The solid-state molecular conformations and crystal structures of three analogues of the CP-96,345 molecule, an important nonpeptidic SP antagonist, namely the (±)-2-(3-phenylbenzilidene)-3-(2-benzylamino) quinuclidine, theo-chloro- and theo-methoxy-derivatives, have been determined by X-ray diffusion analyses and refined to finalR values of 0.055, 0.045, and 0.056, respectively. All three molecules in the solid state show the same disposition of the substituents of the double bond and differences in the conformation mainly caused by the need of releasing intramolecular strains and/or nonbonded interactions. The observed molecular structures are compared to the reported solid-state structure of the CP-96,345 and correlated to the biological activity as NK antagonists.  相似文献

The synthesis of the novel tetracyclic rings 5 and 6 is described starting from 4-cyano-1-methyl-5(1H-pyrrol-1-yl)-1H-pyrazole and its 3-(1H-pyrrol-1-yl)- isomer, respectively. The pathway employed is based on a one-pot double annelation reaction involving the transformation of 13 into the required tetracyclic derivative 16 . Lithium aluminum hydride/sulfuric acid reduction of the last compound furnished 5 . A similar way was used for the synthesis of 6 by cyclization of 20 .  相似文献

Singlet oxygen, 1O2, can be generated by molecules that upon photoexcitation enable the 3O21O2 transition. We used a series of atomically precise Au24M(SR)18 clusters, with different R groups and doping metal atoms M. Upon nanosecond photoexcitation of the cluster, 1O2 was efficiently generated. Detection was carried out by time-resolved electron paramagnetic resonance (TREPR) spectroscopy. The resulting TREPR transient yielded the 1O2 lifetime as a function of the nature of the cluster. We found that: these clusters indeed generate 1O2 by forming a triplet state; a more positive oxidation potential of the molecular cluster corresponds to a longer 1O2 lifetime; proper design of the cluster yields results analogous to those of a well-known reference photosensitizer, although more effectively. Comprehensive kinetic analysis provided important insights into the mechanism and driving-force dependence of the quenching of 1O2 by gold nanoclusters. Understanding on a molecular basis why these molecules may perform so well in 1O2 photosensitization is instrumental to controlling their performance.

Atomically precise Au24M(SR)18 clusters were used as singlet-oxygen photosensitizers. Comprehensive kinetic analysis provided insights into the mechanism and driving-force dependence of the quenching of 1O2 by gold nanoclusters.  相似文献

This report describes the synthesis of derivatives of two nitrogen tetracyclic ring systems, respectively 9H,11H-pyrimido[4,3-c]pyrrolo[1,2-a][1,4]benzodiazepine and spiro[piperidine-4,4′-[4H]pyrrolo[1,2-a][1,4]-benzodiazepine], by the use of the diethyl ester of 5,6-dihydro-4H-pyrrolo[1,2-a][1,4]benzodiazepine-4,4-diacetic acid as a synthon. This compound was obtained by condensation of 1-(2-aminomethylphenyl)-1H-pyrrole with diethyl 1,3-acetonedicarboxylate in acid medium. Pyrimidopyrrolobenzodiazepine derivatives were obtained by treating either the pyrrolobenzodiazepine 4,4-diacetate or the related 4-methyl-4-acetate with phenylisocyanate in boiling diethyl ether in the presence of sodium metal. The structure of 12,13-dihydro-11,13-dioxo-12-phenyl-9H,11H-pyrimido[4,3-c]pyrrolo[1,2-a][1,4]benzodiazepine, a product formed by loss of an acetate unit when 5,6-dihydro-4H-pyrrolo[1,2-a][1,4]benzodiazepine-4,4-diacetate, sodium metal and phenyl-isocyanate reacted in boiling xylene, was proved by catalytic reduction to 11,13-dioxo-12-phenyl-12,13,14,14a-tetrahydro-9H,11H-pyrimido[4,3-c]pyrrolo[1,2-a][1,4]benzodiazepine, which was synthesized by unambiguous pathway via 5,6-dihydro-4H-pyrrolo[1,2-a][1,4]benzodiazepine-4-acetate. The 2,6-dioxospiro[piperidine-4,4′-[4H]pyrrolo[1,2-a][1,4]benzodiazepine] derivatives were synthesized from the N-BOC derivative of 5,6-dihydro-4H-pyrrolo[1,2-a][1,4]benzodiazepine-4,4-diacetic acid diethyl ester, by hydrolysis followed by treatment with 2 equivalents of 1,1′-carbonyldiimidazole (CDI) and then with aniline or benzylamine. Removal of BOC from the N-phenyl-2,6-dioxopiperidine derivative was obtained by heating the related spiroderivative in toluene in the presence of p-toluenesulphonic acid. Similar reaction failed when the N-benzyl-2,6-dioxopiperidine analog was used as substrate.  相似文献

