Alzheimer’s disease (AD) is an age-related neurodegenerative disorder characterized by cognitive deficits, which are accompanied by memory loss and cognitive disruption. Rhodiola sachalinensis (RSE) is a medicinal plant that has been used in northeastern Asia for various pharmacological activities. We attempted to carry out the bioconversion of RSE (Bio-RSE) using the mycelium of Bovista plumbe to obtain tyrosol-enriched Bio-RSE. The objective of this study was to investigate the effects of Bio-RSE on the activation of the cholinergic system and the inhibition of oxidative stress in mice with scopolamine (Sco)-induced memory impairment. Sco (1 mg/kg body weight, i.p.) impaired the mice’s performance on the Y-maze test, passive avoidance test, and water maze test. However, the number of abnormal behaviors was reduced in the groups supplemented with Bio-RSE. Bio-RSE treatment improved working memory and avoidance times against electronic shock, increased step-through latency, and reduced the time to reach the escape zone in the water maze test. Bio-RSE dramatically improved the cholinergic system by decreasing acetylcholinesterase activity and regulated oxidative stress by increasing antioxidant enzymes (superoxide dismutase (SOD) and catalase (CAT)). The reduction in nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) signaling in the brain tissue due to scopolamine was restored by the administration of Bio-RSE. Bio-RSE also significantly decreased amyloid-beta 1–42 (Aβ1–42) and amyloid precursor protein (APP) expression. Moreover, the increased malondialdehyde (MDA) level and low total antioxidant capacity in Sco-treated mouse brains were reversed by Bio-RSE, and an increase in Nrf2 and HO-1 was also observed. In conclusion, Bio-RSE protected against Sco-induced cognitive impairment by activating Nrf2/HO-1 signaling and may be developed as a potential beneficial material for AD.  相似文献

Human embryonic stem cell-derived cardiomyocytes (hESC-CMs) have been reported to exhibit immature embryonic or fetal cardiomyocyte-like phenotypes. To enhance the maturation of hESC-CMs, we identified a natural steroidal alkaloid, tomatidine, as a new substance that stimulates the maturation of hESC-CMs. Treatment of human embryonic stem cells with tomatidine during cardiomyocyte differentiation stimulated the expression of several cardiomyocyte-specific markers and increased the density of T-tubules. Furthermore, tomatidine treatment augmented the number and size of mitochondria and enhanced the formation of mitochondrial lamellar cristae. Tomatidine treatment stimulated mitochondrial functions, including mitochondrial membrane potential, oxidative phosphorylation, and ATP production, in hESC-CMs. Tomatidine-treated hESC-CMs were more sensitive to doxorubicin-induced cardiotoxicity than the control cells. In conclusion, the present study suggests that tomatidine promotes the differentiation of stem cells to adult cardiomyocytes by accelerating mitochondrial biogenesis and maturation and that tomatidine-treated mature hESC-CMs can be used for cardiotoxicity screening and cardiac disease modeling.Subject terms: Heart failure, Embryonic stem cells, Stem-cell differentiation  相似文献

The measurement of radial directional natural frequency and damping ratio in a vehicle tire has been studied. Natural frequencies and damping ratios in the radial direction of various tires, from passenger car tires to truck bus tires, are reported. The radial direction modal parameters of tires subjected to different levels of inflation pressure, have been determined by using a frequency response function method. To obtain the theoretical natural frequency and mode shape, the plane vibration of a tire has been modeled as though it were that of a circular beam. By using the Tielking method that is based on Hamilton’s principle, theoretical results have been determined by considering the rotational velocity, tangential and radial stiffness, radial directional velocity and tension force which is due to tire inflation pressure. The results show that experimental conditions can be considered as the parameters that shift the natural frequency and damping ratio.  相似文献

Abstract

Three new homologous series of mesogens containing three rings in the main structure and a lateral aromatic branch have been synthesized. When the lateral aromatic branch is attached to the middle ring, monotropic phases are observed, and there is a strong tendency to form the smectic C phase. When the lateral aromatic branch is attached to an outer ring, the compounds possess purely enantiotropic nematic phases. The terminal alkoxy chain in the outer ring which has a lateral aromatic substituent can be replaced by a rigid group without disturbing the large nematic range. Selective introduction of a particular substitutent may be useful for the construction of devices possessing desirable characteristics such as non-linear optical properties.  相似文献

