The three proteins phosphorylase b, calmodulin and fibrinogen are adsorbed onto thioalkyl derivatives of Sepharose much more strongly than onto gels carrying the same alkyl residue coupled via a carbamate linkage. This enhancement of binding onto alkyl-S-Sepharoses compared with alkyl-N-agaroses is not primarily due to an increase in the extent of conformational changes of the proteins occurring on the gel surface. This can be shown in experiments with the tripeptide Trp-Trp-Trp. The Trp tripeptide is also adsorbed with a much higher affinity to butyl-S-Sepharose than to butyl-N-Sepharose, showing that the primary interaction between the immobilized alkyl residue and the amino acids of the protein is decisive for adsorption. A model stressing the strong influence of an atom or a group of atoms at the base of an immobilized alkyl residue is described as "base-atom recognition".  相似文献

Reproducibility for successive determinations with a hanging mercury drop electrode is assessed in relation to solution stirring, drop size and back-diffusion to the mercury thread. The effect of experimental parameters such as drop size, deposition time and gallium concentration on the observed stripping current is investigated. The interference of zinc present in a 0.5M NaSCN + 4.2M NaClO(4) supporting electrolyte on the gallium detection limit and calibration plots is described. Formation of an intermetallic compound with a Zn:Ga ratio of 2:3 and its destruction by co-deposition of zinc with Sb are reported. A detection limit of 10(-8)M gallium was obtained in the presence of 10(-5)M Sb(III).  相似文献

Five Upper Miocene fossil Hipparion molars were studied for U-distribution by Fission Track. U uptake has reached the whole mass of the teeth and no saturation fronts seem to occur in the interior layers. Mean U-concentration in dentine, cement and enamel was 157, 139 and 78 ppm, respectively. Uenamel/Udentine 0.5 is considerably higher than 0.1 reported in the past for Upper-Quaternary mammoth teeth. These features are disadvantageous for ESR-dating in Hipparion enamel, although a long-lived signal (g=2.0018) is apparent. Leaching phenomena occur in the outer regions of these teeth. Inner enamel folding seems to have hindered a more uniform U-distribution in the teeth.  相似文献

Conclusions Refractive-index fluctuations caused by heating alter the intensity and phase fluctuation spectra arising from the turbulence. The large-scale part of the spectra is weakened, whereas the small-scale part is strengthened, and the spatial-frequency range in which the fluctuations are attenuated becomes narrower as the beam energy increases. Correspondingly, the fluctuation structure functions alter. For values of the arguments in those functions less than the radius of the first Fresnel zone , the thermal nonlinearity causes increases in the structural functions D, with the energy. For , the dependence of D, on energy is nonmonotone, with the fluctuations at first weakened but then strengthened. The reduction in fluctuation variance and the improvement in coherence is less pronounced over long lines than on short ones. The range in which geometrical optics can be used to derive the fluctuations is independent of the nonlinearity and is defined by k2z/2k /4. A differential manifold (d-manifold, for short) can be defined as a pair (M, C), where M is any set and C is a family of real functions on M which is (i) closed with respect to localization and (ii) closed with respect to superposition with smooth Euclidean functions; one also assumes that (iii) M is locally diffeomorphic to Rn. These axioms have a straightforward physical interpretation. Axioms (i) and (ii) formalize certain compatibility conditions which usually are supposed to be assumed tacitly by physicists. Axiom (iii) may be though of as a (nonmetric) version of Einstein's equivalence principle. By dropping axiom (iii), one obtains a more general structure called a differential space (d-space). Every subset of Rn turns out to be a d-space. Nevertheless it is mathematically a workable structure. It might be expected that somewhere in the neighborhood of the Big Bang there is a domain in which space-time is not a d-manifold but still continues to be a d-space. In such a domain we would have a physics without the (usual form of the) equivalence principle. Degenerated four-wave mixing (DFWM) was realised in five commercial semiconductor-doped glasses (Corning CS3–66, CS3–67, CS3–68, CS3–69, CS3–70). The reflectivity obtained for each glass seems very dependent on the energy excess (E=hvE g) and relatively independant on l, the product of the absorption coefficient () by the sample thickness (l). The decay time of the DFWM signal was measured for each glass. The relaxation was found to be different when the photon energy is under or over the gap.  相似文献

We report experimental observations of the phonon focusing pattern in [100] GaAs using low temperature electron beam scanning for phonon generation. The typical dispersive effects for high-frequency phonons expected from the calculations by Tamura have clearly been observed using PbIn tunnel junctions for phonon detection. The quantitative comparison of our experimental results with the frequency dependent calculations by Tamura allowed to determine the dominant phonon frequencies contributing to the detector signal in our different experiments. Above the temperature of the -point the dominant phonon frequencies appear to be shifted considerably to lower values, which could be explained by a heating effect in the liquid-He layer adjacent to the tunnel junction detector. By comparing the observed magnitude of the detector signal with different theoretical treatments of the detector response, we have found satisfactory agreement for a model where the perturbation due to the high-frequency phonons is restricted to the base electrode of the detector reached first by the phonons following their passage through the crystal.  相似文献

In this work we used x-ray structural analysis to investigate the martensitic transformations (MTs)B2R andRB19 in Ti(Ni, Fe) alloys containing 1, 2.5, 4, 5 at. % Fe, and the effect of heat treatment on the MT temperatures and sequence. In the majority of cases, it was found that the MTB2R proceeds through a two-phase region (+R). Heat treatment leads to a narrowing of the temperature interval of the existence of the two-phase region, weakly affects the onset temperatures of the MTsB2R TR andRB19 Ms, and sharply increases the temperature-of the end of the MT Me.
