Llama-derived single-domain antibodies for the detection of botulinum A neurotoxin

Single-domain antibodies (sdAb) specific for botulinum neurotoxin serotype A (BoNT A) were selected from an immune llama phage display library derived from a llama that was immunized with BoNT A toxoid. The constructed phage library was panned using two methods: panning on plates coated with BoNT A toxoid (BoNT A Td) and BoNT A complex toxoid (BoNT Ac Td) and panning on microspheres coupled to BoNT A Td and BoNT A toxin (BoNT A Tx). Both panning methods selected for binders that had identical sequences, suggesting that panning on toxoided material may be as effective as panning on bead-immobilized toxin for isolating specific binders. All of the isolated binders tested were observed to recognize bead-immobilized BoNT A Tx in direct binding assays, and showed very little cross-reactivity towards other BoNT serotypes and unrelated protein. Sandwich assays that incorporated selected sdAb as capture and tracer elements demonstrated that all of the sdAb were able to recognize soluble (“live”) BoNT A Tx and BoNT Ac Tx with virtually no cross-reactivity with other BoNT serotypes. The isolated sdAb did not exhibit the high degree of thermal stability often associated with these reagents; after the first heating cycle most of the binding activity was lost, but the portion of the protein that did refold and recover antigen-binding activity showed only minimal loss on subsequent heating and cooling cycles. The binding kinetics of selected binders, assessed by both an equilibrium fluid array assay as well as surface plasmon resonance (SPR) using toxoided material, gave dissociation constants (K _D ) in the range 2.2 × 10⁻¹¹ to 1.6 × 10⁻¹⁰ M. These high-affinity binders may prove beneficial to the development of recombinant reagents for the rapid detection of BoNT A, particularly in field screening and monitoring applications.     

Ligand influence in the selective gold-mediated synthesis of allenes

The use of [(IPr)AuOH] permits the generation of gold(i)-amine complexes by a silver-free protocol. These in situ or well-defined complexes are used in a straightforward synthetic route to substituted allenes from propargylic acetates. The catalytic activity can be modulated as a function of the nature of the ligand bound to the gold-NHC moiety.     

Analysis of large and non-standard geometry samples of ancient potteries by internal monostandard neutron activation analysis using in situ detection efficiency

The k ₀-based internal monostandard neutron activation analysis (IM-NAA) method was used for the analysis of 30 large and non-standard geometry ancient pottery samples obtained from Buddhist sites of Andhra Pradesh, India. One freshly finished pottery and a sun-drenched pottery were also analyzed for comparison. Samples were irradiated in thermal column facility of Apsara reactor and also in graphite reflector position of critical facility of Bhabha Atomic Research Centre. Radioactive assay was carried out using a 40% relative efficiency HPGe detector coupled to MCA. Concentration ratios of 15 elements with respect to Sc were determined. The La/Ce values as well as statistical cluster analysis utilizing concentration ratios of elements were used for grouping/provenance of the potteries.     

Higher-order Poincaré sphere, stokes parameters, and the angular momentum of light

A higher-order Poincaré sphere and Stokes parameter representation of the higher-order states of polarization of vector vortex beams that includes radial and azimuthal polarized cylindrical vector beams is presented. The higher-order Poincaré sphere is constructed by naturally extending the Jones vector basis of plane wave polarization in terms of optical spin angular momentum to the total optical angular momentum that includes higher dimensional orbital angular momentum. The salient properties of this representation are illustrated by its ability to describe the higher-order modes of optical fiber waveguides, more exotic vector beams, and a higher-order Pancharatnam-Berry geometric phase.     

Autoresonant excitation of antiproton plasmas

We demonstrate controllable excitation of the center-of-mass longitudinal motion of a thermal antiproton plasma using a swept-frequency autoresonant drive. When the plasma is cold, dense, and highly collective in nature, we observe that the entire system behaves as a single-particle nonlinear oscillator, as predicted by a recent theory. In contrast, only a fraction of the antiprotons in a warm plasma can be similarly excited. Antihydrogen was produced and trapped by using this technique to drive antiprotons into a positron plasma, thereby initiating atomic recombination.     

Synthesis,structural and thermal characterization of metaphosphatecobalt(II) salt

A new inorganically template metaphosphate of Co(II) complex has been synthesized and characterized by different measurements such as DSC, FT-IR, C–H–N–O–S, ESR, TG-DTA and X-RD. Differential Scanning Calorimeter (DSC) elucidated negative specific heat of the system and has used to evaluate some thermo dynamical constants like activation energy (E _a), frequency factor (A), enthalpy and entropy of that system. The specific heat capacity of the system is measured both in atmospheric O₂ and N₂ atmosphere at different heating rates of 278, 283, 293 and 298 K min⁻¹ in room atmosphere and 288 K min⁻¹ in N₂ atmosphere.
