Novel quinolone derivatives featuring an 1,3,4-oxadiazole ring as a metal-chelating component and a benzyl group base on HIV-1 integrase inhibitors pharmacophore were designed and synthesized. An antiviral assay revealed that most analogues inhibited HIV-1 replication in the cell culture. Our results showed that compounds bearing small alkyl groups as R group were inactive in anti-HIV-1 assay, whereas compounds possessing benzyl or substituted benzyl at the same position showed good anti-HIV activity with the range of 20–57% at 100 μM concentration. Among them, 3-(5-((2-fluorobenzyl)thio)-1,3,4-oxadiazol-2-yl)-8-phenylquinolin-4-(1H)-one (compound 13) showed reasonable cell-based antiviral activity (EC50 = 50 μM) with no considerable cytotoxicity (CC50 > 100 μM) in the cell viability assay, suggesting that it may be amenable to further development for identifying new anti-HIV-1 agents. Docking studies using the later crystallographic data available for PFV integrase corroborate favorable binding to the active site of HIV integrase, providing a basis for the design of more potent analogues.  相似文献

In recent decades, nanotechnology is growing rapidly owing to its widespread application in science and industry. The aim of the experiment was chemical characterization and evaluation of cytotoxicity, antioxidant, antibacterial, antifungal, and cutaneous wound healing activities of titanium nanoparticles using aqueous extract of Ziziphora clinopodioides Lam leaves (TiNPs@Ziziphora). These nanoparticles were characterized by fourier transformed infrared spectroscopy (FT‐IR), field emission scanning electron microscopy (FE‐SEM), energy dispersive X‐ray spectroscopy (EDS), and UV–visible spectroscopy. The synthesized TiNPs@Ziziphora had great cell viability dose‐dependently (Investigating the effect of the plant on human umbilical vein endothelial cells (HUVECs) cell line) and revealed this method was nontoxic. Then, 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) free radical scavenging test was done to assess the antioxidant properties, which indicated similar antioxidant potentials for TiNPs@Ziziphora and butylated hydroxytoluene. Agar diffusion tests were applied to determine the antibacterial and antifungal characteristics. Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC), and Minimum Fungicidal Concentration (MFC) were specified by macro‐broth dilution assay. The data were analyzed by SPSS 21 software (Duncan post‐hoc test). TiNPs@Ziziphora indicated higher antibacterial and antifungal effects than all standard antibiotics (p ≤ 0.01). Also, TiNPs@Ziziphora inhibited the growth of all bacteria at 2‐16 mg/ml concentrations and removed them at 2‐32 mg/ml concentrations (p ≤ 0.01). In case of antifungal properties of TiNPs@Ziziphora, they prevented the growth of all fungi at 2‐8 mg/ml concentrations and destroyed them at 2‐16 mg/ml concentrations (p ≤ 0.01). In vivo experiment, after creating the cutaneous wound, the rats were randomly divided into six groups: untreated control, treatment with Eucerin basal ointment, treatment with 3% tetracycline ointment, treatment with 0.2% TiO2 ointment, treatment with 0.2% Z. clinopodioides ointment, and treatment with 0.2% TiNPs@Ziziphora ointment. These groups were treated for 10 days. For histopathological and biochemical analysis of the healing trend, a 3 × 3 cm section was prepared from all dermal thicknesses at day 10. Use of TiNPs@Ziziphora ointment in the treatment groups substantially reduced (p ≤ 0.01) the wound area, total cells, neutrophil, and lymphocyte and remarkably raised (p ≤ 0.01) the wound contracture, hydroxyl proline, hexosamine, hexuronic acid, fibrocyte, and fibrocytes/fibroblast rate compared to other groups. In conclusion, the results revealed the useful non‐cytotoxic, antioxidant, antibacterial, antifungal, and cutaneous wound healing effects of TiNPs@Ziziphora.  相似文献

In this work, surfactant-coated Fe3O4@decanoic acid nanoparticles was synthesized as a viable nanosorbent for coextraction of drugs with different polarities (hydrophobic, hydrophilic). To reach desirable enrichment factors, efficient clean-up and low limits of detection (LODs), the method was combined with dispersive liquid–liquid microextraction (DLLME). The coupling of these extraction methods with GC-FID detection was applied to simultaneous extraction and quantification of venlafaxine (VLF) as a hydrophilic model drug and desipramine (DESI) and clomipramine (CLO) as hydrophobic model drugs in urine samples. The effect of sample pH, nanosorbent amount, sorption time, surfactant concentration, eluent type, eluent volume, salt content, elution time in magnetic solid phase extraction step and extraction solvent and its volume along with sample pH in DLLME step were optimized. Under the selected conditions, linearity was achieved within the range of 5–5000 µg L?1. The LOD values were obtained in the range of 1.5–3.0 µg L?1 for DESI, 1.2–2.5 µg L?1 for VLF and 2.0–4.0 µg L?1 for CLO, respectively. The percent of extraction recoveries and relative standard deviations (n?=?5) were in the range of 82.4–95.9 and 6.1 for DESI, 60.5–92.8 and 6.9 for VLF and 57.2–58.0 and 5.5 for CLO, respectively. Ultimately, the applicability of the new method was successfully confirmed by the extraction and quantification of DESI, VLF and CLO from human urine samples.  相似文献

