Synthesis and crystal structures of novel calix[4]azacrowns

A series of novel 1,3‐altemate calix[4]arene azacrowns having mono and bis crown ethers on the lower rim of the calix[4]arene framework were synthesized. Solid‐state structures confirmed the three dimensional conformation of compounds 1–3.     

Cyclization of 1-bromo-2,7- and 1-bromo-2,8-enynes mediated by indium

[reaction: see text] The cyclization of 1-bromo-2,7- and 1-bromo-2,8-enynes mediated by indium in DMF produced five- and six-membered cyclic compounds. Although KI was a necessary additive in the cyclization of terminal 1-bromo-2,7-enynes to give the desired products at 25 degrees C, reactions of terminal 1-bromo-2,8-enynes and internal 1-bromo-2,7-enynes with indium proceeded at 100 degrees C in DMF without KI. After cyclizations, subsequent cross-coupling reaction and iodolysis increase the usefulness of this reaction.     

Computer-assisted optimization of reversed-phase HPLC isocratic separation of neutral compounds

An appropriate optimization strategy should be used to find a desired resolution or selectivity with a minimum number of experiments in a limited time, which could assure the baseline separation of all target compounds. It was usually realized by means of a specialized computer program. In this paper, mapping optimization method and overlapping resolution mapping were compared for the optimization of a reversed-phase high-performance liquid chromatography (HPLC) isocratic separation of neutral compounds. The calculated resolutions and separation time of 7 to 10 experiments are fitted by different equations, which were used to build a contour plot with a minimum effective resolution and maximum retention time as a function of a mobile phase composition. The balance between resolution and analysis time could be easily realized by the overlapping of the final overlapping resolution mapping and analysis time mapping. The validity of the two methods was confirmed by some typical experiments. The models are simple, visual, and common without theoretical arithmetic.     

Improved Performance of an Amperometric Biosensor with Polydiaminonaphthalene on Electrochemically Deposited Au Nanoparticles

The performance of an enzyme sensor fabricated through covalent bond formation on the HRP‐bonded poly(1,8‐diaminonaphthalene) (polyDAN) layer with gold nanoparticles (AuNPs) was applied to catalyze the electrochemical reduction of H₂O₂. The surface characteristics of the sensor probe were studied using cyclic voltammetry, SEM, XPS, QCM, and impedance spectroscopy. The AuNP‐deposited surface resulted in higher conductivity and sensitivity for H₂O₂ detection in phosphate buffer solution. A linear calibration plot was obtained in the H₂O₂ concentration range between 10.0 μM and 25.0 mM with detection limit 5.0±1.25 μM. The lifetime of HRP/polyDAN/AuNP/GC probe was over 70 days without response loss.     

Hydrophilic functionalization of syndiotactic polystyrene via a combination of electrophilic bromination and Suzuki–Miyaura reaction

Postfunctionalization of high‐molecular‐weight syndiotactic polystyrene (sPS) was achieved via combination of electrophilic bromination at the para‐position of the polymer aromatic ring and subsequent Suzuki–Miyaura cross‐coupling reactions with functionalized phenylboronic acids. The concentration of brominated styrene repeating unit in sPS was conveniently controlled by changing the ratio of added bromine relative to the polymer repeating unit. Brominated sPS (8.5 mol %) was converted quantitatively to other polar functional groups via Suzuki–Miyaura cross‐coupling reactions with various functional group‐substituted phenylboronic acids. Phosphorus ligands for iron(III)‐mediated ATRP of styrene via generation of activators by monomer addition

Styrene polymerization via generation of activators by monomer addition (GAMA) for atom transfer radical polymerization (ATRP) has been examined extensively with bulk FeX₃ and FeX₂ at 110 °C in conjunction with various phosphorus‐bearing ligands. It was found that GAMA possesses advantages over normal ATRP. Most importantly, narrower polydispersity index (PDI) values were observed from the styrene polymerizations with Fe(III) over those with Fe(II). Every instance of 2‐(diphenylphosphino)‐N,N′‐dimethyl‐[1,1′‐biphenyl]‐2‐amine and 2‐(diphenylphosphino) pyridine with the Fe(III) system were controlled excellently without addition of any radical initiator or reducing agent additives. Initiator type was found to exert a significant factor to influence on the controllability of polymerization. The initiation of 1‐phenylethyl chloride and methyl‐2‐chloropropionate gave rise to formation of polymers with narrow PDI (1.05–1.20), whereas those from 1‐phenylethyl bromide increased to 1.35. The GAMA of bulk styrene exhibited the best performance in terms of both rate and controllability compared with toluene and anisole. Both formation of block copolymer from the macroinitiator and efficient perturbation of polymerization with 2,2,6,6‐tetramethylpiperidine 1‐oxyl provided firm evidence to support the living and radical characteristics for the GAMA of styrene. © 2009 Wiley Periodicals, Inc. Cytochrome P450 1A1 (CYP1A1) polymorphisms and breast cancer risk in Korean women