Two new thermally crosslinkable hole‐transporting polymers, X‐PTPA and X‐PCz, were synthesized via Yamamoto coupling reactions. The number‐averaged molecular weights (Mn) of X‐PTPA and X‐PCz were found to be 45,000 and 48,000, respectively, and therewith, polydispersity indices were of 1.8 and 1.7, respectively. Thermally crosslinked X‐PTPA and X‐PCz exhibit excellent solvent resistance and stable optoelectronic properties. The UV–visible maximum absorption peaks of X‐PTPA and X‐PCz in the thin film state are at 389 and 322 nm, respectively. The HOMO levels of X‐PTPA and X‐PCz are estimated to be ?5.27 and ?5.39 eV, respectively. Multilayered devices (ITO/crosslinked X‐PTPA or X‐PCz/SY‐PPV/LiF/Al) were fabricated with SY (SuperYellow) as the emitting layer. The maximum efficiency of the multilayered device with a crosslinked X‐PTPA layer is approximately three times higher than that of the device without a crosslinked X‐PTPA layer and much higher than that of the crosslinked X‐PCz device. This result can be explained by the observations that crosslinked X‐PTPA produces increased electron accumulation within the emitter, SY, and also efficient exciton formation due to improved charge balance. © 2013 Wiley Periodicals, Inc. In this paper, tridiagonal Toeplitz matrix (type I, type II) with opposite-bordered rows are introduced. Main attention is paid to calculate the determinants, the inverses and the eigenpairs of these matrices. Specifically, the determinants of an $n\times n$ tridiagonal Toeplitz matrix with opposite-bordered rows can be explicitly expressed by using the $(n-1)$th Fibonacci number, the inversion of the tridiagonal Toeplitz matrix with opposite-bordered rows can also be explicitly expressed by using the Fibonacci numbers and unknown entries from the new matrix. Besides, we give the expression of eigenvalues and eigenvectors of the tridiagonal Toeplitz matrix with opposite-bordered rows. In addition, some algorithms are presented based on these theoretical results. Numerical results show that the new algorithms have much better computing efficiency than some existing algorithms studied recently.  相似文献

The neutron irradiated reactor pressure vessel (RPV) steels at various dose of 0–1018 n/cm2 have been studied with Mössbauer, x-ray diffraction, and VSM. The Mössbauer data shows that the value of magnetic hyperfine field of Fe atom that exist at martensite is 330 kOe at site 1 and 305 kOe at site 2. At room temperature, the total absorption area of Mössbauer spectra with respect to irradiation of neutron is constant for the dose of 0–1016 n/cm2, while over the dose of 0–1017 n/cm2 the absorption area decreases rapidly. But the doublet area for the dose of 0–1016 n/cm2 is constant, while over the dose of 1017 n/cm2 it increases with increasing the fluence level of neutron. The coercivity and remanence of the neutron irradiated samples do not change significantly. But the maximum induction decreases by 5% at 1018 n/cm2, compared with that of the as-received sample.  相似文献

A liquid chromatographic chiral stationary phase based on (+)‐(18‐crown‐6)‐2,3,11,12‐tetracarboxylic acid was applied to the resolution of 15 analytes, including racemic rasagiline, a chiral drug for the treatment of Parkinson's disease, and its analogues. The composition of mobile phase was optimized to be ethanol/acetonitrile/acetic acid/triethylamine (80:20:0.2:0.3, v/v/v/v) by evaluating the chromatographic results for the resolution of five selected analytes under various mobile phase conditions. Under the optimized mobile phase conditions, racemic rasagiline was resolved quite well with a separation factor of 1.48 and resolution of 2.71 and its 14 analogues were also resolved reasonably well with separation factors of 1.06–1.54 and resolutions of 0.54–2.11. Among 15 analytes, racemic rasagiline was resolved best except for just one analyte. The analyte structure–enantioselectivity relationship indicated that racemic rasagiline has the most appropriate structural characteristics for resolution on the chiral stationary phase.  相似文献
