The preparation of highly water‐soluble and strongly fluorescent diketopyrrolopyrrole (DPP) dyes using an unusual taurine‐like sulfonated linker has been achieved. Exchanging a phenyl for a thienyl substituent shifts the emission wavelength to near λ=600 nm. The free carboxylic acid group present in these new derivatives was readily activated and the dyes were subsequently covalently linked to a model protein (bovine serum albumin; BSA). The bioconjugates were characterized by electronic absorption, fluorescence spectroscopy and MALDI‐TOF mass spectrometry, thus enabling precise determination of the labeling density (ratio DPP/BSA about 3 to 8). Outstanding values of fluorescence quantum yield (30 % to 59 %) for these bioconjugates are obtained. The photostability of these DPP dyes is considerably greater than that of fluorescein under the same irradiation conditions. Remarkably low detection limits between 80 and 300 molecules/μm2 were found for the BSA bioconjugates by fluorescence imaging with a epifluorescence microscope.  相似文献

Mitochondria consume oxygen in the respiratory chain and convert redox energy into ATP. As a side process, they produce reactive oxygen species (ROS), whose physiological activities are still not understood. However, current analytical methods cannot be used to monitor mitochondrial ROS quantitatively and unambiguously. We have developed electrochemical biosensors based on peroxidase‐redox polymer‐modified electrodes, providing selective detection of H2O2 with nanomolar sensitivity, linear response over five concentration decades, and fast response time. The release of H2O2 by mitochondria was then monitored under phosphorylating or inhibited respiration conditions. We report the detection of two concomitant regimes of H2O2 release: large fluxes (hundreds of nM ) under complex III inhibition, and bursts of a few nM immediately following mitochondria activation. These unprecedented bursts of H2O2 are assigned to the role of mitochondria as the hub of redox signaling in cells.  相似文献

Polyethylene (PE), alkoxyamine‐ and thiol‐terminated PEs (PE‐TEMPO and PE‐SH, respectively) can be converted to macroradicals using a peroxide, a thermal cleavage of the alkoxyamine and a hydrogen transfer reaction of the thiol, respectively. The addition of these macroradicals to multiwalled carbon nanotubes (MWCNTs) were compared by performing grafting reactions at 160 °C in 1,3‐dichlorobenzene as solvent. Raman spectroscopy was utilized to follow the introduction of PE on the MWCNTs' surface while thermogravimetric and elemental analysis indicated the extent of this grafting. The grafting ratio was found to be in the range of 19–36 wt %. PE‐functionalized MWCNTs were imaged by transmission electronic microscopy showing a PE layer with various thicknesses covering the surface of nanotubes. It was found that higher levels of grafting were obtained using PE‐2,2,6,6‐tetramethylpiperidinyl‐1‐oxy and PE‐SH rather than a radical grafting reaction in which dicumyl peroxide, PE, and MWCNTs were reacted. © 2010 Wiley Periodicals, Inc.
