Experiments with human hepatoma PLC/PRF/5 cells and human embryo skin fibroblasts involving the use of three different tests (colony formation, Trypan blue exclusion, labeled thymidine incorporation) have demonstrated a significantly higher photosensitizing activity of chlorin e6 conjugates with internalizable ligands as compared to that of chlorin e6 itself. Receptor-mediated internalization of chlorin e6 conjugates ensures a greater photosensitization of cells than binding of those conjugates to cell surface receptors. The suitability of such conjugates that permit the delivery of a photosensitizer to sensitive intracellular targets is discussed. 相似文献

Novel N-bonded imidato complexes of general formula [Pd(N-N)(C6F5)(imidate)](imidate = maleimidate, succinimidate or phthalimidate; N-N = 2,2'-bipyridine (bipy), 4,4'-dimethyl-2,2'-bipyridine (Me2bipy) or N,N,N',N'-tetramethylethylenediamine (tmeda)), [NBu4][Pd(C6F5)(H2O)(succinimidate)2] and [NBu4][Pd(C6F5)(L)(succinimidate)2](L = PPh3 or t-BuNC) have been synthesised. These complexes are air-, light- and moisture-stable. The crystal structures of [Pd(tmeda)(C6F5)(maleimidate)].H2O.0.5CHCl3, [NBu4][Pd(C6F5)(H2O)(succinimidate)2].H2O and [NBu4][Pd(C6F5)(t-BuNC)(succinimidate)2].2H2O have been determined by X-ray diffraction. Many of these new complexes are shown to be active phosphine-free palladium catalysts/precatalysts for the Suzuki cross-coupling reactions of aryl bromides and aryl chlorides with phenylboronic acid. 相似文献

Ceramides are important intracellular second messengers that play a role in the regulation of cell growth, differentiation and programmed cell death. Qualitative and quantitative analysis of these second messengers requires sensitive and specific analytical methods to detect endogenous levels of individual ceramide species and to differentiate between them. Nine synthetic ceramides were separated by liquid chromatography coupled to tandem mass spectrometry on a C18 bonded silica column. The lipids were eluted in gradient elution mode using a mixture of water, acetonitrile and 2-propanol as mobile phase. They were detected by reaction monitoring performed on positive ion electrospray generated ions. Collision-induced fragmentations conducted on ceramides produced a well characteristic product ion at m/z 264, making multiple reaction monitoring (MRM) well suited for various ceramides quantitative measurements. After optimization of the extraction step, the proposed methodology was able to identify and quantify different ceramide species issued from human cancer cells. The method could be validated for C16, C18 and C20 ceramides, quantified at the nanogram level. The validation exhibits good results with respect to linearity, accuracy and precision. 相似文献

Anhydrous disodium phosphonate, Na2HPO3, was prepared by dehydration of its pentahydrate. The crystal structure of Na2HPO3 was solved from high resolution X‐ray powder diffraction data (P21/n; Z = 4; a = 9.6987(1), b = 6.9795(1), c = 5.0561(1) Å, β = 92.37(1)°; V = 341.97(1) Å3). The crystal structure consists of two types of sodium‐oxygen polyhedra, which are connected via common edges and vertices forming layers perpendicular to [100]. These Na(1)‐ and Na(2)‐layers are interlinked via common edges, forming in a 3D‐framework. In a cellular environment, the presence of macromolecular cosolutes and membrane interfaces can influence the folding-unfolding behavior of proteins. Here we report on the pressure stability of alpha-chymotrypsin in the ternary system bis(2-ethylhexyl)sodium sulfosuccinate-octane-water using FTIR spectroscopy. The ternary system forms anionic reverse micelles which mimic cellular conditions. We find that inclusion of a single protein molecule in a reverse micelle does not alter its conformation. When pressurized in bulk water, alpha-chymotrypsin unfolds at 750 MPa into a partially unfolded structure. In contrast, in the ternary system, the same pressure increase induces a random coil-like unfolded state, which collapses into an amorphous aggregate during the decompression phase. It is suggested that the unfolding pathway is different in a cell-mimicking environment due to the combined effect of multiple factors, including confinement. A phase transition of the reverse micellar to the lamellar phase is thought to be essential to provide the conditions required for unfolding and aggregation, though the unfolding is not a direct result of the phase transition. Our observations therefore suggest that membranes may cause the formation of alternative conformations that are more susceptible to aggregation. 相似文献

