A microwave-based procedure for arsenic species extraction in alga samples (Sargassum fulvellum, Chlorella vulgaris, Hizikia fusiformis and Laminaria digitata) is described. Extraction time and temperature were tested in order to evaluate the extraction efficiency of the process. Arsenic compounds were extracted in 8 ml of deionised water at 90 degrees C for 5 min. The process was repeated three times. Soluble arsenic compounds extracted accounted for about 78-98% of total arsenic. The results were compared with those obtained in a previous work, where the extraction process was carried out by ultrasonic focussed probe for 30 s. Speciation studies were carried out by high performance liquid chromatography-hydride generation-inductively coupled plasma-atomic emission spectrometry (HPLC-HG-ICP-AES). The chromatographic method allowed us to separate As(III), As(V), monomethylarsonic acid and dimethylarsinic acid in less than 13 min. The chromatographic analysis of the samples allowed us to identify and quantify As(V) in Hizikia sample and Sargasso material, while the four arsenic species studied were found in Chlorella sample. In the case of Laminaria sample, none of these species was identified by HPLC-HG-ICP-AES. However, in the chromatographic analysis of this alga by HPLC-ICP-AES, an unknown arsenic species was detected.  相似文献

A procedure for arsenic species fractionation in alga samples (Sargassum fulvellum, Chlorella vulgaris, Hizikia fusiformis and Laminaria digitata) by extraction is described. Several parameters were tested in order to evaluate the extraction efficiency of the process: extraction medium, nature and concentration (tris(hydroxymethyl)aminomethane, phosphoric acid, deionised water and water/methanol mixtures), extraction time and physical treatment (magnetic stirring, ultrasonic bath and ultrasonic focussed probe). The extraction yield of arsenic under the different conditions was evaluated by determining the total arsenic content in the extracts by ICP-AES. Arsenic compounds were extracted in 5 mL of water by focussed sonication for 30 s and subsequent centrifugation at 14,000 × g for 10 min. The process was repeated three times. Extraction studies show that soluble arsenic compounds account for about 65% of total arsenic.

An ultrafiltration process was used as a clean-up method for chromatographic analysis, and also allowed us to determine the extracted arsenic fraction with a molecular weight lower than 10 kDa, which accounts for about 100% for all samples analysed.

Speciation studies were carried out by HPLC–ICP-AES. Arsenic species were separated on a Hamilton PRP-X100 column with 17 mM phosphate buffer at pH 5.5 and 1.0 mL min−1 flow rate. The chromatographic method allowed us to separate the species As(III), As(V), MMA and DMA in less than 13 min, with detection limits of about 20 ng of arsenic per species, for a sample injection volume of 100 μL. The chromatographic analysis allowed us to identify As(V) in Hizikia (46 ± 2 μg g−1), Sargassum (38 ± 2 μg g−1) and Chlorella (9 ± 1 μg g−1) samples. The species DMA was also found in Chlorella alga (13 ± 1 μg g−1). However, in Laminaria alga only an unknown arsenic species was detected, which eluted in the dead volume.  相似文献