3-Methyl-1-(4-sulfonic acid)butylimidazolium hydrogen sulfate [(CH2)4SO3HMIM][HSO4], a Brønsted acidic ionic liquid, has been used as an efficient, green, and reusable catalyst for the synthesis of 1,2,4,5-tetrasubstituted imidazoles using benzil, an aromatic aldehyde, and a primary amine in the presence of ammonium acetate under solvent-free conditions. The catalyst could be recycled and reused several times without noticeably decreasing the catalytic activity.  相似文献

The present study attempts to develop a detailed numerical approach and a simulation procedure to predict the motion of gas, ions and particles inside a simple parallel plate channel containing a single corona wire. A hybrid Finite Element (FEM)-Flux Corrected Transport (FCT)-Finite Volume (FVM) method is used: the FEM–FCT numerical algorithm is applied for modeling the steady-state corona discharge, while the turbulent gas flow and the particle motion under electrostatic forces are modeled using the commercial CFD code FLUENT. Calculations for the gas flow are carried out by solving the Reynolds-averaged Navier–Stokes equations and turbulence is modeled using the k? turbulence model. An additional source term is added to the gas flow equation to include the effect of the electric field, obtained by solving a coupled system of the electric field and charge transport equations using User-Defined Functions (UDFs). The particle phase is simulated based on the Lagrangian approach, where a large number of particles is traced with their motion affected by the gas flow and electrostatic forces using the Discrete Phase Model (DPM) in FLUENT. The developed model is useful to gain insight into the particle collection phenomena that take place inside an ESP.  相似文献

Biobutanol has attracted significant interest in recent decades and is seriously considered as a potential biofuel to partly replace gasoline. However, some production challenges must be addressed to make butanol economically viable such as the low product concentration and product toxicity inhibiting the microorganism. To alleviate these limitations, several in situ or ex situ separation techniques have been investigated in view of their integration to the biobutanol production process to enhance its economic viability. One of these techniques is adsorption which is one of the most energy-efficient techniques used for biobutanol separation. Considering the number of chemical species present in the ABE fermentation broth, it is essential to develop multicomponent adsorption isotherms for all components as a first step to design a high performance adsorption process. Few multicomponent isotherm models have been proposed such as multicomponent Langmuir and Freundlich. In this study, these two models as well as artificial neural networks were used to model the isotherms of each component in an ABE fermentation broth as a function of the equilibrium concentrations of all components for activated carbon F-400. Results showed that the multicomponent Langmuir model was not accurate due to the many simplifying assumptions. The multicomponent Freundlich and feedforward neural network (FFNN) isotherm models were able to predict the behavior of multicomponent systems very well. Indeed, the predictive model of the experimental data had a coefficient of determination (R2) of 0.97 and 0.99, for multicomponent Freundlich and FFNN isotherm models, respectively.  相似文献

A new ruthenium/dendrimer complex stabilized on the surface of silica-functionalized nano-magnetite was fabricated and well characterized. The nano-catalyst showed good activity in the synthesis of benzil derivatives via the oxidation of stilbenes with high turnover frequency (TOF) at room temperature. Efficient and convenient synthesis of β-acetamido ketones has been achieved by one-pot reaction of acetophenone, aryl aldehydes, acetyl chloride, and acetonitrile in the presence of 3-methyl-1-(4-sulfonylbutyl)imidazolium hydrogensulfate [HO3S(CH2)4MIM][HSO4], a Br?nsted-acidic ionic liquid, as a green and reusable catalyst in solvent-free media at room temperature. The catalyst could be recycled and reused without noticeable decrease in catalytic activity.  相似文献

A novel nitrite biosensor was constructed by simultaneous immobilization of hemoglobin (Hb) and a room temperature ionic liquid, octylpyridinium chloride ([OcPy][Cl]), on multi-walled carbon ionic liquid electrode (MWILE). The direct electron transfer of Hb showed a pair of redox peaks with a formal potential of ?0.187 V vs. Ag/AgCl in pH 5.0 acetate buffer solution. Nitrite (NO2 ?) catalysis on the modified electrode was investigated by cyclic voltammetry and amperometry. The biosensor exhibited a wide linear range for NO2 ? detection from 0.01 to 15 mM, with a detection limit (3σ) of 1.46 μM. MWILE provided an excellent matrix for protein immobilization and biosensor fabrication which could be used for the determination of NO2 ? with a low detection limit, fast response, long linearity, and excellent sensitivity.  相似文献