Several lipophilic perylene and coronene derivatives, employed mainly as liquid crystalline dyes are known and their synthesis has been widely studied. We have applied an analogue strategy using hydrophilic substituents to obtain highly water soluble perylene diimides (4) and a new hydrosoluble coronene derivative (CORON, 6), whose molecular features appear particularly suitable for inducing G-quadruplex DNA structures and inhibiting human telomerase.  相似文献

LiI in refluxing pyridine can remove within a few hours methoxy-, allyloxy-, and benzyloxycarbonyl groups from saccharidic carbinols under conditions compatible with the maintenance of acyl groups. Addition of a stoichiometric excess of acetic acid to the reaction mixture minimizes or even suppresses intramolecular transesterifications. The mechanism and stereochemistry of the intracomplex solvolysis of proton-bound complexes [Y...H...M]+ between M = CH3 (18)OH and Y = 1-arylethanol [(S)-1-(para-tolyl)ethanol (1S), (S)-1-(para-chlorophenyl)ethanol (2S), (S)-1-(meta-alpha,alpha,alpha-trifluoromethylphenyl)ethanol (3S), (S)-1-(para-alpha,alpha,alpha-trifluoromethylphenyl)ethanol (4S), (R)-1-(pentafluorophenyl)ethanol (5R), (R)-alpha-(trifluoromethyl)benzyl alcohol (6R), and (R)-1-phenylethanol (7R)] have been investigated in the gas phase (CH3F; 720 Torr) in the 25-140 degrees C temperature range. Gas-phase solvolysis of [Y...H...M]+ (Y=2S, 3S, 4S, and 7R) leads to extensive racemization above a characteristic temperature t(#) (e.g. at t(#)>60 degrees C for 7R), whereas below that temperature the reaction displays a preferential retention of configuration. Predominant retention of configuration is instead observed in the intracomplex solvolysis of [Y...H...M]+ (Y=1S, 4S, 5R, and 6R) with the temperature range investigated (25 相似文献

The preparation and properties of a series of gold nanoclusters protected by thiolated peptides based on the alpha-aminoisobutyric acid (Aib) unit are described. The peptides were devised to form 0-3 C=O...H-N intramolecular hydrogen bonds, as required by their 3(10)-helical structure. The monolayer-protected clusters (MPCs) were prepared, using a modified version of the two-phase Brust-Schiffrin preparation, and fully characterized with (1)H NMR spectrometry, IR and UV-vis absorption spectroscopies, transmission electron microscopy (TEM), thermogravimetric analysis (TGA), and X-ray photoelectron spectroscopy (XPS). The MPCs were obtained with core diameters in the range of 1.1-2.3 nm, depending on the reaction conditions. Structured peptides formed smaller clusters. The smallest MPC obtained is in agreement with the average formula Au(38)Pep(18). The results showed that the chemical integrity of the peptide is maintained upon monolayer formation and that the average number of peptide ligands per gold cluster is typically 75-85% the value calculated for alkanethiolate MPCs of similar sizes. The IR and NMR spectra indicated that in the monolayer the peptides are involved in both intra- and interligand C=O...H-N hydrogen bonds.  相似文献

Docking simulation and three-dimensional quantitative structure-activity relationships (3D-QSARs) analyses were conducted on four series of HDAC inhibitors. The studies were performed using the GRID/GOLPE combination using structure-based alignment. Twelve 3-D QSAR models were derived and discussed. Compared to previous studies on similar inhibitors, the present 3-D QSAR investigation proved to be of higher statistical value, displaying for the best global model r2, q2, and cross-validated SDEP values of 0.94, 0.83, and 0.41, respectively. A comparison of the 3-D QSAR maps with the structural features of the binding site showed good correlation. The results of 3D-QSAR and docking studies validated each other and provided insight into the structural requirements for anti-HDAC activity. To our knowledge this is the first 3-D QSAR application on a broad molecular diversity training set of HDACIs.  相似文献
