A new NiII complex of 5,6-dihydro-2H-pyran-3-aldehyde thiosemicarbazone (HDPTSC) has been synthesized and characterized by microanalyses, magnetic susceptibility, molar conductance measurements and by spectral methods (i.r., u.v.–vis., 1H-n.m.r.). The structure of [Ni(DPTSC)2] · B DMF has been solved using X-ray diffraction and found to be highly symmetrical with a trans -arrangement of the two bidentate ligands. The thiosemicarbazone coordinates as an anionic ligand via the thiosemicarbazone moietys azomethine nitrogen and thiolate sulfur [on loss of the N2 hydrogen]. The electrochemical behavior of the ligand and its NiII complex, determined by cyclic voltammetry, shows that the redox process of the ligand was highly irreversible, whereas the redox process of the NiII complex was observed as a one electron transfer process in a quasi-reversible and diffusion-controlled reaction.  相似文献

The oxidation-reduction reaction between U(VI) and Ti(III) in HCl solution was studied spectrophotometrically. The reaction is second-order at all concentrations of reactants, HCl, ferrous chloride and mannitol used in this work. In 5M HCl the rate constantk increases with increasing Ti(III) concentration, whereas it decreases with increasing U(VI) concentration, with increasing HCl concentration from 1.00M to 7.17M and increases thereafter from 7.17M to 11.79M. The addition of mannitol causes a consistent decrease in the rate of reaction, whereas ferrous chloride has no effect. The activation energy for this oxidation-reduction reaction was 47.90±0.11 kJ·mol–1. The values of H , G and S were 45.40±0.11 kJ·mol–1, 72.50±0.17 kJ·mol–1 and –91.10±0.22J·k–1·mol–1, respectively. The mode of reaction is discussed in the light of kinetic results.  相似文献

A general methodology has been established for rapid generation and screening of combinatorial glycopeptide library and subsequent mass spectrometric sequencing to identify the mimetics of Galalpha(1,3)Gal epitopes. Using this approach, several active glycopeptide sequences were recognized and found to inhibit the binding of human natural anti-Gal antibodies with comparable IC(50)s to synthetic Galalpha(1,3)Gal oligosaccharides. The most active glycopeptides detected from the library included Gal-Tyr-Trp-Arg-Tyr, Gal-Thr-Trp-Arg-Tyr, and Gal-Arg-Trp-Arg-Tyr. These glycopeptides showed higher affinities to anti-Gal antibodies than known Galalpha(1,3)Gal peptide mimetics, such as DAHWESWL and SSLRGF. Our results suggest that, by combining a peptide sequence (the "functional" mimic part) with a terminal alpha-linked galactose moiety (the "structural" mimic part), the resulting glycopeptide could be a very good Galalpha(1,3)Gal mimetic. Analysis of these active glycopeptides provided first-hand information regarding the binding site of anti-Gal antibodies to facilitate the structurally based design of more potent and stable inhibitors.  相似文献

Phytochemical investigations on the non-alkaloidal extracts of Mitragyna stipulosa bark has led to the isolation of a series of triterpenoids mainly consisting of quinovic acid ([structure: see text]) and its glycoside derivatives [structure: see text] and [structure: see text]. The other constituents isolated include alpha-amyrin, 3beta-acetyl ursolic acid and a mixture of oleanolic and ursolic acid and beta-sitosterol glucopyranoside. Their structures were identified by spectral and chemical studies and compounds [structure: see text] and [structure: see text] were, respectively, identified as quinovic acid 3-O-[beta-D-glucopyranoside] (quinovin glycoside C) and quinovic acid 3-O-[beta-D-quinovopyranoside]-27-O-[beta-D-glucopyranosyl] ester. Summary This paper deals with systematic distribution studies of metal ions on stannic arsenate in butanol-HNO3 systems. Kd values of metal ions have been determined by the batch process in solvent systems containing varying proportions of butanol and 50% HNO3. A simple sensitive and economical method for the determination of labetalol hydrochloride has been proposed, based on the reaction of labetalol with sodium nitroprusside and hydroxylamine hydrochloride in sodium dihydrogen phosphate-sodium hydroxide buffer solution of pH 12. The green-blue color produced due to the formation of a nitroso derivative has been measured at 695 nm. The Beer's range was obeyed in the concentration range of 2-51 μg·mL^-1 with molar absorptivity of 0.48 × 10^4 L·mol^-1·cm^-1. Rigorous statistical analyses were performed for the validation of the method. A detailed investigation of the selectivity of the method has been done to find it to be highly selective for the determination of labetalol hydrochloride in the presence of its acidic degradation product and common excipients of formulations. The proposed method was successfully applied to the determination of labetalol hydrochloride in the laboratory prepared dosage forms. Comparison of the means of the proposed procedure with a reference method using point as well as interval hypotheses showed no statistically significant difference. The developed method was extended to investigate its applicability to biological samples.  相似文献

Dysphania ambrosioides (L.) Mosyakin and Clemants is an annual or ephemeral perennial herb used traditionally in the Mediterranean region in folk medicine to treat various illnesses, including those related to the digestive system. This study aims to assess the antispasmodic, myorelaxant, and antioxidant effects of D. ambrosioides flower hydroethanolic extract and its chloroform and ethyl acetate fractions in a comparative study to evaluate the result of the extraction type on the potential activity of the extract. Both rat and rabbit jejunum were used to evaluate the antispasmodic and myorelaxant effect, while the antioxidant effect was evaluated using DPPH, a ferric reducing power assay, and a beta-carotene bleaching test. LC/MS-MS analysis was carried out to reveal the composition of the different types of extract. Following the results, the hydroethanolic extract showed a significant myorelaxant effect (IC50 = 0.39 ± 0.01 mg/mL). Moreover, it was shown that the hydroethanolic extract demonstrated the best antispasmodic activity (IC50 = 0.51 ± 0.05 mg/mL), followed by the ethyl acetate (IC50 = 4.05 ± 0.32 mg/mL) and chloroform (IC50 = 4.34 ± 0.45 mg/mL) fractions. The antioxidant tests showed that the hydroethanolic extract demonstrated high antioxidant activity, followed by the ethyl acetate and chloroform fractions. The LC/MS-MS analysis indicates that the plant extract was rich in flavonoids, to which the extract activity has been attributed. This study supports the traditional use of this plant to treat digestive problems, especially those with spasms.  相似文献

