An on-line HPLC screening method for detection of inhibitors of human cytochrome P450 1A2 in extracts was developed. HPLC separation of extracts is connected to a continuous methoxyresorufin-O-demethylation (MROD) assay in which recombinant human P450 1A2 converts methoxyresorufin to its fluorescent metabolite resorufin. The system was tested with three P450 1A2 inhibitors, for which minimum detectable amounts (MDA) ranging from 0.7 to 9.5 ng were obtained. Analysis of a kava kava and a basil extract showed that the on-line system is applicable to complex mixtures, since in both extracts, peaks with P450 1A2 inhibiting activity were observed. 相似文献

In this article we analyze in detail the electronic properties of the D(3h)-symmetric tris(ethylene) complexes of nickel, palladium, and platinum ([M(C(2)H(4))(3)] M=Ni, Pd, Pt). In the case of [Pd(C(2)H(4))(3)] the analysis is based on new experimental IR and Raman spectra for the matrix-isolated molecules and in all cases on the results of quantum-chemical (DFT) calculations. The experimental spectra collected for [Pd(C(2)H(4))(3)] provide evidence for several previously unobserved vibrational modes, including the in-phase and out-of-phase nu(C-C) and delta(CH(2)) modes, and the in-phase nu(M-C) mode. Special consideration is given to possible inter-ligand interactions. The interaction force constant f(CC,CC) between two C(2)H(4) ligands can be directly estimated from the spectra, and its very small value (0.002 N m(-1)) indicates the absence of any significant inter-ligand interaction. An analysis of the topology of the theoretical electron density distribution, rho(r), and the corresponding Laplacian, nabla(2)rho(r), for [Pd(C(2)H(4))(3)] and its lighter and heavier homologues [Ni(C(2)H(4))(3)] and [Pt(C(2)H(4))(3)], respectively, is in full agreement with the conclusions drawn from the experimental results. The combined experimental and quantum-chemical results provide detailed insights in the electronic properties of these prototypical ethylene complexes. 相似文献

The Indiana Science Initiative (ISI) is a systemic effort to reform K–8 science education. The program provides teachers with professional development, reform‐oriented science modules, and materials support. To examine the impact of the initiative's professional development, a participant observation study was conducted in the program's pilot year. Five teachers in grades 3–6 were observed and interviewed as they implemented the ISI‐provided modules. Analysis of the observation data revealed that the teachers incorporated each of the features of inquiry science instruction. However, they did not consistently teach in a way that was aligned with the intent of the ISI. Examination of interview data provided insight into influences on teachers' use of inquiry with the ISI‐provided modules. These data revealed that teachers were aware of the intent of the ISI and attempted to align their instruction. However, teachers were influenced by their perceptions of students' behavior and abilities as well as timing and the appropriate level of teacher control needed to facilitate science instruction. The research suggests that professional development activities should prepare teachers to help learners evaluate explanations against alternatives, connect explanations to scientific knowledge, and provide strategies to address teachers' perceptions of students, timing, and teacher control. 相似文献

This paper investigates the uncertainty of a hyper-elastic model by random material parameters as stochastic variables. For its stochastic discretization a polynomial chaos (PC) is used to expand the coefficients into deterministic and stochastic parts. Then, from experimental data in combination with artificial data for elastomers the distribution of the force-displacement curves are known. In the numerical example the PC-based stochastic and the deterministic parameter identification are used for generation of the distribution of Ogden's material parameters. This article deals with stochastic maintenance models that include a repair facility and three types of ‘unreliable’ machines: the main facility of working and reserve machines, and an auxiliary facility of ‘super‐reserve’ machines. Working machines breakdown exponentially and are immediately replaced by available reserve machines. Defective machines line up for repair and refurbished machines are returned to the main facility. This system falls into the category of closed queues with hot standbys and has more options than the basic model. If the main facility is restored to its original quantity and the repair facility leaves on routine maintenance, all reserve machines are temporarily blocked and renewals come from the super‐reserve group until the latter is exhausted. Explicit formulas obtained demonstrate a relatively effortless use of functionals of the main stochastic characteristics, and optimization of their objective function. Magnetic forces are now being utilised in an amazing variety of microfluidic applications. Magnetohydrodynamic flow has been applied to the pumping of fluids through microchannels. Magnetic materials such as ferrofluids or magnetically doped PDMS have been used as valves. Magnetic microparticles have been employed for mixing of fluid streams. Magnetic particles have also been used as solid supports for bioreactions in microchannels. Trapping and transport of single cells are being investigated and recently, advances have been made towards the detection of magnetic material on-chip. The photoinduced interconversion between cinnamido‐substituted cyclodextrins constitutes a gating switch through which the substituent moves to open or block access to the cyclodextrin cavity. Most unusually for a cyclodextrin‐based device, the operation of this gate is solvent‐independent and unaffected by potentially competitive guests. It occurs in MeOH and DMSO, as well as in water. This contrasts with other cyclodextrin inclusion phenomena that are usually driven by hydrophobic effects and limited to aqueous media. 相似文献
