A novel graphical method for assessing the compromise between conditional peak capacity and separation speed for packed bed columns under gradient conditions has been developed and applied to the separation of peptides. This approach is analogous to and complements the conventional "Poppe plot" used to study plate count in isocratic separations. The use of the new plot can assist the design of appropriate column formats (e.g. particle size and column length) for both dimensions in gradient elution two-dimensional liquid chromatography (2DLC). Particularly for the second dimension of 2DLC, we find that smaller particles provide faster separations even though fast separations based on particles smaller than 2 microm are practically limited by the required miniscule column length. We also find that high temperatures strongly enhance the kinetics of peak capacity production whereas higher pressures help achieve larger absolute peak capacities albeit at the cost of longer analysis time.  相似文献

In the past 50 years. gas chromatography (GC) has played a most important role in the analysis of oil. In this review, the early history is briefly reviewed; next developments in this highly relevant application area since about 1985 are highlighted. The main topic of interest are the introduction and decisive role of capillary GC, the use of selective detection techniques, the versatility of coupled-column techniques and, specifically, the additional power of comprehensive two-dimensional GC.  相似文献

The main factors affecting the mesopore porosity of methacrylate-ester based monolithic columns were investigated. We prepared 40 monolithic capillary columns with porosity controlled by varying the proportions of butyl methacrylate (BMA) and ethylene dimethacrylate (EDMA) monomers and of 1,4-butanediol (BUT) and 1-propanol (PROP) as the porogen solvent in the polymerisation mixtures by thermally initiated in situ polymerisation in fused-silica capillaries. Using mixture design software, we systematically varied the composition of the polymerisation mixtures to find significant factors affecting mesopore formation. Multivariate analysis of the experimental data obtained for the fabricated columns yielded a model for prediction of the mesopore porosity in monolithic beds as a function of the composition of the polymerisation mixture used to prepare polymethacrylate monolithic capillary columns. The mean absolute deviation of predicted porosities is 0.029 for most of the columns, with only eight columns showing deviations exceeding 0.050. The main factor affecting the mesopore porosity proved to be the combination of the concentration of hydrophobic monomer (BMA) and the concentration of the less-polar solvent, 1-propanol, in the porogen mixture. The proportion of mesopores in the monolithic capillary columns increases with increasing concentration of 1-propanol and with decreasing concentration ratios of the cross-linker (EDMA) to monomer (BMA) and of BUT to PROP porogenic solvents.  相似文献

The chromatographic characteristics were determined for a set of microfabricated separation channels structured with cylindrical and diamond-shaped pillars with a characteristic size of 5 microm. Channels with different structures and porosities were etched in a silicon wafer using lithographic techniques. The permeability for flow of the channels was shown to increase strongly with the overall porosity. Diamond-shaped pillars appeared to yield a slightly higher permeability than cylindrical pillars at the same channel porosity. Compared to packed columns, permeabilities were higher by a factor of up to 5. Band dispersion in the channels was measured with an unretained fluorescent probe compound using a fluorescence microscope. A relatively large variation in the observed plate heights between channels was found, which was mainly attributed to the inaccurate geometry of the structure close to the side walls. Reduced plate heights between 0.2 and 1.0 were obtained. The lowest plate heights were found for channels with low porosity. The chromatographic impedances were calculated and compared to the values for the traditional chromatographic systems. For one of the structured microchannels the impedance was found to be more than ten times lower than for a column packed with nonporous spherical particles. With the data collected, predictions are given on the possibilities in terms of efficiency and speed offered by structured microchannels for pressure-driven separations, taking practical constraints into account.  相似文献

New phosphorescent Pt(II) compounds based on dimesitylboron (BMes(2) )-functionalized 2-phenylpyridyl (ppy) N,C-chelate ligands and an acetylacetonato ancillary ligand have been achieved. We have found that BMes(2) substitution at the 4'-position of the phenyl ring can blue-shift the phosphorescent emission energy of the Pt(II) compound by approximately 50?nm, compared to the 5'-BMes(2) substituted analogue, without substantial loss of luminescent quantum efficiencies. The emission color of the 4'-BMes(2) substituted Pt(II) compound, Pt(Bppy)(acac) (1) can be further tuned by the introduction of a substituent group at the 3'-position of the phenyl ring. A methyl substituent red-shifts the emission energy of 1 by approximately 10?nm whereas a fluoro substituent blue-shifts the emission energy by about 6?nm. Using this strategy, three bright blue-green phosphorescent Pt(II) compounds 1, 2 and 3 with emission energy at 481, 492, and 475?nm and Φ(PL) =0.43, 0.26 and 0.25, respectively, have been achieved. In addition, we have examined the impact of BMes(2) substitution on 3,5-dipyridylbenzene (dpb) N,C,N-chelate Pt(II) compounds by synthesizing compound 4, Pt(Bdpb)Cl, which has a BMes(2) group at the 4'-position of the benzene ring. Compound 4 has a phosphorescent emission band at 485?nm and Φ(PL) =0.70. Highly efficient blue-green electroluminescent (EL) devices with a double-layer structure and compounds 1, 3 or 4 as the phosphorescent dopant have been fabricated. At 100?cd?m(-2) luminance, EL devices based on 1, 3 and 4 with an external quantum efficiency of 4.7, 6.5 and 13.4?%, respectively, have been achieved.  相似文献

