Structure reinvestigation of gelsemoxonine,a constituent of Gelsemium elegans,reveals a novel,azetidine-containing indole alkaloid

[structure: see text] The structure of gelsemoxonine, isolated from Gelsemium elegans Benth., was revised to be a novel oxindole alkaloid having an azetidine unit. A new alkaloid, 14,15-dihydroxygelsenicine, which was presumed to be a biosynthetic precursor of gelsemoxonine, was also isolated.     

Solvatochromic studies of interactions between surfactants and poly(N-substituted acrylamide)s

Interactions between poly(N-substituted acrylamide)s and surfactants, such as sodium dodecyl sulfate (SDoS) and sodium decyl sulfate (SDeS), in aqueous solutions were investigated using a solvatochromic probe. The polymers used were poly(N,N-dimethylacrylamide) (PDMA), poly(N-isopropylacrylamide) (PIPA), poly(N-acryloylpyrrolidine) (PAPR), and poly(vinylpyrrolidone) (PVPy) for comparison. They were labeled with pyridinium dicyanomethylide chromophore as a solvatochromic probe, and the changes in the microenvironment polarity of the polymer upon association with surfactant micelles were investigated by monitoring the λ_max in the absorption spectra of the probe molecule. It was found that the Gibbs free energy of micelle stabilization by polymer complexation for SDoS is 7.6, 4.1, and 2.2 kJ mol⁻¹, and for SDeS 5.1, 2.9, and 0.8 kJ mol⁻¹ with PIPA, PAPR, and PDMA, respectively. These results indicate that the complexation between polymer and surfactant is influenced not only by the alkyl-chain length of the surfactant, but also by the polymer side groups.

Preparation and photolysis of polyamides having coumarin dimer component

The ring-opening polyaddition reaction of anti and syn head-to-head Coumarin dimers with diamines and the photocleavage behavior of the resulting polyamides were investigated. Anti head-to-head Coumarin dimer successfully reacted with aliphatic and aromatic diamines in an aprotic polar solvent to give corresponding high molecular weight polyamides. The polyamides showed good film forming ability and exhibited solubility behavior typical of polyelectrolytes. Furthermore, these polyamides were found to undergo exclusively asymmetric photocleavage on the cyclobutane rings in the polymer main chain to give a fumaramide unit with elimination of 2,2′-dihydroxystilbene. On the other hand, syn head-to-head Coumarin dimer gave only low molecular weight polyamides. These polymerization and photocleavage behaviors were elucidated in comparison with those of model reactions.     

Synthesis of Optically Active Aromatic Ether Ketone Macrocycles Containing 2,2′-Dimethoxy-1,1′-binaphthyl-6,6′-diyl Moieties

Optically active aromatic ether ketone macrocycles containing 2,2′-dimethoxy-1,1′-binaphthyl-6,6′-diyl moieties were synthesized through electrophilic aromatic substitution reaction and stepwise nucleophilic aromatic substitution reactions. Structures of the resulting macrocycles were confirmed by NMR and matrix assisted laser desorption/ionization time of flight (MALDI-TOF) mass measurements.     

Studies on the mechanism for stereoisomerization of 1-zirconacyclopent-3-yne compounds

The stereoisomerization of 2,5-disubstituted 1-zirconacyclopent-3-yne compounds, stable five-membered cycloalkynes, has been studied with regard to the mechanism. The bimetallic complex of 1,4-bis(trimethylsilyl)butatriene was synthesized and structurally characterized, although it seems unimportant for the stereoisomerization reactions. The isomerization of trans-1,1-bis(η⁵-cyclopentadienyl)-2,5-bis(trimethylsilyl)-1-zirconacyclopent-3-yne 2a into the cis-form in benzene-d₆ solution were observed using ¹H NMR spectroscopy at 50 °C in various concentrations. The reaction was first order with respect to trans-2a. This ruled out the possibility that a bimetallic complex was responsible for the isomerization. A kinetic isotope effect was observed (k_H/k_D = 1.8), suggesting that C–H activation is involved in the rate-determining step. A mechanism via hydrogen elimination from the complex of η⁴-π,π-coordination mode is proposed.     

New mu-opioid receptor agonists with phenoxyacetic acid moiety

New muq-opioid receptor (MOR) agonists containing 4-hydroxypiperidine, piperidine and piperazine moieties were synthesized and evaluated to find a peripheral opioid analgesic. Among the synthesized compounds, 12-[1-[3-(N,N-dimethylcarbamoyl)-3,3-diphenylpropyl]-4-hydroxypiperidin-4-yl]phenoxy]acetic acid (8: SS620) having phenoxyacetic acid and 4-hydroxypiperidine moieties showed the highest agonist potency on the MOR in an isolated guinea-pig ileum preparation, and it also had selectivity to the human MOR expressed in Chinese hamster ovary (CHO)-K1 cells compared with the same types of delta- and kappa-opioid receptors (DOR and KOR). In addition, compound 8 showed a 10 times more potent MOR agonist activity than loperamide. Furthermore, compound 8 showed a peripheral analgesic activity in vivo screening on rat.     

Detection of multiple anions by thin-layer chromatography

A novel detection method for 21 different anions by thin-layer chromatography is presented. Anions on the target plate form salts with amine in a developing solvent and are visualized after staining with citric acid-acetic anhydride reagent as white spots contrasting against a pale red-pink background. This method has particularly high sensitivity for anions of chlorate, sulfate, phosphate, chromate and dichromate (0.02-0.05 microg). The method is demonstrated to efficiently detect toxic arsenite in curry sauce as an example application. The proposed method offers highly efficient indirect detection for a wide range of anions, and serves as a purification procedure for the preparation of anionic sample solutions for other analytical methods.     

Strange giant resonance in (K −,π−) reaction

We point out a possibility of strange giant resonances that can be excited by the (K ^?,π^?) reaction on nuclear targets. Recent experimental data on⁴⁰Ca are analyzed from this point of view.
