A HPLC-UV method has been developed for assaying rifampicin in plasma and liver. The assay involved a liquid-liquid extraction procedure with dichloromethane-pentane (1:1). An Ultrabase-C18 column and a simple mobile phase consisting of a water (pH 2.27)-acetonitrile (40:60, v/v) mixture were used. The flow-rate was 1 ml/min and the effluent was monitored at 333 nm. Results from the HPLC analyses showed that the assay method is linear in the ranges 0.1-1 and 1-50 microg/ml for plasma, and 0.6-40 microg/g for liver. Intra- and inter-day R.S.D. were below 15% for all the sample types. Recoveries averaged 83 and 95% for plasma and liver, respectively. The method is being successfully applied to determine rifampicin in plasma and liver samples taken during pharmacokinetic studies in rats.  相似文献

A capillary zone electrophoresis (CZE) method is proposed for the quantitative determination of anthocyanins in wine as an alternative to high-performance liquid chromatography. The CZE separation was carried out using a 46 cm (effective length)×75 μm I.D. fused-silica capillary at 10 °C and a 50 mM sodium tetraborate buffer at pH 8.4 with 15% of methanol as modifier. A voltage of 25 kV and a hydrodynamic injection of 300 mbar s were used. The electropherograms were recorded at 599 nm. It was found that SO2 (antibacterial and antioxidant agent added to wine during its production) increased the absorbance of anthocyanins at 599 nm in a basic medium. Therefore, a concentration of 250 mg/l of SO2 was added to the samples and the calibration solution before the analysis in order to avoid errors by this matrix effect. The analytical response was linear (R=0.998) between 10 and 700 μg/ml of malvidin-3-O-glucoside. The limit of detection and the reproducibility (as a relative standard deviation, n=11) were 1 μg/ml and 1.5%, respectively. Finally, the CZE method was validated by the analysis of synthetic wine samples (errors less than 8%) and by the comparison of the results obtained in the analysis of different monovarietal wines by CZE with those obtained by the standard HPLC method. In this comparison, a good correlation (R=0.998) with a slope of 1.005±0.044 and an intercept of −0.752±6.690 was obtained for malvidin-3-O-glucoside.  相似文献

The determination of tetracycline by fluorescence spectrophotometry in complex matrices has some difficulties, because the presence of other compounds in the matrix affects the analytical signal. In this work, the effect of some inorganic species that are present in whey milk on the fluorescence signal of tetracycline is studied using a D-optimal experimental design. Next, an experimental strategy is proposed in conjunction with Parallel Factor Analysis, PARAFAC, modeling that leads to suitably modeling the severe matrix effect in the determination of tetracycline in whey milk. A specific design is performed in such a way that the lack of trilinearity due to the effect of the presence of interferents on the signal is obviated. Then, ten test samples from three brands of milk, spiked with different quantities of tetracycline and measured in 2 days were analysed using this methodology (mean of the absolute value of the relative errors: 5.1%). The developed analytical method fulfils the property of trueness, the relative errors being, both in calibration and prediction, inside the interval set by Commission Decision 2002/657/EC at these concentration levels. Decision limits (CCα) at x0 = 0 μg L−1 and at x0 = 100 μg L−1 were 13.2 and 112.4 μg L−1 respectively, for α = 0.05; whereas detection capabilities (CCβ) were 25.9 μg L−1 and 124.4 μg L−1 respectively for α = β = 0.05.  相似文献

