ABSTRACT: A rapid and reproducible stability indicating TLC method was developed for the determination of prednisolone acetate and chloramphenicol in presence of their degraded products. Uniform degradation conditions were maintained by refluxing sixteen reaction mixtures for two hours at 80°C using parallel synthesizer including acidic, alkaline and neutral hydrolysis, oxidation and wet heating degradation. Oxidation at room temperature, photochemical and dry heating degradation studies were also carried out. Separation was done on TLC glass plates, pre-coated with silica gel 60F-254 using chloroform: methanol (14:1 v/v). Spots at Rf 0.21 ± 0.02 and Rf 0.41 ± 0.03 were recognized as chloramphenicol and prednisolone acetate, respectively. Quantitative analysis was done through densitometric measurements at multiwavelength (243 nm, λmax of prednisolone acetate and 278 nm, λmax of chloramphenicol), simultaneously. The developed method was optimized and validated as per ICH guidelines. Method was found linear over the concentration range of 200-6000 ng/spot with the correlation coefficient (r2 ± S.D.) of 0.9976 ± 3.5 and 0.9920 ± 2.5 for prednisolone acetate and chloramphenicol, respectively. The developed TLC method can be applied for routine analysis of prednisolone acetate and chloramphenicol in presence of their degraded products in their individual and combined pharmaceutical formulations. 相似文献

(S)-5-Fluoro-2-(2,2,6,6-tetramethylpiperidin-1-yloxymethyl)-1-tosylindoline, a 2-methyleneoxy-substituted chiral indoline, was synthesized on multigram scale using an efficient copper-catalyzed enantioselective intramolecular alkene aminooxygenation. The synthesis is accomplished in four steps and the indoline is obtained in 89% ee (>98% after one recrystallization). The preparation of the (1-naphthyl)(trifluoromethyl) O-carboxy-anhydride 1 and its use as a chiral derivatizing agent with several alpha-chiral primary amines are reported. The very large Delta delta(RS) values observed in (1)H NMR have been correlated with a marked preference of the corresponding alpha-hydroxy-amides for the eclipsed conformation. In this work, new ways of plasticizing polylactide (PLA) with low molecular poly(ethylene glycol) (PEG) were developed to improve the ductility of PLA while maintaining the plasticizer content at maximum 20 wt.% PLA. To this end, a reactive blending of anhydride-grafted PLA (MAG-PLA) copolymer with PEG, with chains terminated with hydroxyl groups, was performed. During the melt-processing, a fraction of PEG was grafted into the anhydride-functionalized PLA chains. The role of the grafted fraction was to improve the compatibility between PLA and PEG. Reactive extrusion and melt-blending of neat and modified PLA with PEG did not induce any dramatic drop of PLA molecular weight. The in situ reactive grafting of PEG into the modified PLA in PLA/PEG blends showed a clear effect on the thermal properties of PLA. It was demonstrated by DSC that the mobility gained by PLA chains in the plasticized blends yielded crystallization. The grafting of a fraction of PEG into PLA did not affect this process. However, DSC results obtained after the second heating showed an interesting effect on the Tg when 20 wt.% PEG were melt blended with neat PLA or 10 wt.% MAG-PLA. In the latter case, the Tg displayed by the reactive blend was shifted to even lower temperatures at around 14 °C, while the Tg of neat PLA and PLA blended with 20 wt.% PEG was around 60 and 23 °C, respectively. Regarding viscoelastic and viscoplastic properties, the presence of MAG-PLA does not significantly influence the behavior of plasticized PLA. Indeed, with or without MAG-PLA, elastic modulus and yield stress decrease, while ultimate strain increases with the addition of PEG into PLA. 相似文献

We have compared TMAH thermochemolysis with the classical method using acid hydrolysis for carbohydrates analysis in a peat core. Even if TMAH thermochemolysis does not analyse hemicellulosic carbohydrates and discriminate each individual carbohydrate sensu stricto, it allows the analysis of a cellulose pool hidden to acid hydrolysis and the specific analysis of free and terminal carbohydrates. Simple direct comparisons of thermochemolysis data with data generated from acid hydrolysis cannot be done because of the different mechanisms involved in each process. TMAH thermochemolysis must be viewed and used as a pertinent and complementary method for the analysis of carbohydrates protected and trapped by the organic matter in complex environmental systems. 相似文献

The copper(II) complexes of formula [Cu2(2,5-dpp)(H2O)4(CF3SO3)4] · 2H2O (1) and [Cu2(2,5-dpp)(H2O)2(tcnoet)4]n (2) [2,5-dpp = 2,5-bis(2-pyridyl)pyrazine and tcnoet− = 1,1,3,3-tetracyano-2-ethoxypropenide anion] have been prepared and their structures determined by X-ray crystallographic methods. Compound 1 is a dinuclear complex where the 2,5-dpp molecule acts as a bis-bidentate bridge between the two copper centers, the electroneutrality being achieved by four terminally bound triflate anions. Each copper(II) ion presents an elongated octahedral CuN2O4 environment with two nitrogen atoms from 2,5-dpp and two water molecules in the basal plane and two triflate-oxygen atoms in the axial positions. Compound 2 is a zigzag chain of copper(II) ions with regular alternating 2,5-dpp and double tcnoet groups as bridges. Each copper(II) ion exhibits an elongated octahedral CuN5O surrounding with four nitrogen atoms, two from 2,5-dpp, one from a terminally bound tcnoet and the other from a bridging tcnoet occupying the equatorial positions and a water oxygen and a nitrogen from a monodentate tcnoet in the axial sites. The values of the copper–copper separation across 2,5-dpp are 6.763(1) (1) and 6.754(1) Å (2) whereas that through the double tcnoet bridge is 9.559(1) Å (2). The investigation of the magnetic properties of 1 and 2 in the temperature range 1.9–295 K reveal a Curie law behaviour for 1 and a very weak ferromagnetic interaction for 2. The poor ability of the 2,5-dpp ligand to mediate magnetic interactions between the copper(II) ions in the 2,5-dpp-bridged copper(II) complexes contrast with the somewhat better ability of the pyrazine ring in related pyrazine-bridged copper(II) complexes. 相似文献
