We construct traversable wormholes in dilatonic Einstein-Gauss-Bonnet theory in four spacetime dimensions, without needing any form of exotic matter. We determine their domain of existence, and show that these wormholes satisfy a generalized Smarr relation. We demonstrate linear stability with respect to radial perturbations for a subset of these wormholes.  相似文献

The mesomorphic behavior of a calamitic mesogen (4'-undecyloxybiphenyl-4-yl-4-octyloxy-2-(pent-4-en-1-yloxy)benzoate) and of a supermesogenic octapode formed by the side-on attachment of the mesogen to a octasilsesquioxane central core is studied by X-ray diffraction and polarizing optical microscopy. The calamitic compound is found to have a nematic phase that has biaxial domains (cybotactic clusters) of tilted layers throughout its entire temperature range. Domains of analogous structure are also found in both the nematic and the hexagonal columnar mesophases exhibited by the obctapode compound. The spacing of the layers forming the domains is found to have the same, essentially temperature independent value for the calamitic monomer and for the octapode, in both its mesophases. Comparison with compounds of analogous structure shows that this value is determined by the length of the rigid part of the mesogenic unit. Variation of the latter length is shown to have no effect on the size of the hexagonal lattice of the octapode columnar phase or on the stacking distance within the columns. The presence of the biaxial domains in the nematic phase is discussed in connection with the phase biaxiality that has been observed in structurally related tetrapode compounds and the possibility of field induced macroscopic biaxial nematic order.  相似文献

Ultra performance LC (UPLC) was evaluated as an efficient screening approach to facilitate method development for drug candidates. Three stationary phases were screened: C-18, phenyl, and Shield RP 18 with column dimensions of 150 mm x 2.1 mm, 1.7 microm, which should theoretically generate 35,000 plates or 175% of the typical column plate count of a conventional 250 mm x 4.6 mm, 5 microm particle column. Thirteen different active pharmaceutical ingredients (APIs) were screened using this column set with a standardized mobile-phase gradient. The UPLC method selectivity results were compared to those obtained for these compounds via methods developed through laborious trial and error screening experiments using numerous conventional HPLC mobile and stationary phases. Peak capacity was compared for columns packed with 5 microm particles and columns packed with 1.7 microm particles. The impurities screened by UPLC were confirmed by LC/MS. The results demonstrate that simple, high efficiency UPLC gradients are a feasible and productive alternative to more conventional multiparametric chromatographic screening approaches for many compounds in the early stages of drug development.  相似文献

Plasma etching has evolved in an important technology for rapid and cost-effective generation of random or quasi-ordered nanostructures in large areas and in a repeatable manner, if properly controlled. It simultaneously affects the chemical composition of etched surfaces. Thus, plasma etching finds numerous applications in the areas of biomaterials and biomicrosystems, since surface chemistry and topography are proven to influence strongly cell–substrate interactions. Herein, we briefly review published studies addressing cell–surface interactions, especially those focusing on optimal surface properties favoring cell adhesion and proliferation. We show that plasma-based micro- and nano-texturing of polymeric surfaces provides a unique, simple and yet versatile tool for tuning the physicochemical properties of polymeric surfaces to those favoring cell cultures. Plasma etching and nanotexturing is proven indispensable also for the patterning on the same substrate of different chemical and/or topographical areas to induce preferential cell adhesion in predefined areas. In this respect, the implementation of surfaces with extreme wettabilities (superhydrophobic/superhydrophilic patterns) is highly valued and when integrated inside microchannels can add new potential to the current archery of microanalytical devices. The paper concludes with the authors’ view to the future outlook of the niche area of plasma nanotextured polymer surfaces.  相似文献

