Background

Antisense oligonucleotide (AON)-mediated exon skipping is a powerful tool to manipulate gene expression. In the present study we investigated the potential of exon skipping by local injection in the central nucleus of the amygdala (CeA) of the mouse brain. As proof of principle we targeted the splicing of steroid receptor coactivator-1 (SRC-1), a protein involved in nuclear receptor function. This nuclear receptor coregulator exists in two splice variants (SRC-1a and SRC-1e) which display differential distribution and opposing activities in the brain, and whose mRNAs differ in a single SRC-1e specific exon.

Methods

For proof of principle of feasibility, we used immunofluorescent stainings to study uptake by different cell types, translocation to the nucleus and potential immunostimulatory effects at different time points after a local injection in the CeA of the mouse brain of a control AON targeting human dystrophin with no targets in the murine brain. To evaluate efficacy we designed an AON targeting the SRC-1e-specific exon and with qPCR analysis we measured the expression ratio of the two splice variants.

Results

We found that AONs were taken up by corticotropin releasing hormone expressing neurons and other cells in the CeA, and translocated into the cell nucleus. Immune responses after AON injection were comparable to those after sterile saline injection. A successful shift of the naturally occurring SRC-1a:SRC-1e expression ratio in favor of SRC-1a was observed, without changes in total SRC-1 expression.

Conclusions

We provide a proof of concept for local neuropharmacological use of exon skipping by manipulating the expression ratio of the two splice variants of SRC-1, which may be used to study nuclear receptor function in specific brain circuits. We established that exon skipping after local injection in the brain is a versatile and useful tool for the manipulation of splice variants for numerous genes that are relevant for brain function.  相似文献

Feedforward active control of the flexural waves in a single and L-shaped plate has been analytically and experimentally investigated. The plates are simply supported along two parallel edges, and free at the other two ends. Point forces were used to generate the primary and secondary plate excitations. The plate flexural displacement is described by a combination of a travelling wave solution and a modal expansion. The flexural wave coefficients were determined using the boundary conditions, continuity equations at the driving force locations, and continuity equations at the corner junction for the L-shaped plate. The control actuator and error sensor are optimally located in order to achieve the best control performance.  相似文献

Background  

Interruption of mature axons activates a cascade of events in neuronal cell bodies which leads to various outcomes from functional regeneration in the PNS to the failure of any significant regeneration in the CNS. One factor which seems to play an important role in the molecular programs after axotomy is the stearoyl Coenzyme A-desaturase-1 (SCD-1). This enzyme is needed for the conversion of stearate into oleate. Beside its role in membrane synthesis, oleate could act as a neurotrophic factor, involved in signal transduction pathways via activation of protein kinases C.  相似文献

Low-frequency modulation of distortion product otoacoustic emissions (DPOAEs) was measured from the human ears. In the frequency domain, increasing the bias tone level resulted in a suppression of the cubic difference tone (CDT) and an increase in the magnitudes of the modulation sidebands. Higher-frequency bias tones were more efficient in producing the suppression and modulation. Quasi-static modulation patterns were derived from measuring the CDT amplitude at the peaks and troughs of bias tones with various amplitudes. The asymmetric bell-shaped pattern resembled the absolute value of the third derivative of a nonlinear cochlear transducer function. Temporal modulation patterns were obtained from inverse FFT of the spectral contents around the DPOAE. The period modulation pattern, averaged over multiple bias tone cycles, showed two CDT peaks each correlated with the zero-crossings of the bias tone. The typical period modulation pattern varied and the two CDT peaks emerged with the reduction in bias tone level. The present study replicated the previous experimental results in gerbils. This noninvasive technique is capable of revealing the static position and dynamic motion of the cochlear partition. Moreover, the results of the present study suggest that this technique could potentially be applied in the differential diagnosis of cochlear pathologies.  相似文献

A time‐resolved intensified charge coupled device‐based Raman microspectrometer system dedicated to the study of solid samples is described, offering good optical, temporal and spatial resolution. The advantages of this approach are demonstrated on Al2O3:Cr3+, obtaining for the first time the temporal evolution of the excited state transition Ē → 2Ā. Background  

Methamphetamine (MA) use is a worldwide problem. Abusers can have cognitive deficits, monoamine reductions, and altered magnetic resonance spectroscopy findings. Animal models have been used to investigate some of these effects, however many of these experiments have not examined the impact of MA on the stress response. For example, numerous studies have demonstrated (+)-MA-induced neurotoxicity and monoamine reductions, however the effects of MA on other markers that may play a role in neurotoxicity or cell energetics such as glucose, corticosterone, and/or creatine have received less attention. In this experiment, the effects of a neurotoxic regimen of (+)-MA (4 doses at 2 h intervals) on brain monoamines, neostriatal GFAP, plasma corticosterone, creatinine, and glucose, and brain and muscle creatine were evaluated 1, 7, 24, and 72 h after the first dose. In order to compare MA's effects with stress, animals were subjected to a forced swim test in a temporal pattern similar to MA administration [i.e., (30 min/session) 4 times at 2 h intervals].  相似文献

Low band gap D‐A conjugated PNs consisting of 2‐ethylhexyl cyclopentadithiophene co‐polymerized with 2,1,3‐benzothiadiazole (for nano‐PCPDTBT) or 2,1,3‐benzoselenadiazole (for nano‐PCPDTBSe) have been developed. The PNs are stable in aqueous media and showed no significant toxicity up to 1 mg · mL?1. Upon exposure to 808 nm light, the PNs generated temperatures above 50 °C. Photothermal ablation studies of the PNs with RKO and HCT116 colorectal cancer cells were performed. At concentrations above 100 µg · mL?1 for nano‐PCPDTBSe, cell viability was less than 20%, while at concentrations above 62 µg · mL?1 for nano‐PCPDTBT, cell viability was less than 10%. The results of this work demonstrate that low band gap D‐A conjugated polymers 1) can be formed into nanoparticles that are stable in aqueous media; 2) are non‐toxic until stimulated by IR light and 3) have a high photothermal efficiency.

DNA glycosylase enzymes recognize and remove structurally distinct modified forms of DNA bases, thereby repairing genomic DNA from chemically induced damage or erasing epigenetic marks. However, these enzymes are often promiscuous, and advanced tools are needed to evaluate and engineer their substrate specificity. Thus, in the present study, we developed a new strategy to rapidly profile the substrate specificity of 8-oxoguanine glycosylases, which cleave biologically relevant oxidized forms of guanine. We monitored the enzymatic excision of fluorophore-labeled oligonucleotides containing synthetic modifications 8-oxoG and FapyG, or G. Using this molecular beacon approach, we identified several hOGG1 mutants with higher specificity for FapyG than 8-oxoG. This approach and the newly synthesized probes will be useful for the characterization of glycosylase substrate specificity and damage excision mechanisms, as well as for evaluating engineered enzymes with altered reactivities.

First-principles calculations are presented for the layered perovskite Ca3Mn2O7. The results reveal a rich set of coupled structural, magnetic, and polar domains in which oxygen octahedron rotations induce ferroelectricity, magnetoelectricity, and weak ferromagnetism. The key point is that the rotation distortion is a combination of two nonpolar modes with different symmetries. We use the term "hybrid" improper ferroelectricity to describe this phenomenon and discuss how control over magnetism is achieved through these functional antiferrodistortive octahedron rotations.  相似文献
