Capillary electrophoresis (CE) has been combined with atmospheric pressure photoionization (APPI) and electrospray ionization (ESI) for mass spectrometric (MS) detection. Separation conditions using potassium phosphate buffer and ammonium formate buffer have been compared for analysis of eleven pharmaceutical bases. The results showed improvements in separation efficiency and peak symmetry when phosphate buffer was used. The low flow in CE may enable utilization of these advances with MS detection. Compared with ESI, the APPI technique provided a cluster-free background. The enhanced signal-to-noise ratio in the total ion current (TIC) and the reduced spectral background indicated that the APPI process is less affected by non-volatile salts in the CE buffers. This results in a wider range of choice of CE buffers in CE/MS analysis when APPI is the ionization method.  相似文献

The galectins are a family of [small beta]-galactoside-binding proteins that have been implicated in cancer and inflammation processes. Herein, we report the synthesis of a library of 28 compounds that was tested for binding to galectins-1, -3, -7, -8N and -9N. An aromatic nucleophilic substitution reaction between 1,5-difluoro-2,4-dinitrobenzene and a galacto thiol gave 5-fluoro-2,4-dinitrophenyl 2,3,4,6-tetra-O-acetyl-1-thio-beta-D-galactopyranoside. This versatile intermediate was then modified in a two dimensional manner: either by further substitution of the second fluoride by amines or thiols, or by reduction of the nitro groups and acylation of the resulting amines, or both. Deacetylation then gave a library of aromatic beta-galactosides that showed variable inhibitory activity against the different galectins, as shown by screening with a fluorescence-polarisation assay. Particularly efficient inhibitors were found against galectin-7, while less impressive enhancements of inhibitor affinity over methyl beta-D-galactopyranoside were found for galectin-1, -3, -8N and -9N. The best inhibitors against galectin-7 showed significantly higher affinity (K(d) as low as 140 microM) than both beta-methyl galactoside (K(d) 4.8 mM) and the unsubstituted beta-phenyl thiogalactoside (non-inhibitory). The best inhibitors against galectin-7 were poor against the other galectins and thus have potential as structurally simple and selective tools for dissecting biological functions of galectin-7.  相似文献

Dual activity of phosphoryl choline (PC) functional poly(trimethylene carbonate) (PTMC) was found which induces the zwitterionic biomimetic PC group to form physical cross-links with ionomers in the bulk, and at the same time enrich at the surface of cast films. The formation of zwitterionic domains from a bifunctional PC-PTMC-PC (ionomer) provided firm films with a low elastic modulus in contrast to the tacky PTMC starting material (Mn approximately 3900 g/mol) with poor mechanical performance. In addition, the ionomer possessed improved hemocompatible properties that was explained by the enrichment of PC at the surface, suggesting a way to tailor the mechanical performance of biodegradable PTMC-based ionomers while providing its bioactivity. Tailored elasticity while maintaining hemocompatibility of a biodegradable ionomer should be of particular interest for a variety of in vivo applications.  相似文献

The local hydrogen-bonding environment in supercritical water (380 degrees C, 300 bars, density 0.54 gcm3) was studied by x-ray Raman scattering at the oxygen K edge. The spectra are compared to those of the gas phase, liquid surface, bulk liquid, and bulk ice, as well as to calculated spectra. The experimental model systems are used to assign spectral features and to quantify specific local hydrogen-bonding situations in supercritical water. The first coordination shell of the molecules is characterized in more detail with the aid of the calculations. Our analysis suggests that approximately 65% of the molecules in supercritical water are hydrogen bonded in configurations that are distinctly different from those in liquid water and ice. In contrast to liquid water the bonded molecules in supercritical water have four intact hydrogen bonds and in contrast to ice large variations of bond angles and distances are observed. The remaining approximately 35% of the molecules exhibit two free O-H bonds and are thus either not involved in hydrogen bonding at all or have one or two hydrogen bonds on the oxygen side. We determine an average O-O distance of 3.1+/-0.1 A in supercritical water for the H bonded molecules at the conditions studied here. This and the corresponding hydrogen bond lengths are shown to agree with neutron- and x-ray-diffraction data at similar conditions. Our results on the local hydrogen-bonding environment with mainly two disparate hydrogen-bonding configurations are consistent with an extended structural model of supercritical water as a heterogeneous system with small patches of bonded molecules in various tetrahedral configurations and surrounding nonbonded gas-phase-like molecules.  相似文献

