An enhanced hemoglobin–membrane association has been previously documented in sickle cell anemia. However, it is not known
how this interaction is modified during the hemoglobin S polymerization process. In this work, we use a model of reconstituted
erythrocytes from ghost membranes whose cytoskeleton proteins had been previously labeled with the 4-maleimido Tempo spin
label, and that were subsequently resealed with hemoglobin S or A solutions. Using electron paramagnetic resonance spectroscopy,
we studied the time dependence of the spectral W/S parameter, indicative of the conformational state of cytoskeleton proteins
(mainly spectrin) under spontaneous deoxygenation, with the aim of detecting the eventual effects due to hemoglobin S polymerization.
The differences observed in the temporal behavior of W/S in erythrocytes reconstituted with both hemoglobins were considered
as experimental evidence of an increment in hemoglobin S–membrane interaction as a result of the polymerization process of

Liquid Phase Photo-Deposition (LPPD) technique has been used to obtain both colloidal particles and thin films of metallic
and chloride nickel from solutions of only precursor Ni(acac)2 (acac=2,4-pentandionato). Metallic nickel was obtained from ethanol solutions by direct nickel(II) photoreduction at 254 nm
and by acetone sensitised reaction at 300 nm. In this latter process the rate was higher than in the first one. NiCl2 was formed from CCl4 solution by a solvent-initiated reaction. TEM analysis, performed on colloidal particles of nickel, showed that their dimensions
are in the range 2–4 nm. The films did not present carbon contamination and were characterized by AFM, XPS and GIXRD. Metallic
films consisted of particles of 20–40 nm that are the result of the aggregation of smaller crystallites (4–5 nm). Larger agglomerations

Monodisperse magnetizable colloidal silica particles in a stable dispersion have been functionalized with a homogeneous catalyst: a PCP–pincer Pd-complex. In a proof-of-principle experiment we demonstrate the catalytic activity of the colloids in a C–C bond formation reaction. Advantages of the magnetic silica carriers are the large surface-to-volume ratio and the easy recovery by magnetic separation. After magnetic separation, the catalyst-loaded particles are readily redispersed for further use. 相似文献

Patients diagnosed with nodules (NO = 40) in a large university hospital clinic and an age-stratum matched nondiseased group (ND = 200) described adverse outcomes of vocal impairment on work and work-related communications. NOs were significantly more likely than NDs to report symptoms of hoarseness (73% vs. 26%), high-note difficulty (70% vs. 20%), difficulty speaking with a lower voice (53% vs. 13%) and a tired voice (50% vs. 10%), and their greatest source of physical discomfort was associated with scratchiness (61% vs. 3%). The average number of symptoms was four in NOs and less than one in NDs. Nodule patients were most concerned about the effects their voice problem would have on their future career (78% vs. 24%) and 49% of NOS reported their voice problem had an adverse work effect in the past compared with 4% of NDs. Having a voice condition limited current job performance in 39% of the NO group but only in 2% of the ND group. The results suggest that a diagnosis of nodules plays a major role in disrupting careers and work activities and that available educational programs and additional research are needed for improving their functional ability and preventing adverse outcomes in the lives of individuals with voice disorders. 相似文献

In this paper, we have analyzed the signal processing methods both in digital and optical domain to enhance the transmission performance of downstream signalling in long reach passive optical networks (LR-PONs). The impact of non-linear (NL) equalization through signal processing, i.e. Volterra Equalization (VE), Digital Backpropagation (BP) and Optical Phase Conjugation with Non-linearity Module (OPC-NM) is investigated, in 10 Gbit/s (XG) DP-QPSK long-reach wavelength division multiplexed (WDM) PONs without midspan repeaters over 120 km standard single mode fibre (SMF) link for down-stream signals. Due to the compensation of optical Kerr effects, the sensitivity penalty is reduced to 2 dB by BP algorithm, 1.5 dB by VE algorithm and 2.69 dB by OPC-NM. Moreover, with the implementation of NL equalization technique we are able to get the transmission distance of 126.6 km SMF for the 1:1024 split-ratio at 5 GHz channel spacing in the non-linear region. Furthermore, the concept of super passive optical network (S-PON) is also evaluated, which involves a repeater stage consisting of optical amplifiers, to study the feasibility for receiver side signal processing and simplification. 相似文献

