藏药胡芦巴种籽油脂类成分的GC-MS分析

采用GC-MS法对青海产的胡芦巴油脂类化学成分进行了分析,并以面积归一法测其质量分数。共鉴定出41种化学成分,其中34种为该植物种籽中首次得到。实验中,首先采用三氟化硼-甲醇体系对胡芦巴油进行甲酯化,使酯化更完全,测定效果更好。     

海南捕鸟蛛毒素-Ⅲ的固相化学合成和氧化复性

应用芴甲氧羰基(Fmoc)固相化学合成了海南捕鸟蛛毒素-Ⅲ,并优化了其最佳氧化复性条件。最佳的氧化复性条件为pH 7.5的重蒸水溶液体系,样品质量浓度为0.1 g/L,还原型谷胱甘肽和氧化型谷胱甘肽的浓度分别为 1.0 mmol/L和0.1 mmol/L、L-Arg浓度为1.0 mol/L。复性产物经质谱测定其相对分子质量为3607.68;与天然毒素等量混合后用高效液相色谱分析得到单一峰;膈神经-膈肌标本生理实验结果表明,合成的毒素具有与天然毒素相同的生物学活性,从而可确定二者在结构与功能上具有一致性。     

敬钊毒素-V剪切体Y1-JZTX-V的化学合成与氧化复性及其钠通道活性鉴定

应用芴甲氧羰基(Fmoc)固相方法化学合成了敬钊毒素-V（JZTX-V）分子N-端酪氨酸残基剪切体(Y1-JZTX-V),并且通过反相高效液相色谱和质谱对不同条件下的氧化复性结果进行监测,从而得到该剪切体的最佳氧化复性条件:0.1 mol/L Tris-HCl缓冲液、pH 7.50、1 mmol/L还原型谷胱甘肽(GSH)、0.1 mmol/L氧化型谷胱甘肽(GSSG)、样品浓度为0.05 mg/L、复性温度为4 ℃。膜片钳电生理实验结果显示敬钊毒素-V剪切体Y1-JZTX-V对大鼠背根神经节(DRG)细胞上表达的河豚毒素不敏感型(TTX-R)与河豚毒素敏感型(TTX-S)钠电流均有抑制作用,其半数抑制浓度(IC50)分别为(160±2.5) nmol/L和(39.6±3.2) nmol/L。与天然的敬钊毒素-V相比,该剪切体对大鼠DRG细胞上的TTX-S钠电流的抑制作用基本一致,但对TTX-R钠电流的抑制作用却大大降低,表明敬钊毒素-V分子N-端的酪氨酸残基是一个与TTX-R钠通道结合活性相关的氨基酸残基。

Dynamic properties of soy globulin adsorbed films at the air-water interface

In this paper we present surface dilatational properties of soy globulins (beta-conglycinin, glycinin, and reduced glycinin with 10 mM of dithiothreitol (DTT)) adsorbed onto the air-water interface, as a function of adsorption time. The experiments were performed at constant temperature (20 degrees C), pH (8.0), and ionic strength (0.05 M). The surface rheological parameters were measured as a function of protein concentration (ranging from 1 to 1x10(-3)% wt/wt). We found that the surface dilatational modulus, E, increases, and the phase angle, phi, decreases with time, theta, which may be associated with protein adsorption. These phenomena have been related to protein adsorption, unfolding, and/or protein-protein interactions (at long-term adsorption) as a function of protein concentration in solution. From a rheological point of view, the surface viscoelastic characteristics of soy globulin films adsorbed at the air-water interface are practically elastic. The main conclusion is that the dilatational properties of the adsorbed films depend on the molecular structure of the protein.     

高效液相色谱-荧光检测法测定罂粟籽和火锅汤料中的罂粟碱

用所建立的高效液相色谱-荧光检测法测定了罂粟籽和火锅汤料中的罂粟碱。采用的色谱柱为RP-C18柱(250 mm×4.6 mm i.d.,5 μm);检测激发波长为285 nm,发射波长为355 nm;流动相为甲醇-0.02 mol/L乙酸铵(体积比为70∶30),流速0.8 mL/min。实验结果表明,罂粟碱的进样量为1×10-4~0.1 μg时其质量浓度与相应峰面积有良好的线性关系,最低检测限(以信噪比大于3计)达到0.02 ng。罂粟籽中罂粟碱的回收率为99.0%～100.8%。方法快速准确,简便灵敏,分离度高,能够满足有关食品中罂粟碱的检测要求。

高效液相色谱-荧光检测法测定血清中的犬尿氨酸

建立了高效液相色谱-荧光检测(HPLC-FLD)测定血清中犬尿氨酸(kynurenine,Kyn)含量的方法。采用Hypersil C8色谱柱(300 mm×6.0 mm,10 μm), 流动相为0.25 mol/L醋酸锌及50 mmol/L醋酸溶液(含3%乙腈),流速为1.5 mL/min, 荧光检测激发波长和发射波长分别为365 nm和480 nm。血清标本经5%(v/v)高氯酸溶液去除蛋白质后取上层清液直接进样, Kyn经流动相等度洗脱分离后,用FLD进行测定。研究结果表明Kyn保留时间约为8.3 min,线性范围为0.098～19.6 μmol/L,最低检出浓度为0.04 μmol/L,回收率为90.8%～96.2%,日内、日间测定的相对标准偏差均小于5%,苯丙氨酸、酪氨酸、色氨酸、5-羟色胺和犬尿喹啉酸等物质对犬尿氨酸的测定均无干扰。建立的方法简便、快速、灵敏、特异,适用于临床和科研应用。     

COHERENT OPTICAL PHONON EXCITATION BY FEMTOSECOND LASER PULSE IN YBa2Cu3O7-δ THIN FILMS

With a correlation of nonequilibrium carriers relaxation and coherent phonons displacive excitation, the coherent optical phonon oscillations in YBa₂Cu₃O_7-δ thin films excited by femtosecond laser pulse are simulated theoretically. It is revealed that as the oxygen concentration decreases, the coherent phonon oscillations become easier to be observed due to the decrease of the local coupling between the carriers and the lattice vibrations in the CuO₂ plane.     

Directed ortho-lithiation of unprotected diphenylphosphinic acids

Directed ortho lithiation of diphenylphosphinic acid and subsequent electrophilic trapping provides mono ortho-functionalized derivatives including enantiopure γ-aminophosphinic acids in moderate yields. Copper catalyzed coupling of the ortho anion leads to biphenyl-2,2′-diylbis(phenylphosphinic acid), a phosphorus analogue of biphenyl-2,2′-dicarboxylic acid. Preliminary studies of the metal-binding abilities of this O,O-chelating ligand towards a series of metal cations are included.
