Microfabrication processes, especially in silicon, are not compatible with biomolecules. Silicon and metal-based materials having crystalline structures are manipulated under harsh conditions with acids, bases, and organic solvents at high temperature. In comparison, organic biomolecules such as DNA and proteins have complex, three-dimensional structures and are sensitive to denaturation, oxidation, hydrolysis, and thermal destruction. Here, we report on the integration of DNA and the biotin-binding protein NeutrAvidin into microfabrication processes by using a novel approach based on a gold passivation mask. Our data show that this passivation method preserves approximately 84% of the biofunctionality of DNA and approximately 30% of that of NeutrAvidin under harsh process conditions. This novel technology enables the integration of DNA, proteins, and potentially other biological molecules into mass scalable microfabrication processes for biomedical devices, biochips, biosensors, and microelectromechanical systems with biomolecules (BioMEMS).  相似文献

In this contribution, it is demonstrated that inner magnetic field gradients can seriously affect the results of stimulated echo PFG NMR experiments on plant-based materials even if there is no notable content of paramagnetic substances. Such effects could be observed both in experiments on water in pharmaceutical grade cellulose powder materials and on eggplant fruit tissue. In both cases, it was observed that the effects of internal magnetic field gradients led to different relative values of the diffusion coefficient compared to values obtained with a gradient-compensating pulse sequence.  相似文献

We propose and analyze a wall model based on the turbulent boundary layer equations (TBLE) for implicit large-eddy simulation (LES) of high Reynolds number wall-bounded flows in conjunction with a conservative immersed-interface method for mapping complex boundaries onto Cartesian meshes. Both implicit subgrid-scale model and immersed-interface treatment of boundaries offer high computational efficiency for complex flow configurations. The wall model operates directly on the Cartesian computational mesh without the need for a dual boundary-conforming mesh. The combination of wall model and implicit LES is investigated in detail for turbulent channel flow at friction Reynolds numbers from Re τ  = 395 up to Re τ =100,000 on very coarse meshes. The TBLE wall model with implicit LES gives results of better quality than current explicit LES based on eddy viscosity subgrid-scale models with similar wall models. A straightforward formulation of the wall model performs well at moderately large Reynolds numbers. A logarithmic-layer mismatch, observed only at very large Reynolds numbers, is removed by introducing a new structure-based damping function. The performance of the overall approach is assessed for two generic configurations with flow separation: the backward-facing step at Re h = 5,000 and the periodic hill at Re H = 10,595 and Re H = 37,000 on very coarse meshes. The results confirm the observations made for the channel flow with respect to the good prediction quality and indicate that the combination of implicit LES, immersed-interface method, and TBLE-based wall modeling is a viable approach for simulating complex aerodynamic flows at high Reynolds numbers. They also reflect the limitations of TBLE-based wall models.  相似文献

Three different techniques (adiabatic passage Hartman-Hahn cross-polarization, optimal control designed pulses, and EXPORT) are compared for transferring (15)N magnetization to (13)C in solid-state NMR experiments under magic-angle-spinning conditions. We demonstrate that, in comparison to adiabatic passage Hartman-Hahn cross-polarization, optimal control transfer pulses achieve similar or better transfer efficiencies for uniformly-(13)C,(15)N labeled samples and are generally superior for samples with non-uniform labeling schemes (such as 1,3- and 2-(13)C glycerol labeling). In addition, the optimal control pulses typically use substantially lower average RF field strengths and are more robust with respect to experimental variation and RF inhomogeneity. Consequently, they are better suited for demanding samples.  相似文献

We present a 3D video-encoding technique called Holovideo that is capable of encoding high-resolution 3D videos into standard 2D videos, and then decoding the 2D videos back into 3D rapidly without significant loss of quality. Due to the nature of the algorithm, 2D video compression such as JPEG encoding with QuickTime Run Length Encoding (QTRLE) can be applied with little quality loss, resulting in an effective way to store 3D video at very small file sizes. We found that under a compression ratio of 134:1, Holovideo to OBJ file format, the 3D geometry quality drops at a negligible level. Several sets of 3D videos were captured using a structured light scanner, compressed using the Holovideo codec, and then uncompressed and displayed to demonstrate the effectiveness of the codec. With the use of OpenGL Shaders (GLSL), the 3D video codec can encode and decode in realtime. We demonstrated that for a video size of 512×512, the decoding speed is 28 frames per second (FPS) with a laptop computer using an embedded NVIDIA GeForce 9400 m graphics processing unit (GPU). Encoding can be done with this same setup at 18 FPS, making this technology suitable for applications such as interactive 3D video games and 3D video conferencing.  相似文献
