Arachidonic‐acid‐derived prostaglandins (PGs), specifically PGE2, play a central role in inflammation and numerous immunological reactions. The enzymes of PGE2 biosynthesis are important pharmacological targets for anti‐inflammatory drugs. Besides mammals, certain edible marine algae possess a comprehensive repertoire of bioactive arachidonic‐acid‐derived oxylipins including PGs that may account for food poisoning. Described here is the analysis of PGE2 biosynthesis in the red macroalga Gracilaria vermiculophylla that led to the identification of 15‐hydroperoxy‐PGE2, a novel precursor of PGE2 and 15‐keto‐PGE2. Interestingly, this novel precursor is also produced in human macrophages where it represents a key metabolite in an alternative biosynthetic PGE2 pathway in addition to the well‐established arachidonic acid‐PGG2‐PGH2‐PGE2 route. This alternative pathway of mammalian PGE2 biosynthesis may open novel opportunities to intervene with inflammation‐related diseases. 相似文献

The crystal structures of 1,2,3,4,6‐penta‐O‐acetyl‐α‐d ‐mannopyranose, C16H22O11, and 2,3,4,6‐tetra‐O‐acetyl‐α‐d ‐mannopyranosyl‐(1→2)‐3,4,6‐tri‐O‐acetyl‐α‐d ‐mannopyranosyl‐(1→3)‐1,2,4,6‐tetra‐O‐acetyl‐α‐d ‐mannopyranose, C40H54O27, were determined and compared to those of methyl 2,3,4,6‐tetra‐O‐acetyl‐α‐d ‐mannopyranoside, methyl α‐d ‐mannopyranoside and methyl α‐d ‐mannopyranosyl‐(1→2)‐α‐d ‐mannopyranoside to evaluate the effects of O‐acetylation on bond lengths, bond angles and torsion angles. In general, O‐acetylation exerts little effect on the exo‐ and endocyclic C—C and endocyclic C—O bond lengths, but the exocyclic C—O bonds involved in O‐acetylation are lengthened by ~0.02 Å. The conformation of the O‐acetyl side‐chains is highly conserved, with the carbonyl O atom either eclipsing the H atom attached to a 2°‐alcoholic C atom or bisecting the H—C—H bond angle of a 1°‐alcoholic C atom. Of the two C—O bonds that determine O‐acetyl side‐chain conformation, that involving the alcoholic C atom exhibits greater rotational variability than that involving the carbonyl C atom. These findings are in good agreement with recent solution NMR studies of O‐acetyl side‐chain conformations in saccharides. Experimental evidence was also obtained to confirm density functional theory (DFT) predictions of C—O and O—H bond‐length behavior in a C—O—H fragment involved in hydrogen bonding. 相似文献

A new class of neutral bidentate ligands with pnictogenyl-functional sites has been obtained. The reaction of tmeda⋅(BH2I)2 ( 1 , tmeda=tetramethylethylendiamine) with different phosphanides yields the corresponding bidentate phosphanylboranes tmeda⋅(BH2PH2)2 ( 2 a ), tmeda⋅(BH2PPh2)2 ( 2 b ), and tmeda⋅(BH2tBuPH)2 ( 2 c ). This reaction strategy could be further extended to synthesize the first bidentate arsanylborane tmeda⋅(BH2AsPh2)2 ( 3 ). Depending on the substituents on the phosphorus, these compounds form different AuI complexes, to build either polymeric tmeda⋅(BH2PH2AuCl)2 ( 4 a ), or monomeric tmeda⋅(BH2PPh2AuCl)2 ( 4 b ) products. These compounds form also neutral oligomeric group 13/15 chain-like molecules by coordination to a boron moiety such as tmeda⋅(BH2PH2BH3)2 ( 5 a ) and tmeda⋅(BH2AsPh2BH3)2 ( 5 b ). This paper focuses on the technical aspects of chemical screening from 384-well plate nano-scale single-bead combinatorial libraries. The analytical technique utilized is a combination of capillary liquid chromatography with ultraviolet detection and matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. The HPLC/MALDI-MS hyphenation is achieved by means of a micro-fraction collector with a peak detection system that automatically collects the peaks onto the MALDI targets for subsequent characterization. Several experimental parameters such as type of 384-well plate, well-plate sealing foils, and a column-switching procedure were investigated using a small test library of nine components. Additionally, the influence of different MALDI matrices, different MALDI targets and sample-spotting techniques on the MALDI detection sensitivity as well as the ruggedness and sample throughput capacity of this technique were studied. Optimum results for the analytes investigated were obtained with 2,5-dihydroxybenzoic acid using on-line mixing of HPLC effluent and matrix solution. To demonstrate the potential of this capillary HPLC/MALDI-TOFMS method, its application to several single-bead libraries was investigated. The instrumental method allowed for the rapid identification and purity assessment of combinatorial libraries with detection limits down to the higher femtomole level using both UV detection and MALDI mass spectrometry. 相似文献

