Various types of glycosaminoglycans (GAGs) including heparins, chondroitin sulfates, dermatan sulfate and hyaluronic acid were studied from their proton nuclear magnetic resonance (1H NMR) spectra using chemometric techniques. Despite the complexity of the 1H NMR signals, data analysis using principal component analysis enabled the different GAG classes to be distinguished and permitted their classification according to their chemical structure. The analysis of the composition of the major disaccharide unit and other relevant chemical structures in the heparin samples was performed using partial least squares regression. 相似文献

Environmental samples from a wide-range of aquatic and soil deposits, mainly of Scandinavian origin, were analysed for 234Th (238U) using low-level gamma-spectrometry. The diversity of the samples, in terms of composition and ages, allowed a detailed evaluation of the analytical problems associated with gamma-ray spectrometry with focus on the reliability of the 234Th peaks for absolute determination of the 234Th activities. The X-ray contributions in the 93 keV peak were compared with the corresponding self-absorption corrected activities of the 63 keV peak. These X-ray contributions were, also, correlated with the 238U, 232Th, 235U, 40K and 137Cs activities of the samples. Despite the difficulties imposed by the self-absorption corrections, the 63 keV peak is still the best option. Large variability in the 93 keV peak interferences, due to X-rays from Th, exists in sediment and soil samples. Only in the case of young ombrotrophic peat samples was it possible to conclude that the 93 keV peak is free from X-ray contributions and can be as good as the 63 keV Monte-Carlo self-absorption corrected peak. X-ray contributions in the samples correlated with the 238U and 232Th activities, only, in closed environmental systems where a secular equilibrium with the daughters of the U/Th series can occur. 相似文献

A method for determination chemical oxygen demand (COD) assisted by use of ultrasound has been successfully evaluated for the first time. The method uses instrumentation simpler and cheaper and, in some instances, safer than that used by previous methods for the same purpose. The new device used for sonication is an all-glass cylindrical sonotrode that can be introduced directly into the reaction mixture. Use of this device enables more efficient interaction between sample and ultrasonic energy. The optimized experimental conditions are high ultrasonic power (55% amplitude, 0.9-second pulses each second), high sulfuric acid concentration (>60%), and a sonication time of 2 min. Under these conditions the method has limitations similar to those of the official COD method with regard to the type of organic compound. It works adequately with easily oxidized organic matter (potassium hydrogen phthalate and dextrose) and other organic compounds difficult to oxidize by conventional methods (e.g. phenol and acetic acid) but the COD values obtained with volatile compounds and difficult organic matter are poor. Chloride is tolerated up to a concentration of 7000 mg L(-1) without any masking agent. Gasification of the sample is recommended to improve results; use of air and argon resulted in no significant differences - bubbling with air during sonication resulted in COD values for certified materials and real wastewater samples statistically identical with the certified COD values and those obtained by the classic (open reflux) method. The use of ultrasound energy for COD determination thus seems to be an interesting and promising alternative to conventional oxidation methods used for the same purpose. 相似文献

The alkaline 1-phenyl-1H-1, 2, 3, 4-tetrazole-5-thiolate salts, M[C6H5N4CS] (M = Li ( 1 ), Na ( 2 ), K ( 3 ), Rb ( 4 ) and Cs ( 5 )) were obtained and characterized by means of mass spectrometry (FAB+) and NMR (1H; 13C) spectroscopy. The structures of Na ( 2 ), K ( 3 ), Rb ( 4 ) and Cs ( 5 ) compounds were determined by X-ray diffraction methods. The ligand shows a rich variety of coordination patterns with the alkaline cations. The formation of a four-membered ring MSCN in the compounds with heavier alkali cations (K, Rb and Cs) is shown. In all the cations the coordination number around it increases with the ionic radius. Many standard and official sample digestion procedures for trace metal determination are carried out in open vessels on hot plates. A new procedure for the determination of trace metals by flame atomic absorption spectrometry or inductively coupled plasmaatomic emission spectrometry in beer samples was developed to be performed in closed reactors assisted by microwaves. The results are compared with the ones obtained by other procedures by means of the analysis of the variance. The differences between the procedures are attributed to residual organic matter. Voltammetry, absorption molecular spectrophotometry and high pressure liquid chromatography with a photodiode array detector are used to study the nature of these residues. Nitrobenzoic acids, phenolic acids and other organic compounds are often present after digestion. The results obtained are related to the precision in metal determination by atomic spectrometry. The need for elaboration of certified reference materials for trace metals in beer is suggested. 相似文献

Spectroscopic and photophysical properties of safranine O (Sf) were investigated in binary water/solvent mixtures. It was found that these properties are strongly solvent-dependent. A blue shift is observed for both the ground-state absorption and the triplet-triplet main absorption band when the solvent polarity augments. At the same time a red shift of the fluorescence emission band takes place. These facts are interpreted in terms of higher dipole moment of the dye molecule in the S(1) state as compared with the S(0) state, while a decrease in the dipole moment of the triplet state T(n) with respect to the triplet state T(1) occurs. The Stokes' shift and the fluorescence lifetime shows a linear correlation with the E(T)(30) parameter, while a non-linear behavior is observed when a correlation with models of a continuous dielectric solvent is attempted. These results suggest the operation of strong specific interactions of Sf with solvent molecules, most likely hydrogen bonding. From fluorescence lifetime and quantum yield determinations, as well as intersystem-crossing quantum yields, the solvent dependence of the photophysical kinetic parameters were obtained. The radiative fluorescence rate constant can be adequately reproduced by calculations based on the UV-Vis absorption and emission spectra, as given by the Strickler-Berg equation. 相似文献

The reactivity of 2-deoxy-2-iodoglycosyl isothiocyanates toward O- and S-nucleophiles gives an easy access to 2-deoxy-2-iodoglycopyranosyl thiocarbamates and dithiocarbamates. Internal nucleophilic displacement of the iodine by the sulfur atom in these compounds allows the preparation of glycopyranoso[1,2-d]-1,3-thiazoles and glycopyranoso[1,2-d]-1,3-thiazolidin-2-one or -2-thione. Reaction with amines or polyamines as N-nucleophiles led directly to 2-aminoglycopyranoso[1,2-d]-1,3-thiazoles without isolation of the intermediate thioureas. Methyl 2-deoxy-2-iodoglycopyranosyl thiocarbamates also allow the synthesis of 2-deoxyglycopyranosyl thiocarbamates or 2-deoxy-2-iodoglycopyranosyl carbamates. 相似文献

The separation of tetracyclines (TCs) using reversed-phase liquid chromatography (LC) is proposed. The use of an amide-based stationary phase prevents the interaction of tetracyclines with the residual silanol groups and thus avoids the appearance of tailed peaks. Detection was based on using an UV spectrophotometer and gradient elution with acetonitrile-oxalic acid as mobile phase permitted good separation of all the peaks. Specificity was demonstrated by the retention characteristics, UV spectra and peak purity index. Linearity, precision, recovery and sensitivity were satisfactory. The procedure was applied to the analysis of tetracycline residues (tetracycline, oxytetracycline (OTC), chlortetracycline (CTC), doxycycline (DC), minocycline (MINO) and methacycline (MTC)) in honey of different types. Extraction involved using a mild acidic solvent containing EDTA to release protein-bound or sugar-bound tetracyclines. For the clean-up step, solid phase extraction using phenyl cartridges was applied. Detection limits in the honey using the proposed procedure are between 15 and 30 ng g(-1), depending on the tetracycline. 相似文献
