Monadic MV-algebras are an algebraic model of the predicate calculus of the Łukasiewicz infinite valued logic in which only a single individual variable occurs. GMV-algebras are a non-commutative generalization of MV-algebras and are an algebraic counterpart of the non-commutative Łukasiewicz infinite valued logic. We introduce monadic GMV-algebras and describe their connections to certain couples of GMV-algebras and to left adjoint mappings of canonical embeddings of GMV-algebras. Furthermore, functional MGMV-algebras are studied and polyadic GMV-algebras are introduced and discussed. The first author was supported by the Council of Czech Government, MSM 6198959214.  相似文献

The antimicrobial activity of 16 newly prepared quinolizidines derivatives using bacteria (Staphylococcus aureus, Staphylococcus epidermidis, Proteus sp., Escherichia coli) acid fast bacterium Mycobacterium smegmatis, yeasts (Candida albicans, Candida parapsilosis), and filamentous fungi (Fusarium culmorum, Microsporum gypseum, Aspergillus flavus, Botrytis cinerea, Alternaria alternata) was studied in this paper. The best antibacterial properties were demonstrated by derivatives 11Ba, trans10Bb and 11Bb, and the most sensitive microorganism was found to be the gram-positive bacterium S. epidermidis. The derivative 11Bb showed the best antifungal activity, while C. albicans was resistant to all tested derivatives, and C. parapsilosis was fully inhibited in the presence of the derivative 11Ba and 11Bb. Among the filamentous fungi, only the dermatophyte M. gypseum was partially inhibited. Biofilms represent the most prevalent type of microbial growth in nature and are crucial to the development of clinical infections. Newly synthesized derivatives were also added into the medium throughout the biofilm formation. We have observed a significant decrease of biofilm formation in the presence of quinolizidine derivatives, testifying to their significant antimicrobial activity. It seems that the relationship between antimicrobial activity and the structure is based on the alkaline character due to nitrogen, the saturated basic quinolizidine skeleton, and the position of sulfur in the molecule.  相似文献

The Becke3LYP density functional was used to study structural and thermodynamic parameters of bivalent zinc cation complexes with selected substrates and ACE inhibitors (H2O/OH, neutral forms of captopril, zofenoprilat, omapatrilat, CH3CONHCH3, and N-terminal anions of captopril, zofenoprilat, omapatrilat, enalaprilat, perindoprilat, trandolaprilat, and fosinoprilat). The combination of DFT and the conductor-like polarizable continuum model (CPCM) were employed to compute the Gibbs interaction energies (ΔG) between Zn2+ and the selected ACE inhibitors for dielectric media with ɛ = 5 (to simulate the protein environment) and for water media (ɛ = 78.39) for comparison purposes. The results show that ΔG is sensitive to the dielectric constant of the environment and that lower dielectric medium favors the binding of inhibitors to the zinc cation.  相似文献

Prion diseases are incurable neurodegenerative diseases that affect both humans and animals. The infectious agent is a pathogenic form of the prion protein that accumulates in brain as amyloids. Currently, there is neither cure nor reliable preclinical diagnostics on the market available. The growing number of reports shows that passive immunisation is one of the most promising strategies for prion disease therapy, where antibodies against prions may prevent and even cure the infection. Since antibodies are large molecules and, thus, might not be suitable for the therapy, different antibody fragments are a good alternative. Therefore, we have designed and prepared single-chain antibody fragments (scFvs) derived from the PrPSc-specific murine monoclonal antibody V5B2. Using a new expression vector pMD204, we produced scFvs in two opposing chain orientations in the periplasm of Escherichia coli. Both recombinant antibody fragments retained the specificity of the parent antibody and one of these exhibited binding properties comparable to the corresponding murine Fab fragments with the affinity in nM range. Our monovalent antibody fragments are of special interest in view of possible therapeutic reagents for prion diseases as well as for development of a new generation of diagnostics.  相似文献

