Tissue engineering offers the potential of providing vessels that can be used to replace diseased and damaged native blood vessels. The endothelization of a synthetic vascular graft minimizes the failures associated with blood clotting and platelet activation. The aim of this study was to culture vascular-derived endothelial and smooth muscle cells on both untreated and NaOH-treated poly(epsilon-caprolactone) (PCL) films, a biocompatible and bio-resorbable polymer, and to evaluate the behavior of both cell types as a preliminary study for vascular graft development. PCL films were prepared by hot pressing; characterized by DSC, IR, SEM, and scanning force microscopy; and treated with NaOH to increase the surface hydrophilicity before cell culture. Endothelial and smooth muscle cells, isolated from pig cava vein, were characterized by immunofluorescence and confocal microscopy studies of endothelial nitric oxide synthase and alpha-smooth muscle actin. Good adhesion, growth, viability and morphology of both the endothelial and smooth muscle cells on PCL films were obtained, but a light stimulation of mitochondrial activity was observed during short culture times. NaOH treatment improved the adhesion and enhanced the proliferation in both cell types. This verified the possible use of this modified polymer as a support in the preparation of a synthetic vascular graft. The composition of the surface waxes of three apple ( Malus domestica L.) cultivars ("Florina", "Golden B" and "Ozark Gold") has been studied by means of spectroscopic and GC–MS analysis of the class-fractionated mixture of components. Odd n -alkanes, mainly C27 and C29 molecules, are prevalent in the saturated fraction. Small concentrations of alkenes were also found; the C28:1 component is strongly (72%) in excess over the other 1-alkenes. Straight-chain esters (mainly of palmitic acid) of saturated primary alcohols (C18–C30) were also detected; whereas the acyl moiety is made up essentially of an even number of carbons, the alcohol counterpart does not exhibit this characteristic. Aldehydes are present (C20–C30) with the homologue patterns C26–C30 most strongly represented. Straight-chain free secondary alcohols characterize the waxes of "Florina" and "Ozark Gold"; the hydroxy function is located far from the extremity of the carbon framework. Outstanding is the presence of three alcohols with 29 carbon centres. These alcohols are accompanied by free straight-chain primary alcohols, mainly with even-numbered carbon chains in the range C26–C30. Free fatty acids are present; all of have a framework of even-numbered carbon chains mainly in the range C16–C20. C18:1 (oleic acid) is well represented. 相似文献

The reaction of B6 vitamins 1–3 with cyanate, in the presence of equivalent amounts of hydrochloric acid, yields different adducts according to the structure of the starting material. The 1H NMR spectra of rifamycin S in different solvents and at different temperatures strongly suggest the existence of a dominant conformer. The nine dihedral angles of the ansa chain from C-28 to C-19, considered conformationally flexible, were obtained from the vicinal interproton coupling constants by the Karplus equation and the proper alternative for each of them compares well with the corresponding values given by X-ray analysis in solid state.All the possible conformations derivable from the intrinsic alternatives of the NMR method were calculated for the ansa chain between C-28 and C-19 and, by applying geometrical considerations, such as closure of the polygonal path of the ansa and the steric incompatibility between the various atoms of the ansa and of the chromophore, only two of them appeared real. Thus, NMR spectroscopy can be used for studying the conformation of the ansa chain of rifamycins in solution. 相似文献

Abstract Modifications induced by the combined application of pressure (up to 800 MPa) and temperature (up to 80° C) were studied on wet and dry semolina, and on fresh pasta as such and after cooking. Rheological and immunochemical properties of the treated products were investigated, along with parameters indicating the extent of protein denaturation. No modification occurs in semolina treated at less than 45% water, below 60°C/800MPa for 10min. Egg noodles prepared from pressure/heat treated semolina had increased protein digestibility and low recognition by anti-gliadin antibodies either before or after cooking. The same effects were observed in heat/pressure treated pasta, but were lost upon subsequent cooking, although we found decreased immunochemical reactivity after heat/pressure treatment of cooked pasta.
