[reaction: see text] A practical synthesis of the potent class I alpha-mannosidase inhibitor kifunensine (1) beginning from the inexpensive and readily available starting material L-ascorbic acid (15) is described. The protected amino-alcohol ((2R,3R,4R,5R)-5-amino-2,3:4,6-diisopropylidenedioxyhexanol, 11) served as a key intermediate from which several N-1 substituted kifunensine analogues (including N-methyl, N-cyclohexyl, and N-bis(hydroxymethyl)methyl) and 2-desoxakifunensine analogues (including N-H and N-methyl) were prepared and screened for inhibition of human endoplasmic reticulum alpha-mannosidase I (ER Man I) and mouse Golgi alpha-mannosidase IA (Golgi Man IA). In addition, several pseudodisaccharide kifunensine analogues in which a mannose residue was tethered to N-1 of kifunensine via a two-, three-, or four-carbon linker and an affinity-bound kifunensine analogue were also prepared and evaluated for biological activity. While the synthesized N-1 kifunesine analogues were found to be less potent inhibitors of Class I alpha-mannosidases than kifuensine itself, the bis(hydroxymethyl)methylkifunensine analogue 6 was shown to selectively inhibit ER Man I over Golgi Man IA.  相似文献

A reliable computational method for the prediction of organoselenium geometries and bond dissociation energies (BDEs) has been determined on the basis of the performance of density functional theory (DFT: B3LYP and B3PW91) and ab initio molecular orbital procedures (Hartree-Fock (HF)) in conjunction with various Pople basis sets including (but not limited to) the 6-31G(d), 6-31G(d,p), 6-311G(d), 6-311G(d,p), 6-311G(2df,p), and 6-311G(3df,3pd) sets. Predicted geometries and BDEs are compared with available experimental data and quadratic configuration interaction including single and double substitutions (QCISD) results. The B3PW91/6-311G(2df,p) level of theory is recommended for the prediction of the geometries and energetics of organoselenium compounds.  相似文献

X‐ray exposure during crystallographic data collection can result in unintended redox changes in proteins containing functionally important redox centers. In order to directly monitor X‐ray‐derived redox changes in trapped oxidative half‐reaction intermediates of Paracoccus denitrificans methylamine dehydrogenase, a commercially available single‐crystal UV/Vis microspectrophotometer was installed on‐line at the BioCARS beamline 14‐BM‐C at the Advanced Photon Source, Argonne, USA. Monitoring the redox state of the intermediates during X‐ray exposure permitted the creation of a general multi‐crystal data collection strategy to generate true structures of each redox intermediate.  相似文献

A new experiment named MTV-G, probing a large electron spin-precession due to a possible strong gravitational field, which predicted by large extra dimension model, is started at TRIUMF from 2011. In an electron-nuclear scattering experiment, a strong gravitational field is tested as a large spin precession effect caused by geodetic precession predicted by general relativity theory as a result of a warped space-time around nuclei. Experimental design using spin polarized electron source and Mott-spin analyzer, commissioning experiment and the preliminary results are described.  相似文献

We report an NMR experimental realization of a rapid quantum deletion algorithm that deletes marked states in an unsorted database.Unlike classical deletion,where search and deletion are equivalent,quantum deletion can be implemented with only a single query,which achieves exponential speed-up compared to the optimal classical analog.In the experimental realization,the GRAPE algorithm was used to obtain an optimized NMR pulse sequence,and the efficient method of maximum-likelihood has been used to reconstruct the experimental output state.  相似文献

Reproducible proton chemical shifts in the porphyrin series are obtained when the spectrum is measured in chloroform using the zinc (II) complex of the porphyrin in the presence of an excess of pyrrolidine. This method is specifically demonstrated for the case of 2,4-dicyanodeuteroporphyrin-IX dimethyl ester which, as the free base, shows dramatic effects due to aggregation phenomena. The shifts obtained using the zinc (II) complex plus pyrrolidine method, which allow compilation of substituent chemical shifts in the porphyrin series, are shown to be the same as the less easily accessible infinite dilution shifts of the porphyrin free bases.  相似文献

Irradiation of terephthalophenone in isopropyl alcohol gave a bright yellow-colored intermediate which decayed rather rapidly. The products were complex and depended upon the mode of irradiation. They ranged from a mixed polymer I to a dimer II. I consisted of about 5 terephthalophenone units capped with hydrogen or isopropyl alcohol radicals or terminated by cyclization. The effects of copolymerizing small amounts of an ionic moiety (0–7 mol %) into the backbone of a polycondensation polymer have been investigated by a combination of linear viscoelastic, steady-shear, and extensional testing. At a constant viscosity in dilute solution, both the zero-shear-rate viscosity and the maximum relaxation time in the melt increase monotonically with an increase in the ionic character. These effects are present after the data are normalized to remove the increase in the glass transition temperature Tg with increased ionic content. Large-strain, steady-shear flow experiments showed that the viscoelastic memory of the melt is affected by ionic content. An increase in ion content causes a decrease in the effect of strain on the memory function. This effect is not apparent in extensional flow experiments, which indicate that the ionic content does not affect the response if the tests are conducted at a constant temperature above Tg.  相似文献