Gradient-elution liquid chromatography (GELC) is a powerful tool for the characterization of synthetic polymers. However, gradient-elution chromatograms often suffer from breakthrough phenomena. Breakthrough can be averted by using a sample solvent as weak as the mobile phase. However, this approach is only applicable to polymers for which a sufficiently strong solvent exists which is at the same time a weak eluent. Finding such a solvent for a given polymer can be laborious or may even be impossible. Besides, when working with comprehensive two-dimensional LC the effluent of the first dimension is the injection solvent of the second dimension. In this case, it is not possible to avoid breakthrough by adjusting the eluent strength of the second-dimension injection solvent. Therefore, another strategy to avert breakthrough has to be implemented. In this work, we successfully avoided breakthrough in GELC by mixing the mobile phase not before, but after the autosampler. This was demonstrated measuring a blend of poly(methyl methacrylate) standards with different molecular-weights as model mixture with comprehensive two-dimensional GELC × size-exclusion chromatography. The strategy is thought to be applicable to all substances with a sufficiently strong dependence of retention on mobile-phase composition. This typically applies to large molecules (synthetic and natural polymers) and allows efficient refocusing. Unretained and barely retained substances are not refocused and therefore suffer in the proposed setup from peak broadening.  相似文献

There is a significant need to characterize and classify lignins and sulfonated lignins. Lignins have so far received a good deal of attention, whereas this is not true for sulfonated lignins. There is a clear demand for a better understanding of sulfonated lignins on a chemical as well as physical level. Many of the analytical methods have been developed with different goals in mind, for example, detection of sulfonated lignins in pulp‐mill effluents, elucidation of structural changes in lignosulfonates during a pulping process, or identification of properties that may affect a formulation when sulfonated lignins are used as a dispersant. When sulfonated lignins are used in industrial applications, analytical data obtained using different techniques may be necessary to enable prediction of their behavior in the target application. In the present review, a critical discussion of established and promising analytical techniques for the characterization of sulfonated lignins is presented.  相似文献

Vitamin D is an important determinant of bone health at all ages. The plasma concentrations of 25-hydroxy vitamin D (25-OH D) and other metabolites are used as biomarkers for vitamin sufficiency and function. To allow for the simultaneous determination of five vitamin D metabolites, 25-OH D3, 25-OH D2, 24,25-(OH)2 D3, 1,25-(OH)2 D3, and 1,25-(OH)2 D2, in low volumes of human plasma, an assay using ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) was established. Plasma samples were spiked with isotope-labeled internal standards and pretreated using protein precipitation, solid-phase extraction (SPE) and a Diels–Alder derivatization step with 4-phenyl-1,2,4-triazoline-3,5-dione. The SPE recovery rates ranged from 55% to 85%, depending on the vitamin D metabolite; the total sample run time was <5 min. Mass spectrometry was conducted using positive ion electrospray ionization in the multiple reaction monitoring mode on a quadrupole–quadrupole-linear ion trap instrument after pre-column addition of methylamine to increase the ionization efficiency. The intra- and inter-day relative standard deviations were 1.6–4.1% and 3.7–6.8%, respectively. The limit of quantitation for these compounds was determined to be between 10 and 20 pg/mL. The 25-OH D results were compared with values obtained for reference materials (DEQAS). In addition, plasma samples were analyzed with two additional Diasorin antibody assays. All comparisons with conventional methods showed excellent correlations (r 2 = 0.9738) for DEQAS samples, demonstrating the high degree of comparability of the new UHPLC-MS/MS technique to existing methods.  相似文献

Spatial comprehensive two-dimensional chromatography is discussed as a potentially alternative to the conventional column-based approach. In “spatial” separations each analyte ends up in a specific location, rather than being eluted at a specific time. Ultimately, higher peak-capacity-production rates (peak capacity per unit time) may be attained by spatial two- and three-dimensional separations. While low-pressure planar chromatography is well developed, the high-pressure equivalent is still in its infancy. We discuss the requirements for a device for high-pressure spatial two-dimensional chromatography and we describe a possible design. A prototype instrument has been constructed in-house. The preparation of a polymer monolithic separation body and a valve configuration that allows manual sample injection are described. Initial tests of this study included the investigation of the homogeneity of the monolith and the flow profile through the separation body. Furthermore, in order to evaluate the current chromatographic performance of the device, a mixture of dyes was separated in one dimension within 30 s.  相似文献
