Lanthanide-encoded polystyrene particles synthesized by dispersion polymerization are excellent candidates for mass cytometry based immunoassays, however they have previously lacked the ability to conjugate biomolecules to the particle surface. We present here three approaches to post-functionalize these particles, enabling the covalent attachment of proteins. Our first approach used partially hydrolyzed poly(N-vinylpyrrolidone) as a dispersion polymerization stabilizer to synthesize particles with high concentration of -COOH groups on the particle surface. In an alternative strategy to provide -COOH functionality to the lanthanide-encoded particles, we employed seeded emulsion polymerization to graft poly(methacrylic acid) (PMAA) chains onto the surface of these particles. However, these two approaches gave little to no improvement in the extent of bioconjugation. In our third approach, seeded emulsion polymerization was subsequently used as a method to grow a functional polymer shell (in this case, poly(glycidyl methacrylate) (PGMA)) onto the surface of these particles, which proved highly successful. The epoxide-rich PGMA shell permitted extensive surface bioconjugation of NeutrAvidin, as probed by an Lu-labeled biotin reporter (ca. 7 × 10(5) binding events per particle with a very low amount of non-specific binding) and analyzed by mass cytometry. It was shown that coupling agents such as EDC were not needed, such was the reactivity of the particle surface. These particles were stable and the addition of a polymeric shell was shown did not affect the narrow lanthanide ion distribution within the particle interior as analyzed by mass cytometry. Synthetic bacteriochlorins are of interest for fundamental studies in photochemistry because of their strong absorption in the near-infrared spectral region and close similarity with natural bacteriochlorophylls. A de novo route to 5-methoxybacteriochlorins entails self-condensation of a dihydrodipyrrin-acetal, which in turn is prepared from a 2-(2-nitroethyl)pyrrole species and an α,β-unsaturated ketone-acetal (e.g., 1,1-dimethoxy-4-methylpent-3-en-2-one). Here, four new results are reported concerning the synthesis of substituted bacteriochlorins. First, a new, scalable route to 1,1-dimethoxy-4-methylpent-3-en-2-one removes a significant previous impediment to the overall route. Second, the new route was employed to gain access to new α,β-unsaturated ketones and corresponding dihydrodipyrrins bearing alternative substituents in place of the dimethoxy unit. Third, a dihydrodipyrrin bearing a 1,3-dioxolan-2-yl moiety afforded the bacteriochlorin (30% yield) containing a 2-hydroxyethoxy substituent at the 5-position. Fourth, subsequent bromination proceeded regioselectively at the 15-position to give a trans-(5,15)-AB-bacteriochlorin building block. The linear 5,15-substitution pattern is attractive for a number of molecular designs. The results taken together afford deeper understanding of the scope and limitations of the de novo route and also advance the capabilities for tailoring synthetic bacteriochlorins. 相似文献

The linear photophysical, excited state absorption (ESA), superfluorescence, and two-photon absorption (2PA) properties of 4,4'-(1E,1'E)-2,2'-(7,7'(1E,1'E)2,2'(4,4'-sulfonylbis(4,1-phenylene))bis(ethane-2,1-diyl)bis(9,9-didecy-9H-fluorene7,2-diyl))bis(ethane-2,1-diyl)bis(N,N-diphenylaniline) (1) were investigated in organic and aqueous media with respect to its potential application in biological imaging. The analysis of linear photophysical properties revealed a rather complex nature of the main one-photon absorption band, strong solvatochromic effects in the steady-state fluorescence spectra, single-exponential fluorescence decay, and high fluorescence quantum yields in organic solvents (≈1.0). The ESA spectra of 1 suggested potential for light amplification in nonpolar media while efficient superfluorescence in cyclohexane was demonstrated. The degenerate 2PA spectra of 1 were obtained over a broad spectral range (640-900 nm), using a standard two-photon induced fluorescence method under 1 kHz femtosecond excitation. Two well defined 2PA bands with maximum 2PA cross sections up to 1700 GM in the higher energy, short wavelength band and ≈1200 GM in the lower energy, long wavelength band of 1 were shown. The potential use of 1 in bioimaging was demonstrated via one- and two-photon in vitro fluorescence imaging of HCT 116 cells. 相似文献

Relative values of the 1H and 13C isotropic hyperfine couplings in the cationic oxidized tryptophan radical TrpH*+ in aqueous solution are determined. The data are obtained from the photo-CIDNP (chemically induced dynamic nuclear polarization) enhancements observed in the microsecond time-resolved NMR spectra of the diamagnetic products of photochemical reactions in which TrpH*+ is a transient intermediate. The method is validated using the tyrosyl neutral radical Tyr*, whose 1H and 13C hyperfine couplings have previously been determined by electron paramagnetic resonance spectroscopy. Good agreement is found with hyperfine coupling constants for TrpH*+ calculated using density functional theory methods but only if water molecules are explicitly included in the calculation. 相似文献

