjugated to a cyclopeptide (i.e., octreotide) and radiolabeled with 111In. In vitro and in vivo studies of the resulting bioconjugate were conducted, highlighting the potential of these BODIPY‐based bimodal probes. This work also confirmed that the biovector and/or the bimodal probes must be chosen carefully, due to the impact of the MOMIP on the overall properties of the resulting imaging agent.  相似文献

A series of conformationally restrained epothilone analogues with a short bridge between the methyl groups at C6 and C8 was designed to mimic the binding pose assigned to our recently reported EpoA-microtubule binding model. A versatile synthetic route to these bridged epothilone analogues has been successfully devised and implemented. Biological evaluation of the compounds against A2780 human ovarian cancer and PC3 prostate cancer cell lines suggested that the introduction of a bridge between C6-C8 reduced potency by 25-1000 fold in comparison with natural epothilone D. Tubulin assembly measurements indicate these bridged epothilone analogues to be mildly active, but without significant microtubule stabilization capacity. Molecular mechanics and DFT energy evaluations suggest the mild activity of the bridged epo-analogues may be due to internal conformational strain.  相似文献

The weathering of a specimen kaolinite clay was studied over the course of 369 d via solid-state 29Si magic angle spinning (MAS) nuclear magnetic resonance (NMR) spectroscopy and high-field 27Al MAS NMR. The chosen baseline solution conditions (0.05 mol kg-1 of Al, 2 mol kg-1 of Na+, 1 mol kg-1 of NO3-, 1 mol kg-1 of OH-, and pH approximately 13.8) approximate those of solutions leaking from waste tanks at the Hanford Site in Richland, WA. Nonradioactive Cs and Sr cations were added to this synthetic tank waste leachate (STWL) solution at concentrations of 10(-3), 10(-4), and 10(-5) molal (m) to represent their radionuclide counterparts. The transformations of silicon- and aluminum-containing solid phase species were monitored quantitatively by using NMR spectroscopy, with the resulting spectra directly reporting the influence of the initial Cs and Sr on formation and transformation of the neo-formed solids. At the lowest concentration of Cs and Sr employed (10(-5) m in each cation) peaks consistent with the formation of zeolite-like minerals were detected via 29Si and 27Al MAS NMR as early as 33 d. At concentrations of 10(-3) m in each cation, new silicon species are not detected until 93 d, although neophases containing four-coordinate aluminum were detectable at earlier reaction times via 27Al MAS NMR. At the highest magnetic field strengths employed in this NMR study, deconvolutions of resonances detected in the tetrahedral region of the 27Al MAS spectra yielded multiple components, indicating the existence of at least four new aluminum-containing phases. Two of these phases are identified as sodalite and cancrinite through comparison with diffuse-reflectance infrared (DRIFT) spectra and powder X-ray diffraction (XRD) results, while a third phase may correlate with a previously detected aluminum-rich chabazite phase. All measurable solid reaction products have been quantified via their 27Al MAS resonances acquired at high magnetic field strengths (17.6 T), and the quantitative nature of the 27Al NMR data shows that cancrinite growth increases while sodalite reaches a steady state with respect to total aluminum in the solid phases. The data also relate the coupling of Cs sorption to the ripening of feldspathoid phases in this heterogeneous system as a function of time, and illustrate the important influence of co-contaminants on the environmental reaction kinetics studied here.  相似文献

We describe methods for the rapid generation of minilibraries of substituted alkoxy benzenes (consisting of 4-5 compounds), for screening as insect olfaction or gustation inhibitors. Synthetic or commercially available monoalkoxy benzene compounds were mixed and reacted with various alkyl halides to afford a first set of minilibraries. A second and third set were generated from allyloxy minilibraries via the Claisen rearrangement and subsequent alkylation of the ortho-allyl phenols. We have chosen to prepare a collection of small libraries (as opposed to one large library) to test the response insects exhibit toward blends of compounds. We demonstrate how our minilibraries can be screened, both against insect antennae and against expressed pheromone-binding proteins from the gypsy moth, Lymantria dispar.  相似文献