Cu (II) complexes with the sterically hindered diphenol derivatives 3,5-di(tert-butyl)-1,2-benzenediol (I), 4,6-di(tert-butyl)-1,2,3-benzenetriol (II) and the sulfur-containing 4,6-di(tert-butyl)-3-(2-hydroxyethylsulfanyl)-1,2-benzenediol (III) and 2-[4,6-di(tert-butyl)-2,3-dihydroxyphenylsulfanyl]acetic acid (IV) have been synthesized and characterized by elemental analysis, TG/DTA, FT-IR, ESR, XPS, XPD and conductivity measurements.
Compounds I–III can coordinate in their singly deprotonated forms and act as bidentate ligands. These compounds yield Cu (II) complexes of the stoichiometry Cu(L)2, which have square planar geometry (g| > g⊥ > ge). Unlike them, compound IV behaves as a terdentate ligand, and its complex Cu(LIV)2 has distorted octahedral geometry. We have constructed a very large virtual diversity space containing more than 1013 chemical compounds. The diversity space is built from about 400 combinatorial libraries, which have been expanded by choosing sizeable collections of suitable R-groups that can be attached to each link point of their scaffolds. These R-group collections have been created by selecting reagents that have drug-like properties from catalogs of available chemicals. As members of known combinatorial libraries, the compounds in the diversity space are in general synthetically accessible and useful as potential drug leads. Hence, the diversity space can be used as a vast source of compounds by a de novo drug design program. For example, we have used such a program to generate inhibitors of HIV integrase enzyme that exhibited activity in the micromolar range. 相似文献

Abstract. Fluorescence quantum yield and lifetime measurements of the tyrosine residues in ribonuclease-A (RNase) were used to study the conformational changes involved in the denaturation of the enzyme. Measurements were done on RNase and on selectively acetylated RNase in the native, the partly denatured (reductive cleavage of S-S bridges or treatment with 8 M urea) and in the fully denatured state. The data were interpreted to mean that the opening of the S-S bridges causes large parts of the enzyme chain to unfold while leaving a hydrophobic region; including one of the tyrosine residues, intact. The biological activity of RNase is destroyed by this unfolding. Urea apparently does penetrate the protein coil but does not greatly affect the RNase structure since some of its biological activity is still retained. The opening of the S-S bridges in the presence of urea destroys the native conformation (and biological activity) completely leaving the protein in the form of an uncoiled polypeptide chain. It is suggested which parts of the protein structure might be affected by partial denaturation. 相似文献

We report the synthesis and characterization of covalent dyads and multiads of electron acceptors (A) and donors (D), with the purpose of exploiting their nanophase separation behavior toward (a) two-dimensional (2D) surface patterning with well-defined integrated arrays of dissimilar molecular electronic features and (b) bulk self-assembly to noncovalent columnar versions of the so-called "double cable" systems, the likes of which could eventually provide side-by-side percolation pathways for electrons and holes in solar cells. Soluble, alkylated hexa-peri-hexabenzocoronenes (HBCs) bearing tethered anthraquinones (AQs) are shown by scanning tunneling microscopy (STM) to self-assemble at the solution-graphite interface into either defect-rich polycrystalline monolayers or extended 2D crystalline domains, depending on the number of tethered AQs. In the bulk, the thermal stability of the room-temperature HBC columnar phase is increased, which is attributed to the desired nanotriphase separation of HBC columns, insulating alkyl sheaths, and AQ units. Homeotropic alignment (columns normal to surfaces), predicted to be ideal for potential exploitation of such "double cables" in photovoltaic devices, is demonstrated. 相似文献

Corn stover is currently being evaluated as a feedstock for ethanol production. The corn stover suspensions fed to reactors
typically range between 10 and 40% solids. To simulate and design bioreactors for processing highly loaded corn stover suspensions,
the rheologic properties of the suspension must be measured. In systems with suspended solids, rheologic measurements are
difficult to perform owing to settling in the measurement devices. In this study, viscosities of corn stover suspensions were
measured using a helical ribbon impeller viscometer. A calibration procedure is required for the impeller method in order
to obtain the shear rate constant, k, which is dependent on the geometry of the measurement system. The corn stover suspensions are described using a power law