Cytochrome P450 1A1 (CYP1A1) is involved in the 2-hydroxylation of estrogen, the hormone that plays a critical role in the etiology of breast carcinoma. We evaluated the associations between two CYP1A1 polymorphisms [MspI (rs4646903); Ile462Val (rs1048943)] and breast cancer in a multicenter case-control study of 513 breast cancer cases and 447 controls in Korea. Women carrying the T allele of the CYP1A1 MspI polymorphism were found to have a 1.72-fold (95% CI 1.11-2.68) greater risk of developing breast cancer. No association was found between any CYP1A1 Ile462Val polymorphism and breast cancer. Haplotype analysis of the two loci showed that the CA haplotype was associated with the lowest risk of breast cancer, and CA/CA diplotypes were associated with a lower risk of breast cancer [OR=0.28 (0.13-0.61)] than others/others diplotypes. Moreover, this reduced risk was more pronounced among women with a lower body mass index (BMI) [OR=0.18 (0.06-0.58)] or with a shorter lifetime exposure to estrogen [OR=0.23 (0.07-0.81)]. The results obtained suggest that the CYP1A1 MspI polymorphisms could affect susceptibility to breast cancer.     

Photocatalytic decomposition of orange II over TiO2-loaded on SBA-15 prepared using a microwave process

SBA-15 mesoporous material was prepared by microwave-hydrothermal method and was used as support in TiO₂-loaded SBA-15 photocatalysts. The physical properties of these particles were investigated. We also examined the activity of these samples as photocatalysts for the decomposition of orange II. Titania loaded on a silica matrix decreases the surface area of the support as expected for TiO₂ incorporation. For TiO₂-loaded SBA-15 photocatalysts, the IR absorption at ∼960 cm⁻¹ commonly accepted as the characteristic vibration of the Ti-O-Si bond. The photocatalytic activity increases with an increase of the TiO₂ loading.     

TNF-alpha upregulates PTEN via NF-kappaB signaling pathways in human leukemic cells

TNF-alpha plays a variety of biological functions such as apoptosis, inflammation and immunity. PTEN also has various cellular function including cell growth, proliferation, migration and differentiation. Thus, possible relationships between the two molecules are suggested. TNF-alpha has been known to downregulate PTEN via NF-kappaB pathway in the human colon cell line, HT-29. However, here we show the opposite finding that TNF-alpha upregulates PTEN via activation of NF-kappaB in human leukemic cells. TNF-alpha increased PTEN expression at HL-60 cells in a time- and dose-dependent manner, but the response was abolished by disruption of NF-kappaB with p65 antisense phosphorothioate oligonucleotide or pyrrolidine dithiocarbamate. We found that TNF-alpha activated the NF-kappaB pathways, evidenced by the translocation of p65 to the nucleus in TNF-alpha-treated cells. We conclude that TNF-alpha induces upregulation of PTEN expression through NF-kappaB activation in human leukemic cells.     

X-ray crystal structure of the tetra(tert-butyl)erbate anion and attempts to prepare tetravalent organolanthanide complexes

The new [Li(DME)₃⁺] salt of the previously-known tetra(tert-butyl)erbate(III) anion [Er(t-Bu)₄⁻] has been prepared and structurally characterized. The erbium(III) center is ligated by four tert-butyl groups in an approximately tetrahedral arrangement. The C–Er–C angles between the tert-butyl groups range from 108.8(3)° to 111.2(3)° and the Er–C distances range from 2.352(6) to 2.395(6) Å. The lithium cation is surrounded by three DME molecules, which form a distorted octahedral coordination sphere. Attempts to oxidize the analogous terbate complex [Li(DME)₃][Tb(t-Bu)₄] and its cerium analog to electrically neutral tetra(alkyl)lanthanide(IV) compounds are described.