A new method toward vertically oriented poly(3,4‐ethylenedioxythiophene) (PEDOT) nanotube arrays on transparent conductive oxide substrates is presented. The approach is based on the use of ZnO nanowire arrays as templates for the electropolymerization of PEDOT. Robust arrays of vertically oriented PEDOT nanotubes with different lengths and wall thicknesses were obtained by modifying the ZnO nanowire length and charge density passed during the electropolymerization, respectively. Furthermore, PEDOT nanotubes with different morphologies (top‐closed and mushroom‐like) were successfully designed by varying the PEDOT electropolymerization kinetics or monomer diffusion or both. © 2010 Wiley Periodicals, Inc. This study describes the development of a new analytical method for determining 14 personal care products (PCPs) – nine synthetic musks, four parabens and one insect repellent – in air samples. The method is based on active sampling on sorbent tubes and thermal desorption-gas chromatography–mass spectrometry analysis, and is rapid, sensitive and drastically reduces the risk of sample contamination. Three kinds of tubes and traps were tested, those filled with Tenax TA being the most suitable for this study. Method validation showed good repeatability and reproducibility, low detection limits (between 0.03 ng m−3 for DPMI and 12.5 ng m−3 for propyl paraben) and good linearity for all compounds. Stability during storage indicated that samples must be kept refrigerated at 4 °C and analysed within 1 week of collection. The applicability of the technique to real samples was tested in different indoor and outdoor atmospheres. The total PCP values for indoor air ranged from 135 ng m−3 in a pharmacy to 2838 ng m−3 in a hairdresser's, whereas the values for outdoor air ranged from 14 ng m−3 for a suburban environment to 26 ng m−3 for an urban environment. In general, the most abundant synthetic musks were galaxolide (5.9–1256 ng m−3), musk xylene (1.6–766 ng m−3) and tonalide (1.1–138 ng m−3). Methyl and ethyl paraben (2.4–313 ng m−3 and 1.8–117 ng m−3, respectively) were the most abundant parabens. Although thermal desorption methods have been widely used for determining volatile organic compounds, they are rarely used with semi-volatile compounds. This study thus demonstrates that the thermal desorption method performs well with semi-volatile compounds and, for the first time, that it can be used for determining PCPs.  相似文献

In this work, experimental data of isobaric vapour–liquid equilibria for the ternary system ethanol + water + 1-hexyl-3-methylimidazolium chloride ([C6mim][Cl]) and for the corresponding binary systems containing the ionic liquid (ethanol + [C6mim][Cl], water + [C6mim][Cl]) were carried out at 101.300 kPa. VLE experimental data of binary and ternary systems were correlated using the NRTL equation. In a previous work [N. Calvar, B. González, E. Gómez, A. Domínguez, J. Chem. Eng. Data 51 (2006) 2178–2181], the VLE of the ternary system ethanol + water + [C4mim][Cl] was determined and correlated, so we can study the influence of different ionic liquids in the behaviour of the azeotropic mixture ethanol–water.  相似文献

Summary A method for the supercritical fluid extraction of carbamate pesticides (propoxur, aminocarb, carbaryl and methiocarb) from soil and cereal samples using CO2 is proposed. Extractions were at 378 bar and 54 °C. Analytes were determined in the extracts by HPLC with fluorescence detection after post-column derivatization. Recoveries from spiked soil ranged between 39.6 and 91.7%, depending on analyte and soil components. Lowest recoveries were from sandy soils. Aminocarb could not be recovered from any soil using CO2. Recovery of aminocarb from diatomaceous earth was improved by adding methanol to the extraction cell prior to SFE, but the effect was not observed in soil samples. Recoveries for propoxur and aminocarb from spiked wheat were about 75%, and only between 30–50% for aminocarb from corn and oats, and carbaryl from wheat. Fat was coextracted using CO2 and retained in the trap together with the analytes, however, appropriate rinsing solvent allowed on-line clean-up of the extract.  相似文献

This work studied the possibility of using polyethyleimine (PEI) as an affinity ligand for the purification of plasmid DNA (pDNA) from alkaline lysates using aqueous two-phase systems (ATPSs). The goal was to find conditions under which this cationic polymer could steer the partition of pDNA to the phase where less impurities accumulate. In poly(ethylene glycol) (PEG)/ammonium sulphate systems, neither free nor PEGylated PEI (pPEI) were able to change the partition of pDNA. This is probably due to the high salt concentration present in these systems that impair the interaction between pDNA and PEI. In PEG 3350/dextran 110 systems, the desired effect could be observed but 0.2-0.5M ammonium sulphate had to be added to prevent the co-partition of RNA to the same phase. These results were used to develop a methodology to obtain polyplexes from alkaline lysates in a two-step ATPSs extraction process. In the first step, a PEG 600/ammonium sulphate system is used to remove most impurities to the top phase. The pDNA-containing bottom phase is then isolated and contacted with a second PEG 3350/dextran 110 system supplemented with a small amount of pPEI (0.2%). Plasmid yield was 100% and the final preparation had no RNA and only small amounts of contaminant protein. Additionally, pDNA was obtained in the form of 53nm-sized polyplexes which are likely to suit specific gene delivery applications.  相似文献