A new chromatography system, AKTAxpress (GE Healthcare, Amersham Biosciences, Uppsala, Sweden) has been designed to meet the demand for high-throughput purification of proteins in structural genomics and drug discovery. The system offers a number of automated multistep purification protocols for affinity-tagged proteins. All protocols start with affinity chromatography followed by combinations of desalting, ion exchange chromatography and gel filtration. As an option, tag removal can be included in the purification protocols. Up to 16 proteins can be purified per day and the yield can be as high as 50 mg of each protein at > 90% purity. To highlight the versatility of the system, this paper presents several case studies; purifications of hexahistidine- and glutathione S-transferase-tagged proteins using different protocols, automated on-column tag cleavage and optimization studies for a hexahistidine-tagged kinase.  相似文献

First, standard mixtures of trimethylsilyl (TMS) derivatives of amino acid and organic acid are analyzed by comprehensive two-dimensional (2D) gas chromatography (GC) coupled to time-of-flight mass spectrometry (GC × GC/TOFMS) in order to illustrate important issues regarding application of the technique. Specifically of interest is the extent to which the peak capacity of the 2D separation space has been utilized and the procedure by which the derivative standards are identified in the 2D separations using the mass spectral information. The resulting 2D separation is found to make extensive use of the GC × GC separation space provided by the complementary stationary phases employed.Second, in order to demonstrate GC × GC/TOFMS on two real sample types, trimethylsilyl metabolite derivatives were analyzed from extracts of common lawn grass samples (i.e., perennial rye grass), as a means to provide insight into both the pre and post harvest physiology. Various chemical components in the two rye grass extract samples were found to either emerge or disappear in relation to the trauma response. For example, a significant difference in the peak for the TMS derivative of malic acid was found. The successful analysis of various components was readily facilitated by the 2D separation, while a 1D separation would have produced too much peak overlap, thus impeding the analysis. The importance of using a GC × GC separation approach for the analysis of complex samples, such as metabolite extracts, is therefore demonstrated. The real-time analysis capability of GC × GC/TOFMS for multidimensional metabolite analysis makes this technique well suited to the high-throughput analysis of metabolomic samples, especially compared to slower, stopped-flow type separation approaches.  相似文献

Solid-phase microextraction (SPME) has been applied to the quantitative analysis of 60 volatile organic compounds (VOCs) in drinking water. Equilibration curves for the partitioning of the VOCs between the fiber coating and fortified water obtained at 20, 50, and 80 °C are found between the theoretical curves for completely agitated and non-agitated samples. Two important factors for the amount adsorbed by the SPME fiber coating are the extraction time and the fiber coating/water distribution coefficient, KFW . Both depend on the sample temperature, but in a counteracting manner: Increasing the temperature shortened the equilibration times, especially for the heavier VOCs, but also lead to lower KFW values, and consequently a lower sensitivity of the method. KFW values are determined for 33 of the VOCs at 40, 60, and 80°C and the heats of adsorption,–ΔH, are calculated. The nature of the adsorption is found to be exothermic which explains the decreasing sensitivity of the method with increasing temperature. Detection limits were typically from 20 ng/l to 200 ng/l, except for the very light VOCs with which detection difficulties were encountered. For all of the VOCs the linear range extended from the lowest concentration at which they were actually detected to at least 5 mg/l. The precision, 3% average standard deviation when an internal standard was used, was satisfactory for most quantitative routine analysis. SPME was also applied to head-space (HS) analysis of drinking water through the coupled equilibrium between water/head-space/fiber coating. HS-SPME is demonstrated to have shorter equilibration times than SPME directly from the water and equal sensitivities, except for the very light VOCs. Water samples from a drinking water plant contaminated in the low μg/l range with 1,1,1-trichloroethane, trichloroethene and tetrachloroethene were analyzed. There seems to be a reasonable agreement between results obtained by SPME and purge & trap. It is concluded that SPME has a great potential for drinking water analysis.  相似文献

The age of plutonium is defined as the time since the last separation of the plutonium isotopes from their daughter nuclides. In this paper, a method for age determination based on analysis of 241Pu/241Am and 240Pu/236Pu using ICP-SFMS is described. Separation of Pu and Am was performed using a solid phase extraction procedure including UTEVA, TEVA, TRU and Ln-resins. The procedure provided separation factors adequate for this purpose. Age determinations were performed on two plutonium reference solutions from the Institute for Reference Materials and Measurements, IRMM081 (239Pu) and IRMM083 (240Pu), on sediment from the Marshall Islands (reference material IAEA367) and on soil from the Trinity test site (Trinitite). The measured ages based on the 241Am/241Pu ratio corresponded well with the time since the last parent-daughter separations of all the materials. The ages derived from the 236U/240Pu ratio were in agreement for the IRMM materials, but for IAEA367 the determination of 236U was interfered by tailing from 238U, and for Trinitite the determined age was biased due to formation of 236U in the detonation of the “Gadget”.  相似文献