The inorganic ceramic compounds based on the CeO2 belong into the group of high-temperature pigments. The pigments have been prepared by the classical dry process (i.e. solid-state reaction) in the temperature range from 1,300 to 1,600 °C and by the coprecipitation at the three different temperatures: 400, 600 and 1,100 °C. The principal of these pigments makes the host lattice of the CeO2, which is doped by terbium ions. This incorporation of the doped ions leads to obtaining of the interesting dark orange colour after application into ceramic glaze. The aim of our research was to improve and optimize the synthesis conditions of these pigments. The samples were submitted to thermal analysis (TG–DTA) for determination of the temperature interval of the pigment formation and the thermal stability of pigments. The compounds were also measured from the point of view of their colouring, structure and particle size distribution.  相似文献

A method for the fast determination of yohimbine, a potent adrenoreceptor antagonist used for the treatment of sexual dysfunctions, is proposed in this article. MEKC under basic and acidic conditions (sodium borate, pH 9.5 and sodium phosphate, pH 2.5) with SDS was developed. The effect of the experimental parameters, e.g. pH, SDS concentration and injection time, on yohimbine migration was also studied. Both methods were validated in terms of linearity, limits of detection and quantification, accuracy, and precision using caffeine as an internal standard. The application for the determination of yohimbine in hand-made medicaments is also investigated in this study.   相似文献

In this paper the response of an amperometric biosensor at mixed enzyme kinetics and diffusion limitations is modelled in the case of the substrate and the product inhibition. The model is based on non-stationary reaction–diffusion equations containing a non-linear term related to non-Michaelis–Menten kinetics of an enzymatic reaction. A numerical simulation was carried out using a finite difference technique. The complex enzyme kinetics produced different calibration curves for the response at the transition and the steady-state. The biosensor operation is analysed with a special emphasis to the conditions at which the biosensor response change shows a maximal value. The dependence of the biosensor sensitivity on the biosensor configuration is also investigated. Results of the simulation are compared with known analytical results and with previously conducted researches on the biosensors.  相似文献

Various types of carbon nanoparticles were directly mixed with microbial cells of Gluconobacter oxydans within a 3-D bionanocomposite in order to prepare a highly sensitive ethanol biosensor with a short response time. From all carbonaceous nanomaterials tested, single- or multi-walled carbon nanotubes provided the highest sensitivity of detection (117–121 µA cm?2 mM?1), but from a practical point of view, Ketjen black 300 and 600 provide very low detection limit (2–6 µM) and high sensitivity for the ethanol analysis (84–88 µA cm?2 mM?1)with a shortresponse time (14–33 s). Moreover, the price of Ketjen black is a few orders of magnitude lower compared to that of carbon nanotubes. Finally, the study showed that the morphology of nanoparticles rather than their surface modification is the key element in achieving high sensitivity of ethanol detection.  相似文献

The thickness shear mode acoustic method was used to study the binding of thrombin to DNA aptamers immobilised on the gold surface covered by DNA tetrahedrons. The binding of thrombin to conventional aptamers sensitive to fibrinogen (FBT) and heparin (HPT) exosites as well as to HPT in a loop configuration (HPTloop) made it possible to determine the constant of dissociation (KD) and the limit of detection (LOD). The sensing system composed of a HPTloop was characterised by KD = (66.7 ± 22.7) nM, which was almost twice as low as that of FBT and HPT. For this biosensor, a lower LOD of 5.2 nM compared with 17 nM for conventional HPT aptamers was also obtained. Less sensitive sensors based on FBT aptamers revealed an LOD of 30 nM which is in agreement with the lower affinity of these aptamers to thrombin in comparison with that of HPT. The surface concentration of DNA tetrahedrons was determined by the electrochemical method using [Ru(NH3)6]3+ as a redox probe. These experiments confirmed that the “step by step” method of forming the sensing layer, consisting first in chemisorption of DNA tetrahedrons onto a gold surface and then in hybridisation of the aptamer-supporting part with complementary oligos at the top of the tetrahedron, is preferable. In addition, atomic force microscopy was applied to analyse the topography of the gold layers modified stepwise by DNA tetrahedrons, DNA aptamers and thrombin. The height profiles of the layers were in reasonable agreement with the dimensions of the adsorbed molecules. The results indicate that DNA tetrahedrons represent an efficient platform for immobilisation of aptamers.  相似文献