The larvicidal action of the entomopathogen Bacillus sphaericus towards Culex quinquefasciatus is due to the binary (Bin) toxin present in crystals, which are produced during bacterial sporulation. The Bin toxin needs to recognize and bind specifically to a single class of receptors, named Cqm1, which are 60-kDa α-glucosidases attached to the apical membrane of midgut cells by a glycosylphosphatidylinositol anchor. C. quinquefasciatus resistance to B. sphaericus has been often associated with the absence of the α-glucosidase Cqm1 in larvae midgut microvilli. In this work, we aimed to investigate, at the ultrastructural level, the midgut cells from C. quinquefasciatus larvae whose resistance relies on the lack of the Cqm1 receptor. The morphological analysis showed that midgut columnar cells from the resistant larvae are characterized by a pronounced production of lipid inclusions, throughout the 4th instar. At the end of this stage, resistant larvae had an increased size and number of these inclusions in the midgut cells, while only a small number were observed in the cells from susceptible larvae. The morphological differences in the midgut cells of resistant larvae found in this work suggested that the lack of the Cqm1 receptor, which also has a physiological role as being an α-glucosidase, can be related to changes in the cell metabolism. The ultrastructural effects of Bin toxin on midgut epithelial cells from susceptible and resistant larvae were also investigated. The cytopathological alterations observed in susceptible larvae treated with a lethal concentration of toxin included breakdown of the endoplasmic reticulum, mitochondrial swelling, microvillar disruption and vacuolization. Some effects were observed in cells from resistant larvae, although those alterations did not lead to larval death, indicating that the receptor Cqm1 is essential to mediate the larvicidal action of the toxin.This is the first ultrastructural study to show differences in the cell morphology of resistant larvae and further investigation is needed to understand the impact of the lack of expression of midgut enzymes on the physiology of resistant insects. 相似文献

A series of metallocenes, namely, [Cp2ZrCl2], [(MeCp)2ZrCl2], [(nBuCp)2ZrCl2], [(iBuCp)2ZrCl2], [(tBuCp)2ZrCl2], [Et(Ind)2ZrCl2], [Et(IndH4)2ZrCl2], and [MeSi2(Ind)2ZrCl2)] were analyzed by extended x‐ray absorption fine structure (EXAFS) and x‐ray photoelectron spectroscopy (XPS). Complementary techniques, UV–vis spectroscopy and cyclic and differential pulse voltammetry, were employed to characterize the organometallic complexes. The catalysts were evaluated in ethylene polymerization, having methylaluminoxane (MAO) as the cocatalyst, and the resulting polymers were characterized by gel permeation chromatography. The structural and electronic effects caused by the coordination sphere around the metal center and their effects on the catalytic activity and polymer characteristics are discussed. Three cis-dioxovanadium(V) complexes with similar N-salicylidenehydrazide ligands modeling hydrogen bonding interactions of vanadate relevant for vanadium haloperoxidases are studied by (51)V solid-state NMR spectroscopy. Their parameters describing the quadrupolar and chemical shift anisotropy interactions (quadrupolar coupling constant C(Q), asymmetry of the quadrupolar tensor eta(Q), isotropic chemical shift delta(iso), chemical shift anisotropy delta(sigma), asymmetry of the chemical shift tensor eta(sigma) and the Euler angles alpha, beta and gamma) are determined both experimentally and theoretically using DFT methods. A comparative study of different methods to determine the NMR parameters by numerical simulation of the spectra is presented. Detailed theoretical investigations on the DFT level using various basis sets and structural models show that by useful choice of the methodology, the calculated parameters agree to the experimental ones in a very good manner. 相似文献

Neodymium based fluorescence presents several advantages in comparison to conventional rare earth or enzyme-substrate based fluorescence emitting sources (e.g.Tb, HRP) . Based on this fact we have herein explored a Nd-based fluoroimmunoassay. We efficiently detected the presence of an oxidized low-density lipoprotein (oxLDL) in human plasma a well-known marker for cardiovascular diseases, which causes around 30% of deaths worldwide. Conventional fluoroimmunoassay uses time-resolved luminescence techniques, with detection in the visible range, to eliminate the fluorescence background from the biological specimens. By using an immunoassay based on functionalized Y2O3:Nd3+ nanoparticles, where the excitation and emission processes in the Nd3+ ion occur in the near-infrared (NIR) region, we have succeeded in eliminating the interferences from the biological fluorescence background, avoiding the use of time-resolved techniques. This yields higher emission intensity from the Nd3+-nanolabels and efficient detection of anti-oxidized low-density lipoproteins (anti-oxLDL) by Y2O3:Nd3+-antibody-antigen conjugation, leading to a novel biolabeling method. 相似文献