Nigella species are widely used to cure various ailments. Their health benefits, particularly from the seed oils, could be attributed to the presence of a variety of bioactive components. Roasting is a critical process that has historically been used to facilitate oil extraction and enhance flavor; it may also alter the chemical composition and biological properties of the Nigella seed. The aim of this study was to investigate the effect of the roasting process on the composition of the bioactive components and the biological activities of Nigella arvensis and Nigella sativa seed extracts. Our preliminary study showed that seeds roasted at 50 °C exhibited potent antimicrobial activities; therefore, this temperature was selected for roasting Nigella seeds. For extraction, raw and roasted seed samples were macerated in methanol. The antimicrobial activities against Streptococcus agalactiae, Streptococcus epidermidis, Streptococcus pyogenes, Candida albicans, Escherichia coli, Enterobacter aerogenes, Klebsiella pneumoniae, and Klebsiella oxytoca were determined by measuring the diameter of the zone of inhibition. The cell viability of extracts was tested in a colon carcinoma cell line, HCT-116, by using a microculture tetrazolium technique (MTT) assay. Amino acids were extracted and quantified using an automatic amino acid analyzer. Then, gas chromatography–mass spectrometry (GC–MS) analysis was performed to identify the chemical constituents and fatty acids. As a result, the extracts of raw and roasted seeds in both Nigella species showed strong inhibition against Klebsiella oxytoca, and the raw seed extract of N. arvensis demonstrated moderate inhibition against S. pyogenes. The findings of the MTT assay indicated that all the extracts significantly decreased cancer cell viability. Moreover, N. sativa species possessed higher contents of the measured amino acids, except tyrosine, cystine, and methionine. The GC–MS analysis of extracts showed the presence of 22 and 13 compounds in raw and roasted N. arvensis, respectively, and 9 and 11 compounds in raw and roasted N. sativa, respectively. However, heat treatment decreased the detectable components to 13 compounds in roasted N. arvensis and increased them in roasted N. sativa. These findings indicate that N. arvensis and N. sativa could be potential sources of anticancer and antimicrobials, where the bioactive compounds play a pivotal role as functional components.  相似文献

Therapeutic effects of anticancer medicines can be improved by targeting the specific receptors on cancer cells. Folate receptor (FR) targeting with antibody (Ab) is an effective tool to deliver anticancer drugs to the cancer cell. In this research project, a novel formulation of targeting drug delivery was designed, and its anticancer effects were analyzed. Folic acid-conjugated magnetic nanoparticles (MNPs) were used for the purification of folate receptors through a novel magnetic affinity purification method. Antibodies against the folate receptors and methotrexate (MTX) were developed and characterized with enzyme-linked immunosorbent assay and Western blot. Targeting nanomedicines (MNP-MTX-FR Ab) were synthesized by engineering the MNP with methotrexate and anti-folate receptor antibody (anti-FR Ab). The cytotoxicity of nanomedicines on HeLa cells was analyzed by calculating the % age cell viability. A fluorescent study was performed with HeLa cells and tumor tissue sections to analyze the binding efficacy and intracellular tracking of synthesized nanomedicines. MNP-MTX-FR Ab demonstrated good cytotoxicity along all the nanocomposites, which confirms that the antibody-coated medicine possesses the potential affinity to destroy cancer cells in the targeted drug delivery process. Immunohistochemical approaches and fluorescent study further confirmed their uptake by FRs on the tumor cells’ surface in antibody-mediated endocytosis. The current approach is a useful addition to targeted drug delivery for better management of cancer therapy along with immunotherapy in the future.  相似文献

The effect of the progressive increase in temperature (off-line pyrolysis in an open-medium system) on a monomaceralic coal (low rank and pure vitrinite) has been studied on the basis of changes in its chemical and physical structure. The properties of the solid residues and liquid effluents obtained were studied using a wide range of petrographic and geochemical techniques. Results support the validity of this pyrolysis method for following the physico–chemical processes that occur in vitrinite of bituminous coal rank. Of special interest is the information concerning the control of the structure on the vitrinite reflectance parameter in the bituminization stage. The evolution of properties and changes occurring at this stage show that the increase in vitrinite reflectance is strongly related and depends on changes in the chemical structure of this maceral during heating rather than on changes in elemental composition. However, any relationship between reflectance and chemical properties needs to be established empirically. The mechanism during heating consists of a complex mixture of depolymerization and condensation reactions with the progressive homogenisation of the botanical structure of the vitrinite. Moreover, pyrolysis conditions lead to a rapid and massive expulsion of the thermally generated products, causing a significant decrease in the H/C atomic ratio and higher thermostability of the solid residues. The enhancement of cross-linking with the formation of C–O–C bridges, which have high activation energy also contributes to an increase in thermal stability. Results obtained from the production and expulsion of oil suggest that this method is suitable for providing an estimation of the amount of hydrocarbons that might be produced and migrate from coals of similar rank. This is of special interest for coals with significant petroligenous potential or for coals that are gas-prone, depending on their composition and rank.  相似文献
