Scoparone, which is a major constituent of Artemisia capillaries, has been identified as an anticoagulant, hypolipidemic, vasorelaxant, anti-oxidant and anti-inflammatory drug, and it is used for the traditional treatment of neonatal jaundice. Therefore, we hypothesized that scoparone could suppress the proliferation of VSMCs by interfering with STAT3 signaling. We found that the proliferation of these cells was significantly attenuated by scoparone in a dose-dependent manner. Scoparone markedly reduced the serum-stimulated accumulation of cells in the S phase and concomitantly increased the proportion of cells in the G0/G1 phase, which was consistent with the reduced expression of cyclin D1, phosphorylated Rb and survivin in the VSMCs. Cell adhesion markers, such as MCP-1 and ICAM-1, were significantly reduced by scoparone. Interestingly, this compound attenuated the increase in cyclin D promoter activity by inhibiting the activities of both the WT and active forms of STAT3. Similarly, the expression of a cell proliferation marker induced by PDGF was decreased by scoparone with no change in the phosphorylation of JAK2 or Src. On the basis of the immunofluorescence staining results, STAT3 proteins phosphorylated by PDGF were predominantly localized to the nucleus and were markedly reduced in the scoparone-treated cells. In summary, scoparone blocks the accumulation of STAT3 transported from the cytosol to the nucleus, leading to the suppression of VSMC proliferation through G1 phase arrest and the inhibition of Rb phosphorylation. This activity occurs independent of the form of STAT3 and upstream of kinases, such as Jak and Src, which are correlated with abnormal vascular remodeling due to the presence of an excess of growth factors following vascular injury. An F-box protein, β-TrCP recognizes substrate proteins and destabilizes them through ubiquitin-dependent proteolysis. It regulates the stability of diverse proteins and functions as either a tumor suppressor or an oncogene. Although the regulation by β-TrCP has been widely studied, the regulation of β-TrCP itself is not well understood yet. In this study, we found that the level of β-TrCP1 is downregulated by various protein kinase inhibitors in triple-negative breast cancer (TNBC) cells. A PI3K/mTOR inhibitor PI-103 reduced the level of β-TrCP1 in a wide range of TNBC cells in a proteasome-dependent manner. Concomitantly, the levels of c-Myc and cyclin E were also downregulated by PI-103. PI-103 reduced the phosphorylation of β-TrCP1 prior to its degradation. In addition, knockdown of β-TrCP1 inhibited the proliferation of TNBC cells. We further identified that pharmacological inhibition of mTORC2 was sufficient to reduce the β-TrCP1 and c-Myc levels. These results suggest that mTORC2 regulates the stability of β-TrCP1 in TNBC cells and targeting β-TrCP1 is a potential approach to treat human TNBC. 相似文献
High energy‐density lithium‐ion batteries are in demand for portable electronic devices and electrical vehicles. Since the energy density of the batteries relies heavily on the cathode material used, major research efforts have been made to develop alternative cathode materials with a higher degree of lithium utilization and specific energy density. In particular, layered, Ni‐rich, lithium transition‐metal oxides can deliver higher capacity at lower cost than the conventional LiCoO2. However, for these Ni‐rich compounds there are still several problems associated with their cycle life, thermal stability, and safety. Herein the performance enhancement of Ni‐rich cathode materials through structure tuning or interface engineering is summarized. The underlying mechanisms and remaining challenges will also be discussed. 相似文献
Reducing the noble-metal catalyst content of acid Polymer Electrolyte Membrane (PEM) water electrolyzers without compromising catalytic activity and stability is a goal of fundamental scientific interest and substantial technical importance for cost-effective hydrogen-based energy storage. This study presents nanostructured iridium nanodendrites (Ir-ND) supported on antimony doped tin oxide (ATO) as efficient and stable water splitting catalysts for PEM electrolyzers. The active Ir-ND structures exhibited superior structural and morphological properties, such as particle size and surface area compared to commercial state-of-art Ir catalysts. Supported on tailored corrosion-stable