Summary: In order to further evaluate the potential of FT-IR spectroscopic investigations on molecular processes during tensile testing experiments, the behavior of monolayer LLDPE films, made with ethylene-butene and ethylene-octene copolymers, was studied. Additionally, multilayer LLDPE films based on the same C4 and C8 copolymers were investigated. The stress-strain data obtained from the monolayer films indicate differences in the strain hardening region. It seems that the film samples PE 469-30-2 (C4-gas phase process) and PE 469-30-5 (C8 solution process) behave similarly whereas the strain hardening for the PE 469-30-3 (C4 solution process) requires lower stress values. The orientation function changes during the stretching of the films indicating that unfolding of the polymer chains occurs at lower strain for PE 469-30-5 (C8) than in the C4 materials. In the multilayer systems the Primplast 44 material (C8) shows a lower tendency for reorientation in the strain hardening region than the Coex 82 (C4) material. In this region of the stress-strain curve the lamellar structure is already transformed into the fibrillar arrangement. Regarding the orientation behavior of the material above 200% strain, a small increase in fb was observed, which led to a decrease of fc. In the octene product possibly the bulky side chains influence the unfolding significantly, producing a higher resistance to unfolding and alignment along the stretching direction. In part, this is potentially caused by the more perfect lamellae in the octene copolymer, which do not include the side chains, while the butene copolymer may have weaker lamellae because they contain a fraction of the side chains which create defects. Consequently, the octene copolymer requires higher stress values to be stretched and finally results in a lower stretchability of this material, as observed on an industrial scale during pallet wrapping tests. Based on the ratio of the structural absorbance parameters of the signals at 729 and 719 cm−1 changes in the crystallinity were studied. For the continuous stretching experiment, no monoclinic phase was detected even after Fourier self-deconvolution and peak fitting approaches. Literature data, however, describe that this crystalline transformation takes place as a result of mechanical deformation. Therefore, stepwise stretching experiments which allow an improvement of the spectral resolution to 1 cm−1 were carried out. In the deconvoluted spectra the monoclinic, orthorhombic and amorphous LLDPE modifications could be assigned. Ultimate stretchability and stretching force of the films, both monolayer and multilayer, was well correlated to the development of crystalline orientation in the films upon stretching. Other mechanical properties like Elmendorf tear and dart impact can also be better understood with these results.  相似文献

Often, deregulation of protein activity and turnover by tyrosine nitration drives cells toward pathogenesis. Hence, understanding how the nitration of a protein affects both its function and stability is of outstanding interest. Nowadays, most of the in vitro analyses of nitrated proteins rely on chemical treatment of native proteins with an excess of a chemical reagent. One such reagent, peroxynitrite, stands out for its biological relevance. However, given the excess of the nitrating reagent, the resulting in vitro modification could differ from the physiological nitration. Here, we determine unequivocally the configuration of distinct nitrated-tyrosine rings in single-tyrosine mutants of cytochrome?c. We aimed to confirm the nitration position by a non-destructive method. Thus, we have resorted to (1)H-(15)N heteronuclear single quantum coherence(HSQC) spectra to identify the (3)J(N?H) correlation between a (15)N-tagged nitro group and the adjacent aromatic proton. Once the chemical shift of this proton was determined, we compared the (1)H-(13)C HSQC spectra of untreated and nitrated samples. All tyrosines were nitrated at ε positions, in agreement to previous analysis by indirect techniques. Notably, the various nitrotyrosine residues show a different dynamic behaviour that is consistent with molecular dynamics computations.  相似文献

A chiral bisurea‐based superhydrogelator that is capable of forming supramolecular hydrogels at concentrations as low as 0.2 mM is reported. This soft material has been characterized by thermal studies, rheology, X‐ray diffraction analysis, transmission electron microscopy (TEM), and by various spectroscopic techniques (electronic and vibrational circular dichroism and by FTIR and Raman spectroscopy). The expression of chirality on the molecular and supramolecular levels has been studied and a clear amplification of its chirality into the achiral analogue has been observed. Furthermore, thermal analysis showed that the hydrogelation of compound 1 has a high response to temperature, which corresponds to an enthalpy‐driven self‐assembly process. These particular thermal characteristics make these materials easy to handle for soft‐application technologies.  相似文献

To quantify several acoustic features of the voice in patients withParkinson's disease (PD), 41 patients and 28 age and sex-matched controls were studied. PD severity was assessed with the Unified PD Rating Scale (UPDRS) and the Hoehn and Yahr staging. The Computerized Speech Lab 4300 program (Kay Elemetrics) was used. Two seconds of a sustained /a/ and a sentence were captured with a microphone and laryngograph equipment. Measures included fundamental frequency (FO), frequency perturbation (fitter), intensity perturbation (shimmer), and harmonic/noise ratio (H/N) of the vowel /a/, and frequency and intensity variability of a sentence, phonational range, dynamic range at the natural frequency, maximum phonational time and s/z ratio. All subjects underwent indirect laryngoscopy and/or laryngeal fibroscopy. When compared with controls, PD patients showed higher jitter, lower H/N ratio, lower frequency and intensity variability of the sentence, and lower phonational range and reported a higher frequency of the presence of low voice-intensity, monopitch, voice arrests, and struggle. These features seem to be unaffected by the duration and severity of the disease.  相似文献