Hemoglobin-based oxygen carriers (HBOCs) are blood substitutes based on hemoglobin of either bovine or human origin and they can potentially be misused in elite sports to improve endurance performance. Recently, three methods have been proposed in doping control analysis to allow HBOCs screening and identification by application of electrophoresis, size-exclusion chromatography coupled with HPLC and LC coupled with tandem mass spectrometry (LC/MSMS). In view of the Athens 2004 Olympic Games, modifications were introduced in order to increase the specificity of these methods. The sample preparation protocols of the electrophoretic and SEC-HPLC methods were modified with the introduction of sequential ultra filtration steps to remove all heme containing material below 100 kDa, thus leaving only HBOCs material for analysis. Furthermore, a modification of the LC/MSMS methodology was introduced to allow full scan MS-MS spectra of peptide segments arising from the tryptic digestion of bovine HBOCs. These relatively simple methodological modifications have major impact, as far as time and cost effectiveness is concerned in doping control procedures, because they provide a useful tool in order to identify which suspect samples from the initial visual screening are due to hemolysis and exclude them from further analysis.  相似文献

A simple gas chromatographic method with mass spectrometry detection was developed and validated for the determination of lacosamide in human plasma. Lacosamide and the internal standard, levetiracetam‐d6, were extracted from 200 μL plasma, by a solid‐phase extraction through HF Bond Elut C18 columns, and derivatized using N‐methyl‐Ntert‐butyldimethylsilyltrifluoroacetamide with 1% tert‐butyldimethylsilylchloride in acetonitrile. The limit of quantification was found to be 0.20 μg/mL and the assay was linear up to 20.0 μg/mL with correlation coefficient ≥0.994. The intra‐ and interday precision values were <4.1% in terms of relative standard deviation (%) and the values of intra‐ and interday accuracy were found to be within –7.2 and 5.3% in terms of relative error (%). Absolute recovery of the method for lacosamide was determined at three concentration levels and ranged from 92.5 to 97.6%. The developed method uses small volumes of plasma and proved to be simple, rapid, and sensitive for the determination of lacosamide in plasma. This method can be used in routine every day analysis of plasma samples obtained from patients who follow respective antiepileptic treatment and for the investigation of clinical and forensic cases where lacosamide is involved.  相似文献

The complexes [Ni(4-Spy)(triphos)]BPh(4) and [Ni(2-Spy)(triphos)]BPh(4) {triphos = PhP(CH(2)CH(2)PPh(2))(2), 4-Spy = 4-pyridinethiolate, 2-Spy = 2-pyridinethiolate} have been prepared and characterized both spectroscopically and using X-ray crystallography. In both complexes the triphos is a tridentate ligand. However, [Ni(4-Spy)(triphos)](+) comprises a 4-coordinate, square-planar nickel with the 4-Spy ligand bound to the nickel through the sulfur while [Ni(2-Spy)(triphos)](+) contains a 5-coordinate, trigonal-bipyramidal nickel with a bidentate 2-Spy ligand bound to the nickel through both sulfur and nitrogen. The kinetics of the reactions of [Ni(4-Spy)(triphos)](+) and [Ni(2-Spy)(triphos)](+) with lutH(+) (lut = 2,6-dimethylpyridine) in MeCN have been studied using stopped-flow spectrophotometry, and the two complexes show very different reactivities. The reaction of [Ni(4-Spy)(triphos)](+) with lutH(+) is complete within the deadtime of the stopped-flow apparatus (2 ms) and corresponds to protonation of the nitrogen. However, upon mixing [Ni(2-Spy)(triphos)](+) and lutH(+) a reaction is observed (on the seconds time scale) to produce an equilibrium mixture. The mechanistic interpretation of the rate law has been aided by the application of MSINDO semiempirical and ADF calculations. The kinetics and calculations are consistent with the reaction between [Ni(2-Spy)(triphos)](+) and lutH(+) involving initial protonation of the sulfur followed by dissociation of the nitrogen and subsequent transfer of the proton from sulfur to nitrogen. The factors affecting the position of protonation and the coupling of the coordination state of the 2-pyridinethiolate ligand to the site of protonation are discussed.  相似文献