conductive oxides, the Ir-ND catalysts exhibited a more than 2-fold larger kinetic water splitting activity compared with supported Ir nanoparticles, and a more than 8-fold larger catalytic activity than commercial Ir blacks. In single-cell PEM electrolyzer tests, the Ir-ND/ATO outperformed commercial Ir catalysts more than 2-fold at technological current densities of 1.5 A cm–2 at a mere 1.80 V cell voltage, while showing excellent durability under constant current conditions. We conclude that Ir-ND/ATO catalysts have the potential to substantially reduce the required noble metal loading, while maintaining their catalytic performance, both in idealized three-electrode set ups and in the real electrolyzer device environments. 相似文献
Highly conductive poly(3,4‐ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) films as transparent electrodes for organic light‐emitting diodes (OLEDs) are doped with a new solvent 1,3‐dimethyl‐2‐imidazolidinone (DMI) and are optimized using solvent post‐treatment. The DMI doped PEDOT:PSS films show significantly enhanced conductivities up to 812.1 S cm−1. The sheet resistance of the PEDOT:PSS films doped with DMI is further reduced by various solvent post‐treatment. The effect of solvent post‐treatment on DMI doped PEDOT:PSS films is investigated and is shown to reduce insulating PSS in the conductive films. The solvent posttreated PEDOT:PSS films are successfully employed as transparent electrodes in white OLEDs. It is shown that the efficiency of OLEDs with the optimized DMI doped PEDOT:PSS films is higher than that of reference OLEDs doped with a conventional solvent (ethylene glycol). The results present that the optimized PEDOT:PSS films with the new solvent of DMI can be a promising transparent electrode for low‐cost, efficient ITO‐free white OLEDs.
The present study demonstrates that one-step peptide backbone fragmentations can be achieved using the TEMPO [2-(2,2,6,6-tetramethyl piperidine-1-oxyl)]-assisted free radical-initiated peptide sequencing (FRIPS) mass spectrometry in a hybrid quadrupole time-of-flight (Q-TOF) mass spectrometer and a Q-Exactive Orbitrap instrument in positive ion mode, in contrast to two-step peptide fragmentation in an ion-trap mass spectrometer (reference Anal. Chem. 85, 7044–7051 (30)). In the hybrid Q-TOF and Q-Exactive instruments, higher collisional energies can be applied to the target peptides, compared with the low collisional energies applied by the ion-trap instrument. The higher energy deposition and the additional multiple collisions in the collision cell in both instruments appear to result in one-step peptide backbone dissociations in positive ion mode. This new finding clearly demonstrates that the TEMPO-assisted FRIPS approach is a very useful tool in peptide mass spectrometry research.
Turbulence-intensity measurements were made in a Taylor-Couette flow reactor consisting of two counter-rotating concentric cylinders designed for the purpose of studying turbulent premixed-flame propagation. In the annulus separating the two cylinders, a nearly homogeneous turbulent flow is generated. The intensities of turbulent velocity fluctuations in the annulus in both axial and circumferential directions were measured by using laser-Doppler velocimetry for a wide range of cylinder rotation rates, corresponding to low through high (120 cm/s) intensities relative to typical laminar flame speeds for lean methane-air mixtures. The experimental measurements indicate a linear relation between turbulence intensities and average cylinder surface speed and demonstrate the usefulness of the Taylor-Couette apparatus for studies of premixed-flame propagation in high-intensity turbulent flow. 相似文献
A new certified reference material (CRM) for radionuclides in sea water from the Irish sea (IAEA-443) is described and the results of the certification process are presented. Ten radionuclides (3H, 40K, 90Sr, 137Cs, 234U, 235U, 238U, 238Pu, 239+240Pu and 241Am) have been certified, and information values on massic activities with 95% confidence intervals are given for four radionuclides (230Th, 232Th, 239Pu and 240Pu). Results for less frequently reported radionuclides (99Tc, 228Th, 237Np and 241Pu) are also reported. The CRM can be used for quality assurance/quality control of the analysis of radionuclides in water samples, for the development and validation of analytical methods and for training purposes.
