Phytoestrogen resveratrol (R) has been demonstrated to benefit human reproductive health. However, R bioavailability and pharmacokinetics are still problematic under oral supplementation. We used an experimental vaginal gel with R and hyaluronic acid (HA) to improve bioavailability and pharmacokinetic properties. The study aimed to assess the impact of vaginal R-HA gel on the reproductive system in ovariectomized rats. Methods: The study was carried out on Wistar female rats. It investigated the body weight, tail temperature, vaginal pH, estrogen and progesterone blood levels, and immunohistochemical biomarkers (COX2, Casp-3, Bcl-2, and VEGF). Animals were divided into control animals; ovariectomized rats (OVX); and OVX group treated with vaginal 0.5% R-HA gel (0.5%, 0.1 mL, daily 28 days). Results: The R-HA gel’s therapeutic effect was manifested by slowing weight gain by 17% (p < 0.001), less pronounced symptom of fever at the root of the tail by 9% (p < 0.001) and lowering the vaginal pH to 4.4–4.5 compared with OVX rats. The anti-inflammatory effect and the reduction of COX-2 expression in vagina were accompanied by antiapoptotic impact of RA-H on endometrium, associated with the decreased Casp-3 expression (p < 0.001) and elevated Bcl-2 score in endometrial glands (p = 0.01). Together with enhanced VEGF expression in endometrial glands (p < 0.001) and stromal cells (p = 0.007), these changes prevented endometrial atrophy (p < 0.001) after ovariectomy. Thus, this study substantiates the feasibility of developing an innovative topical drug with R and HA for treating hypoestrogenic disorders.  相似文献

The attachment of cells onto solid supports is fundamental in the development of advanced biosensors or biochips. In this work, we characterize cortical neuron adhesion, growth, and distribution of an adhesive layer, depending on the molecular structure and composition . Neuronal networks are successfully grown on amino-terminated alkanethiol self-assembled monolayer (SAM) on a gold substrate without adhesion protein interfaces. Neuron adhesion efficiency was studied for amino-terminated, carboxy-terminated, and 1:1 mixed alkanethiol SAMs deposited on gold substrates. Atomic force microscopy and X-ray photoelectron spectroscopy were used to measure the roughness of gold substrate and thickness of SAM monolayers. Conformational ordering and ionic content of SAMs were characterized by vibrational sum frequency generation (VSFG) spectroscopy. Only pure amino-terminated SAMs provide efficient neuronal cell attachment. Ordering of the terminal amino groups does not affect efficiency of neuron adhesion. VSFG analysis shows that ordering of the terminal groups improves with decreasing surface roughness; however the number of gauche defects in alkane chains is independent of surface roughness. We monitor partial dissociation of carboxy groups in mixed SAMs that implies formation of NH3+ neighbors and appearance of catanionic structure. Such catanionic environment proved inefficient for neuron adhesion. Surface roughness of metal within the 0.7-2 nm range has little effect on the efficiency of neuron adhesion. This approach can be used to create new methods that help map structure-property relationships of biohybrid systems.  相似文献

This article presents a combined experimental and computational study of novel isothiochromene 2,2-dioxide derivatives, which were synthesized by the electrophilic substitution reactions. The compounds have been studied by NMR, IR spectroscopy and single-crystal X-ray analysis. According to X-ray diffraction data, the six-membered ring of 6 adopts a distorted half-chair conformation, but in case of compounds 7 and 8, we have a twist-boat conformation of the six-membered ring with deviation of the S1 and C8 atoms from the mean plane of the remaining atoms of the ring, respectively. Results of quantum-chemical calculations of possible isomers and tautomers of the isolated oxime 8 and azo compounds 10 at the B3LYP/aug-cc-pVDZ level indicate that the oxime-oxo tautomer (8a) is more stable than the nitroso-hydroxy tautomer (8b) in agreement with the crystal data and that the keto tautomer (II) is more stable than the hydroxyl tautomer (I) and zwitterion tautomer (III), because of a strong intramolecular hydrogen bond N–H···O.

Virus clearance is required for pharmaceutical preparations derived from animal or human sources such as blood products, vaccines, recombinant proteins produced in mammalian cell lines, etc. High cost and substantial protein losses during virus inactivation are significant problems for protein/peptide manufacturing. The goal of this project was to develop a method to perform virus inactivation in a course of protein chromatographic purification. Another goal was to show that the chromatographic adsorbent can serve as reliable “sieva” for mechanical washing away of infecting viruses. Using chromatographic, photometric, IFA, and RT-PCR approaches, it was discovered that high temperature-depending dynamic capacity of adsorbent allowed to perform a virus inactivation directly in a chromatographic column by solvent/detergent treatment. The peptide/protein biological activity was completely preserved. Using this new approach enveloped and nonenveloped viruses were effectively removed protein preparation. In addition, it was shown that RT-PCR method demonstrates more precise and reproducible results and robust properties for assessment of virus reduction than virus titer followed by infectivity studies. Presented method allowed to obtain the factor of virus concentration decrease (FVD) values that were higher than those provided by known technologies and was sufficient for a full inactivation of viruses. The method is recommended to use in pharmaceutical industry.  相似文献

The temperature dependence of the minority carrier diffusion length and lifetime in bulk n-type ZnO was studied using Electron-Beam-Induced Current (EBIC) and cathodoluminescence (CL) techniques. The diffusion length was observed to increase exponentially over the temperature range from 25 to 125 C, yielding an activation energy of 45 ± 2 meV. A concomitant decrease of the cathodoluminescence intensity for the near-band-edge transition was also observed. The activation energy determined by optical measurements was 58 ± 7 meV. The larger minority carrier diffusion length and smaller luminescence intensity are attributed to the increased lifetime of non-equilibrium holes in the valence band at elevated temperatures. Carrier trapping on Li-related levels with activation energy 283 ± 9 meV is also addressed.  相似文献

Efficient syntheses of trisubstituted cryptophane-A derivatives that are versatile host molecules for many applications are reported. Trihydroxy cryptophane was synthesized in six or seven steps with yields as high as 9.5%. By a different route, trihydroxy cryptophane modified with three propargyl, allyl, or benzyl protecting groups was synthesized with yields of 4.1-5.8% in just six steps. Hyperpolarized (129)Xe NMR chemical shifts of 57-65 ppm were measured for these trisubstituted cryptophanes.  相似文献

Oxidative aromatic coupling of meso-substituted porphyrins bearing one electron-rich naphthalene unit has been studied in detail. After thorough optimization of oxidant, naphthalene-fused porphyrins were prepared in high yield without contamination from chlorinated side-products using Fe(ClO(4))(3)·2H(2)O. Copper and nickel complexes were successfully transformed into π-expanded porphyrins in 40-83% yield.  相似文献
