Tamarindus indica is widely used as a food and beverage and in traditional medicine. The apparent lack of dose standardization in herbal medicine necessitates the evaluation of the lethality T. indica on Artemia salina nauplii and chicken embryos via in vitro and in vivo techniques. Furthermore, hepatotoxicokinetics of the crude extract and fractions on Wister rats was also assessed. At concentrations of 200, 20 and 2 μg/mL, crude extract and fractions showed brine shrimp death percentages ranging from 86.70% to 3.30% and the sub-fractions showed death percentage ranges of 46.70% to 3.30%. Calculated LD?? values ranged from 832 μg/mL to 5,019 μg/mL. Dosing Wister rats with 25% and 50% concentration of LD?? determined for crude extract and fractions on chicken embryos showed an elevation in the ALT and AST levels in the serum. Brine shrimps and chicken embryos showed a positive correlation, with R2 values of 0.541 and 0.588 (P ≤ 0.05) for fractions and subfractions, respectively, as media for the lethality assay. Dose standardization in folk herbal medicine is imperative as T. indica used as food and medicine has been shown to be toxic at high doses. Brine shrimp and chicken embryos may be comparably used as medium for toxicity assay.  相似文献

This study aims to use solid phase microextraction (SPME), a simple tool to investigate diffusion rate (time) constant of selected pharmaceuticals in gel and fish muscle by comparing desorption rate of diffusion of the drugs in both agarose gel prepared with phosphate-buffered saline (PBS; pH 7.4) and fish muscle. The gel concentration (agarose gel model) that could be used to simulate tissue matrix (fish muscle) for free diffusion of drugs under in vitro and in vivo conditions was determined to model mass transfer phenomena between fibre polymer coating and environmental matrix such that partition coefficients and desorption time constant (diffusion coefficient) can be determined. SPME procedure involves preloading the extraction phase (fibre) with the standards from spiked PBS for 1 h via direct extraction. Subsequently, the preloaded fibre is introduced to the sample such fish or agarose gel for specified time ranging from 0.5 to 60 h. Then, fibre is removed at specified time and desorbed in 100 μL of desorption solution (acetonitrile: water 1:1) for 90 min under agitation speed of 1000 rpm. The samples extract were immediately injected to the instrument and analysed using liquid chromatography coupled with tandem mass spectrometry (LC/MS/MS). The limit of detection of the method in gel and fish muscle was 0.01–0.07 ng mL−1 and 0.07–0.34 ng g−1, respectively, while the limit quantification was 0.10–0.20 ng mL−1 in gel samples and 0.40–0.97 ng g−1 in fish sample. The reproducibility of the method was good (5–15% RSD). The results suggest that kinetics of desorption of the compounds in fish tissue and different viscosity of gel can be determined using desorption time constant. In this study, desorption time constant which is directly related to desorption rate (diffusion kinetics) of selected drugs from the fibre to the gel matrix is faster as the viscosity of the gel matrix reduces from 2% (w/v) to 0.8% (w/v). As the concentration of gel reduces, viscosity of the gel will be reduced therefore allowing faster diffusion which invariably affect desorption time constant. Also, desorption time constant of model drugs in the fish muscle and 0.8–0.9% (w/v) gel model are similar based on free diffusion of studied compounds. In addition, in vitro and in vivo desorption time constant comparison shows that desorption time constant in an in vivo system (live fish muscle) is generally higher than an in vitro system (dead fish muscle) except for sertraline and nordiazepam. This study demonstrates SPME as a simple investigative tool to understand kinetics of desorption in an in vivo system with a goal to measure desorption rate of pharmaceuticals in fish.  相似文献

Six fractions, named TiA - TiF, were obtained by fractionating the crude ethanol extract of the stem bark of Tamarindus indica using column chromatographic techniques. On TLC, fraction TiB showed five bands, TiC three bands, while TiD and TiE showed two bands each. TiC, TiD and TiE were re-eluted with different solvent systems to yield two fractions each, while TiB yielded four. These subfractions were designated B1-B4; C1-C2; D1-D2 and E1-E2, respectively. Tannins, flavonoids and alkaloids, among other components, were detected, albeit in different proportions with respect to fractions and subfractions and were compartmentalized with respect to the solvent systems used. The in vitro antibacterial activity of fractions and subfractions was tested separately and in combinations using the agar well diffusion technique. The susceptibly of test strains (expressed as %) were: 83.3% (TiA and TiB), 75.0% (crude extract and TiC), 66.7% (TiD), 50.0% (TiE) and 16.7% (TiF) when used singly, whereas in combination, the corresponding susceptibilities were 100% (CE), 83.3% (DE), 66.7% (AB, AF, BC, BD, DE and EF), 50% (AC and CD), 33.3% (BE and BF) and 16.7% (AD) against Gram negative bacteria strains and 100% (EF), 80% (DE), 60% (AB, BC and CE), 40% (AC, BD, BF, CF and DF) and 20% (AE, AF, BE and CD) against Gram positive strains. Percentage susceptibility with combinatorial use of re-fractions ranged from 85.7-57.1% and 60-40% against Gram negative and positive strains (TiB subfractions), respectively, 100-85.7% and 40-0% against Gram negative and positive strains (TiC, TiD and TiE sub-fractions).  相似文献