Methyl radicals react in fast reactions, with rate constants k>1×10(8) M(-1) s(-1), with Au(0), Ag(0) and TiO(2) nanoparticles (NPs) dispersed in aqueous solutions to form intermediates, (NP)-(CH(3))(n), in which the methyl groups are covalently bound to the NPs. These intermediates decompose to form ethane. As n≥2 is required for the formation of C(2)H(6), the minimal lifetime (τ) of the methyls bound to the NPs, (NP)-CH(3), can be estimated from the rate of production of the CH(3)(·) radicals and the NPs concentration. The results obtained in this study, using a very low dose rate γ-source for NP = Ag(0), Au(0), and TiO(2) point out that τ of these intermediates is surprisingly long, for example, ≥8 and ≥188?sec for silver and gold, respectively. These data point out that the NP-C bond dissociation energies are ≥70?kJ mol(-1). Under low rates of production of CH(3)(·), that is, when the rate of formation of ethane is very low, other reactions may occur, consequently the mechanism proposed is "broken". This is observed in the present study only for TiO(2) NPs. These results have to be considered whenever alkyl radicals are formed near surfaces. Furthermore, the results point out that the rate of reaction of methyl radicals with (NP)-(CH(3))(n) depends on n, that is, the number of methyl radicals bound to the NPs affect the properties of the NPs. 相似文献

The UV-absorption, fluorescence excitation and emission spectra of the 6-chloro-, 8-chloro-, 6,8-dichloro-derivatives of nor-harmane, harmane and harmine and the 8-chloro-derivative of harmol were studied. These studies were performed in EtOH and in EtOH+1% perchloric acid solutions (pa). Furthermore, fluorescence quantum yields (phi(f)) in both media and in acetonitrile and acetonitrile + 1% perchloric acid solutions at 298 K were measured. The HOMO and LUMO energy, the positions (lambda(max)) and oscillator strength (f) of the 1S1 <-- 1S0 band for all the neutral and protonated beta-carbolines studied were calculated and compared with the experimental data. The pK(a) values in aqueous solution for for 6-chloro-, 8-chloro- and 6,8-dichloro-nor-harmane, harmane and harmine and 8-chloro-harmol were spectrophotometrically measured (pK(a(H2O)). The change of the acid-base character of these compounds on going from the ground state (pK(a)) to the first electronic excited singlet state (pKa*) as DeltapKa = pKa*-pKa, in ethanol solution at 298 K were calculated (DeltapK(a(EtOH))). Ground-state proton affinity (PA) for all the compounds studied defined as minus the enthalpy change of the reaction M + H(+) --> MH+ (gas state) were calculated. Basicity relative to pyridine (DeltaH(rPy)) defined as the enthalpy change of the isodesmic reaction MH(+) + Py --> M + PyH+ in gas state and in water solution, were also calculated (ab initio calculations). The effect of chlorine as substituent on the photochemistry and acid-base properties of the beta-carboline alkaloids is discussed. 相似文献

The unprecedented selective encapsulation of trans-[Co(en)2Cl2]+ from the mixture of trans and cis isomers into the cavity of macrocyclic cavitand cucurbit[8]uril (C48H48N32O16, CB[8]) leads to the inclusion compound {trans-[Co(en)2Cl2]@CB[8]}Cl.17H2O (1). Single-crystal X-ray analysis, 1H NMR, and ESI-MS spectra confirm the formation of host-guest complex 1 in both solid state and solution. The geometry of the complex cation alters significantly upon inclusion, which causes appreciable hypsochromic shifts of the absorption bands of the guest complex. According to TGA data, inclusion of trans-[Co(en)2Cl2]+ dramatically stabilizes the complex toward thermal decomposition. Encapsulation of trans-[Co(en)2Cl2]+ into the CB[8] cavity also increases the stability of the metal complex toward isomerization into the cis form. The supramolecular adduct {cis-[Co(en)2(H2O)2](CB[8])6}Cl3.ca109.5H2O (2) was isolated only after prolonged heating of an aqueous solution of 1 at 120 degrees C in an evacuated tube; it was characterized by X-ray crystallography, IR, and elemental analysis. 相似文献

Dielectric properties, thermal stability and sub-molecular structure of metal-polymer composites on the basis of polyvinylidene fluoride (PVDF) modified by sub-micro sized nickel powder were studied in this work by different methods such as dielectric spectroscopy, resistivity measurement, breakdown strength measurement, differential scanning calorimetry and optical microscopy. It was found that modification of PVDF by nickel particles results in the increase of both the volume resistivity by a factor of 1.24…15 and the melting and decomposition temperature by 5 °C and 48 °C for composite with an optimal nickel content compared with non-modified PVDF. 相似文献