Background: Infectious diseases represent a significant global strain on public health security and impact on socio-economic stability all over the world. The increasing resistance to the current antimicrobial treatment has resulted in the crucial need for the discovery and development of novel entities for the infectious treatment with different modes of action that could target both sensitive and resistant strains. Methods: Compounds were synthesized using the classical organic chemistry methods. Prediction of biological activity spectra was carried out using PASS and PASS-based web applications. Pharmacophore modeling in LigandScout software was used for quantitative modeling of the antibacterial activity. Antimicrobial activity was evaluated using the microdilution method. AutoDock 4.2® software was used to elucidate probable bacterial and fungal molecular targets of the studied compounds. Results: All compounds exhibited better antibacterial potency than ampicillin against all bacteria tested. Three compounds were tested against resistant strains MRSA, P. aeruginosa and E. coli and were found to be more potent than MRSA than reference drugs. All compounds demonstrated a higher degree of antifungal activity than the reference drugs bifonazole (6–17-fold) and ketoconazole (13–52-fold). Three of the most active compounds could be considered for further development of the new, more potent antimicrobial agents. Conclusion: Compounds 5b (Z)-3-(3-hydroxyphenyl)-5-((1-methyl-1H-indol-3-yl)methylene)-2-thioxothiazolidin-4-one and 5g (Z)-3-[5-(1H-Indol-3-ylmethylene)-4-oxo-2-thioxo-thiazolidin-3-yl]-benzoic acid as well as 5h (Z)-3-(5-((5-methoxy-1H-indol-3-yl)methylene)-4-oxo-2-thioxothiazolidin-3-yl)benzoic acid can be considered as lead compounds for further development of more potent and safe antibacterial and antifungal agents.  相似文献

Hot flashes are considered the most bothersome complaint during menopause. Although hormone therapy is an effective option to relieve hot flashes, it has been associated with significant side effects. The aim of our study is to suggest a novel combination of different plant extracts with distinct mechanisms of action against hot flashes. We selected the rhizome of Glycyrrhiza glabra L. (Fabaceae), the rhizome of Actaea racemosa L. (Ranunculaceae), the aerial parts of Hypericum perforatum L. (Hypericaceae) to produce extracts rich in bioactive phytochemicals and the seed oil of Oenothera biennis L. (Onagraceae). We investigated their estrogenic and antioxidant potential and their inhibitory effect against prostaglandin D2 receptor 1 (DP1) as a novel mechanistic pathway for vasodilation in hot flashes, alone or in combination. The phytochemical footprint of the extracts was analyzed using HPLC-PDA and UPLC-HRMS. We observed that the tested extracts possess different mechanisms of action. A. racemosa exerts a beneficial activation of the estrogen receptor, H. perforatum possesses the highest antioxidant capacity and the seed oil of O. biennis inhibits the DP1 receptor. The triple combination in the optimal doses pertains to efficacy against all three mechanisms of action, serves as a multitarget plant-based therapy and could serve as a novel strategy for the alleviation of hot flashes in postmenopausal women.  相似文献

Vibrational spectroscopic techniques and especially Raman spectroscopy are gaining ground in substituting the officially established chromatographic methods in the identification of ethanol and other volatile substances in body fluids, such as blood, urine, saliva, semen, and vaginal fluids. Although a couple of different carriers and substrates have been employed for the biochemical analysis of these samples, most of them are suffering from important weaknesses as far as the analysis of volatile compounds is concerned. For this reason, in this study three carriers are proposed, and the respective sample preparation methods are described for the determination of ethanol in human urine samples. More specifically, a droplet of the sample on a highly reflective carrier of gold layer, a commercially available cuvette with a mirror to enhance backscattered radiation sealed with a lid, and a home designed microscope slide with a cavity coated with gold layer and covered with transparent cling film have been evaluated. Among the three proposed carriers, the last one achieved a quick, simple, and inexpensive identification of ethanol, which was used as a case study for the volatile compound, in the biological samples. The limit of detection (LoD) was found to be 1.00 μL/mL, while at the same time evaporation of ethanol was prevented.  相似文献