Morinda lucida (L.) Benth. (Rubiacae) is used in traditional medicine in many West African countries for the treatment of various human diseases. The leaves and roots of this plant were subjected to hydro-distillation to obtain volatile oils which were analyzed by high resolution GC/MS. Fifty compounds were identified in the leaf volatile oil and the major compounds were alpha-terpinene (17.8%) and beta-bisabolene (16.3%). In the root oil, 18 compounds were identified, the major constituents being 3-fluoro-p-anidine (51.8%) and hexadecanoic acid (12.0%). Antioxidant activities of the oils were examined using the DPPH, ABTS, reducing power and lipid peroxidation assays. All assays were concentration dependent with varying antioxidant potentials. The antioxidant activity of the root volatile oil of M. lucida was similar to that of the standard drugs used.  相似文献

In recent times, there has been a cumulative apprehension regarding organophosphate flame retardants (OPFRs) owing to their high manufacturing and usage after brominated flame retardants were strictly regulated and banned from being distributed and used in many countries. OPFRs are known as the main organic pollutants in the terrestrial and aquatic environment. They are very dangerous to humans, plants and animals. They are also carcinogenic and some have been implicated in neurodevelopmental and fertility challenges. OPFRs are distributed into the environment through a number of processes, including the usage, improper disposal and production of materials. The solid phase extraction (SPE) method is suggested for the extraction of OPFRs from water samples since it provides high quality recoveries ranging from 67% to 105% and relative standard deviations (RSDs) below 20%. In the same vein, microwave-assisted extraction (MAE) is highly advocated for the extraction of OPFRs from sediment/soil. Recoveries in the range of 78% to 105% and RSDs ranging from 3% to 8% have been reported. Hence, it is a faster method of extraction for solid samples and only demands a reduced amount of solvent, unlike other methods. The extract of OPFRs from various matrices is then followed by a clean-up of the extract using a silica gel packed column followed by the quantification of compounds by gas chromatography coupled with a mass spectrometer (GC–MS) or a flame ionization detector (GC-FID). In this paper, different analytical methods for the evaluation of OPFRs in different environmental samples are reviewed. The effects and toxicities of these contaminants on humans and other organisms are also discussed.  相似文献

Abstract

Right S -acts A can also be defined, and a tensor product A ??? s B (a set)can be defined that has the customary universal property with respect to balanced maps from A?×?B into arbitrary sets. Over the past three decades, an extensive theory of flatness properties has been developed (involving free and projective acts, and flat acts of various sorts, defined in terms of when the tensor product functor has certain preservation properties). A recent and complete discussion of this area is contained in the monograph Monoids, Acts and Categories by M. Kilp et al. (New York: Walter de Gruyter, 2000). To date, there have been only a few attempts to generalize this material to ordered monoids acting on partially ordered sets ( S -posets). The present paper is devoted to such a generalization. A unique decomposition theorem for S -posets is given, based on strongly convex, indecomposable S -subposets, and a structure theorem for projective S -posets is given. A criterion for when two elements of the tensor product of S -posets given, which is then applied to investigate several flatness properties.  相似文献

Any class of domains, in particular a class of domains that arises from generalizations of factoriality, invites questions about its stability under the standard operations. One of these generalizations of factoriality is the one that requires that every nonzero element be contained in only finitely many principal prime ideals of height one. We use this property to settle all the open cases in the literature on stability of generalizations of factoriality under the standard ring extensions. The paper provides a compendium on the stability, under ring extensions, of all the known generalizations of factoriality. We also use stability properties of factorization in extensions of valuation domains to give a new characterization of discrete valuation domains.  相似文献

The increasing numbers of cases of antibiotic resistance among pathogenic bacteria such as Vibrio species poses a major problem to the food and aquaculture industries, as most antibiotics are no longer effective in controlling pathogenic bacteria affecting these industries. Therefore, this study was carried out to assess the antibacterial potentials of crude aqueous and n-hexane extracts of the husk of Cocos nucifera against some selected Vibrio species and other bacterial pathogens including those normally implicated in food and wound infections. The crude extracts were screened against forty-five strains of Vibrio pathogens and twenty-five other bacteria isolates made up of ten Gram positive and fifteen Gram negative bacteria. The aqueous extract was active against 17 of the tested bacterial and 37 of the Vibrio isolates; while the n-hexane extract showed antimicrobial activity against 21 of the test bacteria and 38 of the test Vibrio species. The minimum inhibitory concentrations (MICs) of the aqueous and n-hexane extracts against the susceptible bacteria ranged between 0.6-5.0 mg/mL and 0.3-5.0 mg/mL respectively, while the time kill study result for the aqueous extract ranged between 0.12 Log?? and 4.2 Log?? cfu/mL after 8 hours interaction in 1 x MIC and 2 x MIC. For the n-hexane extract, the log reduction ranged between 0.56 Log?? and 6.4 Log?? cfu/mL after 8 hours interaction in 1 x MIC and 2 x MIC. This study revealed the huge potential of C. nucifera extracts as alternative therapies against microbial infections.  相似文献