Our research involves the development of new cationic materials for anion-based applications. We report the solvothermal synthesis and characterization of Pb(3)F(5)NO(3), a new layered lead fluoride material that, unlike the majority of layered and open-framework materials, is cationic in charge. The structure consists of polyhedral lead centers connected by doubly and triply bridging fluoride groups. We quantitatively exchanged the interlamellar nitrate groups of Pb(3)F(5)NO(3) for dichromate, under ambient aqueous conditions. Nuclear magnetic resonance and UV-vis spectroscopy show the reaction proceeds to 61.0% completion in several days. The material is also stable to 450 degrees C, which is vastly superior to organic resins that are still the standard for anion-exchange. The presence of extraframework anions also opens up other potentially unique anion-based properties, such as new catalytic reactions, anion intercalation, or growth of anionic clusters within the void spaces of the cationic material. 相似文献

Many photoactive metal complexes can act as electron donors or acceptors upon photoexcitation, but hydrogen atom transfer (HAT) reactivity is rare. We discovered that a typical representative of a widely used class of iridium hydride complexes acts as an H-atom donor to unactivated olefins upon irradiation at 470 nm in the presence of tertiary alkyl amines as sacrificial electron and proton sources. The catalytic hydrogenation of simple olefins served as a test ground to establish this new photo-reactivity of iridium hydrides. Substrates that are very difficult to activate by photoinduced electron transfer were readily hydrogenated, and structure–reactivity relationships established with 12 different olefins are in line with typical HAT reactivity, reflecting the relative stabilities of radical intermediates formed by HAT. Radical clock, H/D isotope labeling, and transient absorption experiments provide further mechanistic insight and corroborate the interpretation of the overall reactivity in terms of photo-triggered hydrogen atom transfer (photo-HAT). The catalytically active species is identified as an Ir(ii) hydride with an IrII–H bond dissociation free energy around 44 kcal mol−1, which is formed after reductive 3MLCT excited-state quenching of the corresponding Ir(iii) hydride, i.e. the actual HAT step occurs on the ground-state potential energy surface. The photo-HAT reactivity presented here represents a conceptually novel approach to photocatalysis with metal complexes, which is fundamentally different from the many prior studies relying on photoinduced electron transfer.Upon irradiation with visible light, an iridium hydride complex undergoes hydrogen atom transfer (HAT) to unactivated olefins in presence of a sacrificial electron donor and a proton source. 相似文献

Canonical Monte Carlo simulations were used to study the adsorption and compression of fluid layers on model substrates with cubic, (111) fcc, and graphite geometries. The effect of the relative size of the fluid and substrate molecules on adsorption was considered for strong molecule-surface interactions. In the case of monolayer formation, it was found that the surface geometry and the size of the adsorbate molecules had a significant effect on the structure of the adsorbed layer. These structures varied from well-ordered, commensurate layers to liquid-like structures. Lateral compression was observed for certain fluid to substrate molecule sizes. For the interactions studied in this work, it was found that maximum lateral compression occurred on the cubic surface when adsorbate molecules had a diameter approximately 15% larger than the substrate diameter. In the case of multilayer formation, it was found that second and higher adsorbed layers could compress into the adsorbed layers below them. For cubic substrates, the interlayer compression was predicted analytically with reasonable accuracy, with maximum interlayer compression found for fluid diameters approximately 90% the size of substrate molecule diameters. 相似文献

The validation data were within the required limits.
